Alim, Zuhal; Kilinç, Namik; Şengül, Bülent; Beydemir, Şükrü
Aldose reductase (AR) inhibitors have vital importance in the treatment and prevention of diabetic complications. In this study, rat kidney AR was purified 19.34-fold with a yield of 3.49% and a specific activity of 0.88 U/mg using DE-52 Cellulose anion exchange chromatography, gel filtration chromatography and 2′5′ ADP Sepharose-4B affinity chromatography, respectively. After purification, the in vitro inhibition effects of some phenolic acids (tannic acid, chlorogenic acid, sinapic acid, protocatechuic acid, 4-hydroxybenzoic acid, p-coumaric acid, ferulic acid, vanillic acid, syringic acid, α-resorcylic acid, 3-hydroxybenzoic acid and gallic acid) were investigated on purified enzyme. We determined IC50, Ki values and inhibition types of these phenolic acids. As a result, tannic and chlorogenic acid had a strong inhibition effect. On the other hand, gallic acid had a weak inhibition effect. In this study, all phenolic acids except for chlorogenic acid and p-coumaric acid showed non-competitive inhibition effects on rat kidney AR.

Ürkek, Bayram; Şengül, Mustafa; Erkaya, Tuba; Aksakal, Vecihi
The aim of this study was to investigate the effects of production systems and milk collection periods on the somatic cell count (SCC), some microbiological properties, total aerobic mesophilic bacteria (TAMB), coliform, Staphylococcus aureus (S. aureus), yeast and mould) and antibiotic residue of milk; in Turkey. Milk samples were collected from 9 conventional farms and 9 organic farms during one year time, at six different months (December 2013 to October 2014), and all farms were selected from the same geographical locations. All organically managed farms had organic production certificates given by the Republic of Turkey Ministry of Food, Agriculture and Livestock. The count of TAMB, coliform, and coagulase positive S. aureus were affected by production systems at the level of p<0.01; yeast and mold, and somatic cell count (SCC) were affected at the level of p<0.05. But, differences according to months were statistically significant only on TAMB (p<0.01) and coliform (p<0.05) counts. The general means of TAMB, coliform and yeast and mould counts of the organic milk (OM) were significantly lower (p<0.05), while the general means of SCC and coagulase positive S. aureus count of the OM was significantly higher (p<0.05) compared to conventional milk (CM). Antibiotic residue was determined in one of the CM sample and in two of the OM samples. Our study is the first research that compared conventional and organic milk in Turkey. This study indicated that the microbiological quality of OM was the higher in terms of TAMB, coliform and yeast and mould, whereas was the lower in relation to SCC and coagulase positive S. aureus counts. But, the quality of both milk types should be improved.

Background: Both insulin-like growth factor-binding protein 7 (IGFBP7) and telomere length (TL) are associated with proliferation and senescence of human breast cancer. This study assessed the clinical significance of both TL and IGFBP7 methylation status in breast cancer tissues compared with adjacent normal tissues. We also investigated whether IGFBP7 methylation status could be affecting TL. Methods: Telomere length was measured by quantitative PCR to compare tumors with their adjacent normal tissues. The IGFBP7 promoter methylation status was evaluated by methylation-specific PCR and its expression levels were determined by western blotting. Results: Telomeres were shorter in tumor tissues compared to controls (P<.0001). The mean TL was higher in breast cancer with invasive ductal carcinoma (IDC; n=72; P=.014) compared with other histological type (n=29), and TL in IDC with HER2 negative (n=53; P=.017) was higher than TL in IDC with HER2 positive (n=19). However, telomeres were shortened in advanced stages and growing tumors. IGFBP7 methylation was observed in 90% of tumor tissues and 59% of controls (P=.0002). Its frequency was significantly higher in IDC compared with invasive mixed carcinoma (IMC; P=.002) and it was not correlated either with protein expression or the other clinicopathological parameters. Conclusion: These results suggest that IGFBP7 promoter methylation and shorter TL in tumor compared with adjacent tissues may be predictive biomarkers for breast cancer. Telomere maintenance may be indicative of IDC and IDC with HER2 (−) of breast cancer. Further studies with larger number of cases are necessary to verify this association.

