Effect of probiotics supplementation on adipokine profile (Visfatin, adiponectin and chemerin), intestinal (citrulline) and thyroid function in japanese quail subjected to heat stress; [Einfluss einer probiotischen futterergänzung auf das adipokin-profil (Visfatin, adiponectin und chemerin), und die darm- (citrullin) und schilddrüsenfunktion japanischer wachteln unter hitzestress]

    Bayraktar, B.; Tekce, E.

In this study, we aimed to determine the effect of probiotic supplementation of Lactobacillus reuteri E81 (LRE; 200, 400, 600 mg/kg) to the diets of Japanese quail (Coturnix coturnix japonica) reared under heat stress (HS) on serum adipokine (visfatin, adiponectin and chemerin), intestinal (citrulline) and thyroid hormone levels. On day 7 of the trial, the animals were assigned to 8 groups [Control temperature (CT): control (C, without LRE supplementation), LRE-200, LRE-400 and LRE-600 and HS: control (HSC), SLRE-200, SLRE-400 and SLRE-600], each composed of 56 animals of equal body weight. Each group was divided into 4 subgroups withg 14 animals per compartment. During the experiments, the temperature applied to the CT-groups C, LRE-200, LRE-400 and LRE-600 was 25°C and to the HS-groups HSC, SLRE-200, SLRE-400 and SLRE-600 was 37°C. At the end of the study (days 21 and 42), a dose-dependent decrease in citrulline, chemerin, adiponectin, and thyroid hormones and a dose-dependent increase in the visfatin hormone level (p < 0.01) due to LRE supplementation were observed in the HS and CT groups compared to its control. In conclusion, the present study showed that dietary supplementation with 600 mg/kg of LRE affected serum chemerin, adiponectin, T3, T4, TSH and visfatin levels in Japanese quail raised under heat stress (p < 0.001).

Effects of an Essential Oil Mixture Added to Drinking Water for Temperature-Stressed Broilers: Performance, Meat Quality, and Thiobarbituric Acid-Reactive Substances

    Tekce, E.; Çınar, K.; Bayraktar, B.; Takma, Ç.; Gül, M.

This study investigated the effects of an essential oil mixture (EOM; Eucalyptus globulus labill, Tymus vulgaris, Cymbopogon nardus, and Syzgium aromaticum) added to drinking water on temperature-stressed broilers. The performance parameters (body weight, average daily weight, feed intake, and feed conversion ratio), meat quality, and thiobarbituric acid-reactive substances (TBARS) were evaluated. In a 42-d study, 400 Ross-308 male chickens (1-d-old) were randomly assigned to 8 different groups (n = 50), each containing 4 subgroups (n = 8) (22°C Control (C), C + 250 mL/1,000 L, C + 500 mL/1,000 L, C + 750 mL/1,000 L), 36°C (stress control (SC), SC + 250 mL/1,000 L, SC + 500 mL/1,000 L, SC + 750 mL/1,000 L). Adding 750 mL/1,000 L at 22°C and 250 mL/1,000 L at 36°C was more beneficial to the fattening performance parameters than those in the control group. EOM reduced liver weight but increased abdominal fat in the SEOM-250 groups but did not affect other organ weights. EOM had no effect on the TBARS or the b* color parameter while it augmented the a* and L* coordinates of meat color.

Microbiological, physicochemical, textural characteristics and oxidative stability of butter produced from high-pressure homogenisation treated cream at different pressures

    Sert, Durmuş; Mercan, Emin

Microbiological, physicochemical, and textural characteristics, and oxidative stability was investigated of butter produced from raw cream treated with high-pressure homogenisation (HPH) at 0 (control), 10, 20, 30, 50, or 70 MPa. HPH treatment of cream increased lightness and decreased yellowness of butter. Firmness of all butter samples was between 97 (control) and 380 N (70 MPa). HPH decreased the spreadability of butter dependent on increasing pressure. A monomodal fat particle size distribution was determined in all butter samples; D[3.2] ranged from 25.25 to 125.5 μm and HPH decreased average particle size compared with the control. The induction time of butter ranged from 27.40 (control) to 39.61 h (70 MPa). Above 50 MPa, Salmonella, and yeast and moulds were completely inactivated whilst coliforms and staphylococci were not detected in butter from HPH treated cream at 70 MPa, indicating that HPH can improve the microbial quality of butter produced from raw cream.

    Tekce, Emre; Bayraktar, B.; Aksakal, V.; Dertli, E.; Kamiloğlu, A.; Çinar, K.; Takma, Ç.; Kaya, H.; Gül, M.

This study investigated the effects of the addition of Lactobacillus reuteri E81 (LRE) into rations of chukar partridges (Alectoris chukar) fed under heat stress (HS) conditions on fattening performance and meat quality. This study included 256 chukar partridges aged 1 day. The study comprised an adaptation period of 7 days and a fattening period of 35 days and included 8 different groups with 32 animals in each group. Each group was further divided into four subgroups with eight animals in each subgroup. At the end of the study, the best results in terms of fattening performance in the non-HS groups were obtained in the LRE 600 ppm group and in the HS groups, SLRE 200 ppm had the best effect on average live weight and average live weight increase, whereas SLRE 400 ppm had the best effect on FCR (p<0.05). The analysis of the samples collected from chukar partridges on day 21 showed that, there was no effect on the colour parameters and Thiobarbituric acid reactive substances (TBARS) level in the LRE in the HS and non-HS groups, whereas the meat pH level decreased in the SLRE 400 ppm group (p<0.05). The analysis of the samples collected on day 42 showed that there was no effect on colour parameters in the HS and non-HS groups. TBARS level decreased at the dose of LRE 200 ppm in the non-HS group, and the meat pH level decreased in both HS and non-HS groups (p<0.05).

The impact of dietary tarragon (Artemisia dracunculus) on serum apelin, brain-derived neurotrophic factor, cardiac troponin concentrations and histopathology of liver tissue in laying hens housed at different stocking densities

Bayraktar, Bulent; Tekce, Emre; Kaya, Hacer; Karaalp, Musa; Turunc, Eylul

Due to its association with several other stress factors (poultry house gases, inadequate ventilation, heat, cold and poor hygiene), the high stocking density is a major stress factor that adversely affects the health and performance of poultry and the quality of the poultry products. Therefore, this experimental study was aimed at analysing the impact of different doses of dietary tarragon (Artemisia dracunculus) on the serum apelin, plasma brain-derived neurotrophic factor (p-BDNF), and cardiac troponin I (cTnI) concentrations, and the correlation between these indicators in laying hens housed at different stocking densities. The aim of this study is to investigate the effects of adding tarragon in different ratios to laying hen rations in the 2ndovulation period on the cTnI, apelin, and BDNF hormone concentrations and the liver histopathology. The experiment was carried out over a period of eight weeks, with 192 Lohman Brown commercial hybrids at 50 weeks of age. Eight groups (four replicates each), composed of laying hens of equal body weight, which were housed at stocking densities of 580 cm2/hen and 810 cm2/hen and received 0, 1, 5 and 10 mmol/kg of tarragon (Artemisia dracunculus) in the feed, were established. At the end of the trial, 96 of the housed egg-laying hens (3 birds in each subgroup, a total of 12 birds in each group) were randomly selected and blood samples were taken from the vena subcutanea ulnaris. The samples collected were analysed for the apelin, p-BDNF, and cTnI contents. The analysis results demonstrated that tarragon supplementation had no effect on the serum apelin, p-BDNF and cTnI concentrations (P > 0.05). The Sub-Groups ST1, ST1.2, and ST6 presented with severe hyperaemia of the sinusoidal, portal and acinar blood vessels, whilst the hyperaemia of these blood vessels was moderate in Sub-Group ST12. Apelin, BDNF, and cTnI can act as protective factors against negative consequences of stress (e.g., stocking density or heat stress).

Bayraktar, Bülent; Tekce, Emre; Aksakal, Vecihi; Takma, Çiğdem; Bayraktar, Fatma Gülten; Şengül, Bülent

Apelin is an important adipokine hormone that is released from adipose tissue, which is considered as the energy store of the body, which plays a role in many physiological processes in the body, as well as cardiovascular, immune functions and energy, nutrients and fluid metabolism. In this study, it was aimed to determine the effect of lactation, pregnancy and gender on apelin hormone levels in blood serums belonging to different races of ewe. In the present study, the hormone levels of the pregnant, non-pregnant ewe and rams of the Akkaraman Kangal and Morkaraman races were thin, with different body condition scores (<2, 3 to 3.5 and ≥4). Apelin hormone level was determined by ELISA technique in blood serum samples of ewe's Jugular vein. It was determined that there was a difference in body scores between races and the interaction between race and body score was important (P<0.05). Apelin level in ewe in lactation and pregnant ewe did not change according to body condition scores (P>0.05). In terms of apelin, gender and body condition score, race and gender, and body condition score interactions were found to be significant.