İspirli, Hümeyra; Demirbaş, Fatmanur; Dertli, Enes
Enterococcus species was isolated from Turkish white cheese and genotypic characterization of these isolates revealed the presence of 12 distinct strains belonging to 5 different species: Enterococcus faecium, E. faecalis, E. durans, E. gallinarum and E. italicus. All strains showed good survival ability under bile salt conditions. The antibiotic resistance of these strains was low but two strains showed high levels of resistance including resistance to vancomycin. PCR detection of virulence determinant in these isolates revealed that cheese isolate enterococci contained virulence genes common in gut and food originated enterococci and importantly two strains harboured cylB gene related to cytolysin metabolism. No complete hemolytic activity was observed for Enterococcus strains but partial hemolytic activity was observed 8 out of 12 strains. Additionally, all strains showed important levels of antimicrobial activity against food-borne pathogens and PCR screening of genes encoding enterocin A and B indicated the presence of enterocin B gene in all tested strains. The antimicrobial activities of the tested strains were lost after proteolytic enzyme treatments but no alteration was observed at different pHs and after heat treatments. In summary, this study reflected characteristics of Enterococcus strains presented in Turkish white cheese in terms of functional and safety perspectives.

The aim of this research was to investigate the effect of cornelian cherry (Cornus mas L.) paste (CP) on the quality properties of ice cream. CP was added to an ice cream mix at four concentrations (0, 5, 10 and 15%, w/w) for ice cream production. The increment of CP level caused the increased of vitamin C content, a values and overrun values, whereas it decreased the viscosity of samples compared to Control ice cream sample. The results indicated that lyophilised water extracts of CP (LWECP) contain remarkable phenolic compounds. The findings showed that there is a positive correlation between the total phenolics and flavonoid contents in LWECP and ice cream samples antioxidant activity. CP has shown to be an effective source of natural antioxidants. CP may be used as a source of natural colour and flavour agent in ice cream manufacture. CP enhanced vitamin C amounts of ice cream, and improve sensory properties. In the literature surveys of Science Direct, Web of Science, Google Scholar and Scopus databases an ice cream study using cornelian cherry fruit was not found.

A rose hip is the fruit of a rose plant and mostly belongs to Rosa canina and Rosa dumalis. These species are genuine found as wild in natural conditions, while in some countries are cultivated for their hips that are picked and used in a wide variety of preparations. Because they contain a variety of antioxidants, carotenoids, flavonoids, polyphenols, leucoanthocyanins and catechins, rose hips are considered to be a good cancer preventative. The Rosa genus has been the focus of many recent studies due to its potential benefic effects on treatment and prevention of several diseases. However, there are few in vitro studies concerning its composition and antioxidant capacity. The aim of the present study was to determine bioactive content, including ascorbic acid, total phenolics, total flavonoids, total carotenoids and antioxidant activity of Rosa dumalis genotypes growing wild in Erzurum province located eastern Anatolia. The results revealed that fruits of Rosa dumalis different genotypes were rich rich in terms of vitamin C, which ranged from 402 to 511 mg/100 g fresh weight base. The total phenolic content varied from 297 to 403 mg/100 g fresh weight. The genotype 'E-09' had the highest total flavonoids content (229 mg quercetin equivalent/100 g fresh weight) and the lowest value was attributed to genotype 'E-04' with 143 mg/ quercetin equivalent/100 g fresh weight. The antioxidant activity of the genotypes was between 12.9-28.6 μg Trolox/ml samples. The results revealed that there was enough diversity among Rosa dumalis genotypes for bioactive content and promising genotypes, with high bioactive content, were determined, which can become study material for future breeding activities.