Characterization of pediococcus acidilactici PFC69 and lactococcus lactis PFC77 bacteriocins and their antimicrobial activities in tarhana fermentation

    Kaya, Halil İbrahim; Şimşek, Ömer

Tarhana is a traditional cereal product fermented by lactic acid bacteria (LAB) and yeast strains that has gained special interest recently as an infant nutrition. Tarhana contains wheat flour, yogurt, and various vegetables that might create a microbiological toxicological risk, especially for Bacillus cereus and Staphylococcus aureus. In this study, characterization of the metabolites responsible for antibacterial activity of Pediococcus acidilactici PFC69 and Lactococcus lactis PFC77 strains obtained from tarhana was performed, and antibacterial effects were detected against B. cereus ATCC 11778 and S. aureus ATCC 29213 during the fermentation. A total of 12,800 AU/mL antibacterial activity was observed for the supernatants of the PFC69 and PFC77 strains that were found to be stable at high temperature and in low pH conditions and sensitive to proteases, suggesting the antimicrobial metabolite is a bacteriocin. These bacteriocins were further purified and their molecular sizes were determined as 4.5 and 3.5 kDa, respectively. Importantly, inoculation of PFC69 and PFC77 to tarhana dough significantly decreased B. cereus ATCC 11778 and S. aureus ATCC 29213 amounts from the fifth day of fermentation compared to the control dough samples. P. acidilactici PFC69 and L. lactis PFC77 strains were concluded as bioprotective cultures for tarhana and these strains were offered for other cereal-based fermentations.

Optimization of cryoprotectant formulation to enhance the viability of Lactobacillus brevis ED25: Determination of storage stability and acidification kinetics in sourdough

Gul, Latife Betul; Gul, Osman; Yilmaz, Mustafa Tahsin; Dertli, Enes; Con, Ahmet Hilmi

In this study, various kinds of cryoprotectant (skim milk, lactose, and sucrose) formulations were tested to enhance the survival of Lactobacillus brevis ED25 after freezing and freeze-drying. A Box–Behnken experimental design was used to optimize cryoprotective medium and the highest cell survival was observed with the 17.28% skim milk, 2.12% lactose, and 10% sucrose cryoprotectant as the optimum condition. The structural and physicochemical characteristics of freeze-dried powder were acceptable for application with regards to particle surface morphology, moisture and water activity (Aw), glass transition temperature (Tg), Fourier transform infrared spectra, X-ray structure, and also storage stability under the refrigeration and room temperature conditions. Accelerated storage test based on Arrhenius equation could be used to predict the freeze-dried bacterial shelf life but only with a certain degree of predictability for long-term storage. The acidification kinetics of fresh and stored culture in sourdough fermentation was also described on the basis of the Gompertz equation. Practical applications: Freezing and storage are crucial factors for the viability and acidification power of starter culture. Therefore, various types and concentrations of cryoprotectants have been used to preserve the microorganisms. L. brevis ED25 has been a good potential for the manufacture of industrial sourdoughs and this research has aimed to investigate long-term protective effects of optimum cryoprotectant formulations on the viability of bacteria and also determine the acidification power in sourdough. The results showed the potential value of freeze-dried L. brevis ED25 culture for commercialization.

The effects of adding garlic (Allium sativum L.) on the volatile composition and quality properties of yogurt

    Arslaner, Ayla

In this study, the effects of adding garlic (Allium sativum L.) of different origins on the volatile composition and quality properties of yogurt were investigated on day 1, 7 and 14 of storage. The addition of garlic did not cause a decrease in the total specific microorganism count (>107) in yogurt samples, and it led to a significant decrease in the yeast–mould count compared to the control. Seventy-four volatile compounds were identified in the samples, consisting of 15 carbonyl compounds, 9 aliphatic hydrocarbons, 8 acids, 3 alcohols, 6 esters, 12 sulfur-containing compounds, 12 terpenes and 9 heterocyclic compounds. Acetaldehyde, 2,3-butanedione, acetoin, hexanoic acid, tetramethylthiourea and 1,2-dihydro-2,2,4-trimethyl-quinoline were the major volatile compounds found in all yogurt samples. Although they were poor in terms of terpenes compared to the control, strong sulfurous components were detected in yogurt samples with garlic. It has been determined that the addition of garlic of different origins significantly changed the aromatic profile of the yogurt. In the sensory analyses, 1.0% domestic garlic was the sample most preferred by the panelists.

Portable infrared sensing technology for phenotyping chemical traits in fresh market tomatoes

Akpolat, Hacer; Barineau, Mark; Jackson, Keith A.; Aykas, Didem P.; Rodriguez-Saona, Luis E.

Our objective was to develop predictive regression algorithms based on infrared spectroscopy to screen for selected quality traits directed at optimizing the selection capabilities of fresh market tomatoes. Fresh tomato (681) samples were harvested from multiple locations (Florida, Virginia, California and South Carolina) during the 2016 and 2018 seasons at various ripening stages. Spectra were collected by transmittance and attenuated total reflectance (ATR) either from the tomato surface or juice. Reference methods included soluble solid content, titratable acidity, sugars, organic acids, and lycopene. Partial least squares regression using surface spectra showed good correlation for lycopene and ascorbic acid (rcv>0.9) but modest correlation coefficients (rcv 0.54–0.80) for all other traits, while juice spectra gave high correlation coefficients (rcv>0.94) and excellent predictive performance (RPD range 3–10) for all quality traits except ascorbic acid (rcv>0.79). Multiple quality traits were simultaneously determined by using a single drop of sample providing fast (<1 min) measurements and minimal sample preparation based on unique spectral fingerprints.

Diversity and functional characteristics of lactic acid bacteria from traditional kefir grains

Purutoğlu, Kübra; İspirli, Hümeyra; Yüzer, Mustafa Onur; Serencam, Hüseyin; Dertli, Enes

Traditional kefir grains were collected from distinct parts of Turkey, and their microbial profile was determined. A wide bacterial biota was observed formed by distinct lactic acid bacteria (LAB) in which Lactococcus lactis strains appeared to be dominant. Yeast species were also identified in kefir grains. Significant levels of antifungal and antibacterial activities were monitored in kefir isolates. All tested LAB produced an exopolysaccharide (EPS) containing glucose and galactose, and some strains formed a fructan-type EPS. Importantly, low levels of antibiotic resistance were observed among the kefir isolates.

Influence of lactobacillus reuteri on internal organ weight, performance and meat quality of Japanese quail (Coturnixcoturnix japonica) under heat stress; [Einfluss von lactobacillus reuteri auf das gewicht der inneren organe, die leistung und die fleischqualität Japanischer Wachteln (Coturnix coturnix japonica) unter Hitzestress]

    Tekce, E.; Bayraktar, B.; Aksakal, V.; Dertli, E.; Kamiloğlu, A.; Çinar Topçu, K.; Kaya, H.; Takma, C.; Yasulergezer, N.

In the present study, the effects of Lactobacillus reuteri E81 (LRE) supplemented at different doses (200, 400 and 600 mg/kg) into the diets on performance, meat quality and internal organ weights of Japanese quail (Coturnixcoturnix japonica) were investigated under heat stress (HS) conditions (37°C). For the experiments, 448 one-day old quails were used. Seven days of training (physical exercise) and a 35-day fattening period were applied. On day 7 of the trial, 8 groups [4 under control temperature of 25°C (CT): control (C), LRE-200, LRE-400 and LRE-600 and 4 under HS: control (HSC), SLRE-200, SLRE-400 and SLRE-600] were formed, each group consisting of four replicates with 14 quail each. At the end of this study, the CT group fed 400 mg/kg LRE showed the best performance results, whereas no effect was found for groups under HS in comparison to the control group (HSC). Samples taken on the 21st and 42nd day revealed that addition of probiotics in the CT and HS groups induced no effect on weight of the internal organs (p > 0.05). Analysis of meat quality on the 21st day showed that LRE caused an increase in pH in both the HS and CT groups. Further, 400-mg/kg LRE supplementation induced an increase in thiobarbituric acid reactive substances (TBARS) in the group under HS. In the CT groups on day 42 no effect of LRE was found on colour parameters or on the TBARS value, except for a decrease in the pH value in the LRE supplemented groups compared with the control. In HS groups a decrease in the L* value and an increase in the TBARS value was found compared to CT groups (p < 0.05), but there was no effect on other colour parameters or pH value. In conclusion, in the present study supplementation of different doses of LRE E81 to quail diets had no effect on performance, internal organ weight or meat quality under HS.