Coating surfaces of fruit with electrospun zein mats with functionalized antimicrobial properties can be a novel strategy to prevent fungal colonization on fruit surfaces. In this study, we tested curcumin-loaded electrospun zein nanofibers (CLZN) in terms of limitation of postharvest decay on CLZN–coated apples infected with Botrytis cinerea and Penicillium expansum. Mixtures of zein and curcumin (the curcumin amounts of 2.5 and 5 wt% based on the weight of zein powder) were electrospun to yield cylindrical and ultrafine (< 350 nm in diameter) polymeric nanofibers. In addition, molecular, thermal, zeta potential and morphological properties of the CLZN as well as their encapsulation efficiency and releasing kinetics were determined, revealing that the developed zein-based scaffolds showed high encapsulation efficiency, molecular interactions with curcumin within nanofibers, alterations in physical states of these components, smooth beadless structure and good thermal (an endothermic peak at 152 °C) and dispersion stability (− 24 mV of ζ potential) properties. In vitro antifungal activity tests conducted at 27 °C for six days showed that CLZN were effective against growth of the tested fungal pathogens, exhibiting almost 40–50% inhibition of mycelial growth of the fungal pathogens; but the antifungal effect against P. expansum was but two-fold higher than that against B. cinerea. In vivo tests conducted at 23 °C with 75% humidity for six days confirmed in vitro test results in terms of both visual inspections on uncoated and coated apples, revealing almost 50% reduction in lesion diameter measured on coated apples infected with Penicillium expansum. Our results suggest that CLZN mats open up new opportunities for a novel application of edible and biodegradable antifungal coating material with the ability to hinder fungal proliferation on coated apples during storage period. Industrial relevance We coated the surfaces of fruits with electrospun mats with functionalized antimicrobial properties to prevent fungal colonization on fruit surface. The coating of apples with curcumin-loaded zein nanoparticles (CLZNs) limited the postharvest decay caused by the fungal pathogens, Penicillium expansum and Botrytis cinerea. This study showed that by encapsulation of curcumin into zein-based nanofibers considerably increased the antifungal effectiveness of curcumin. Our results highlighted the potential use of the CLZN as an effective fungicidal coating material against P. expansum and B. cinerea and suggested that CLZNs can be promising tools to compete with synthetic fungicide counterparts of curcumin. The results of this study should be of great importance to industrial applications in terms of development of natural, but effective preservatives as alternative to synthetic ones.

Kaya, Elif Duygu; Söyüt, Hakan; Beydemir, Şükrü
It is known that heavy metals have toxic effects on fish. Insufficient measures are a serious problem in our country and around the world. This problem can threaten human health in areas where it is common for people to obtain nutrition from local bodies of water. In this study, the toxicological impacts of some heavy metals were investigated on carbonic anhydrase activity in gilthead gills. Carbonic anhydrase (CA) was purified from gilthead sea bream (Sparus aurata) gills with a specific activity of 2872.92EUmg-1 and a yield of 32.84% using affinity chromatography. The overall purification was approximately ~84-fold. SDS-polyacrylamide gel electrophoresis showed a single band, and the MW was approximately 30.5kDa (Soyut et al., 2008, 2012; Soyut and Beydemir, 2008, 2012; Kaya et al., 2013). The kinetic and characteristic properties of CA such as the optimum pH, stable pH, optimum temperature, activation energy (Ea), activation enthalpy (δH), Q10, Km and Vmax were determined. Cadmium (Cd2+), copper (Cu2+), nickel (Ni2+) and silver (Ag+) inhibited CA activity in in vitro conditions. Ki values were calculated for these metals. Ki values were 31.20mM for cadmium (Cd2+), 161.96mM for copper (Cu2+), 10.79mM for nickel (Ni2+) and 0.0082mM for silver (Ag+) based on Lineweaver-Burk plots. Except for cadmium, heavy metals had the same inhibition mechanism. Cadmium was competitive, and the others were noncompetitive.