Response of Japanese quails (Coturnix coturnix japonica) to dietary inclusion of Moringa oleifera essential oil under heat stress condition

Tekce, Emre; Bayraktar, Bülent; Aksakal, Vecihi; Dertli, Enes; Kamiloğlu, Aybike; Çinar Topcu, Kübra; Takma, Çiğdem; Gül, Mehmet; Kaya, Hacer

The present study was aimed at investigating the effects of the dietary use of different doses of Moringa oleifera essential oil (MOEO) on performance parameters, internal organ weights and meat quality in Japanese quails (Coturnix coturnix japonica) exposed to Heat Stress (HS). For this purpose, 320 male day-old quail chicks were used. Seven days of preliminary (physical exercise) and a 35-day fattening period were applied. The animals were divided into 8 groups, each including 40 animals according to temparature (C: 25 °C and SC: 37 °C) and dietary doses (C, 200, 400 and 600). Each group was divided into 4 subgroups, each comprising 10 animals. The consequences of this study showed that, in the stress-free (HSF) groups, while all of the different doses of dietary MOEO positively affected growth performance, feed intake and feed efficiency, in the HS groups, similar positive effects were observed with the use of 200 ppm of dietary MOEO. While no effect was detected on internal organ weights on Days 21 and 42 in the HSF and HS groups, a dose-dependent increase was determined in the intestinal pH value on day 42 in the HS group (p <.05). Additionally, in the analysis of meat quality on the 21st and 42 days compared to the controls, no effect was observed on the meat colour parameters value and TBARS level in the HS and HSF groups. Based on the consequences of this study, it is considered that, MOEO could be used as an alternative product in poultry, at a dose of 200 ppm, to reduce the detrimental effects of HS.HIGHLIGHTS Temperature stress, performance parameters and serious problems in the health of poultry. Moringa olifera helps regulate performance parameters in poultry exposed to heat stress. Since the addition of Moringa oleifera to the diets does not affect the organ weight and meat quality in 21 and 42 days, it does not cause any negative effects for the consumer.

The purpose of this study was to examine the effect of adding an essential fatty acid mixture (EOM; Eucalyptus globulus Labill, thymus vulgaris, Cymbopogon nardus, and Syzygium aromaticum) to the drinking water in the heat stress broilers on the adipokine (Apelin, BDNF) response, the histopathologic changes in liver and intestines, and some biochemical parameters. This study lasted for a total of 42days, including the physical exercise period (7days) and the fattening period (35days). A total of 400 Ross 308 male broilers (1-d-old) were randomly divided into 8 groups (50 animals per group), each of which was exposed to various conditions of temperature (C: 22°C and SC: 36°C) and treatment dose (C, 250, 500 and 750mL/1000 L). Each group was divided into 5 subgroups, each comprising 10 animals. In the stress-free groups, whereas the Apelin level linearly decreased, the BDNF level linearly increased. In histopathology, the liver tissue was found to be normal in all groups whereas the duodenum villi length was found to increase in the group of 750mL/1000 L. No statistically significant difference was found between the stressed groups and the non-stressed groups in terms of VLDL, Glucose, Total bilirubin, ALT, and TG (p >.05). While Apelin level increased in the stressed groups, the BDNF level increased in the group of 250mL/1000 L. In the histopathological examination, a small amount of coagulation necrosis was detected in hepatocyte, a diffuse hydropic degeneration was observed in hepatocytes, and finally a dilatation and hyperaemia were seen in sinusoid in the groups of EOM-500mL/1000 L and EOM-750 m/1000 L compared to the control group. Whereas there was no difference between the group of EOM-250mL/1000 L and the control group in terms of duodenum villa length, there was a significant decrease in other groups (p <.00). In conclusion, this study showed that adding 250mL/1000 L of EOM to the drinking water had a positive effect on the serum levels of Apelin and p-BDNF in the groups exposed to stress (p <.05).HIGHLIGHTS Heat stress results in serious annual economic losses in the poultry industry. Essential oil fatty mix acids have positive effects on poultry health. Apelin and BDNF may be a protective factor against deleterious consequences of chronic exposition to heat stress.

Characterisation of physicochemical, microbiological, thermal, oxidation properties and fatty acid composition of butter produced from thermosonicated cream

    Sert, Durmuş; Mercan, Emin

The effects of thermosonication of cream on physicochemical, microbiological, thermal, oxidation properties and fatty acid composition of butter were determined. Butter samples were produced from cream thermosonicated (TS) for 5, 10, or 15 min and from raw (R) and pasteurised (P) cream as controls. Thermosonication decreased moisture content, water activity, and pH of samples when compared with control groups. Mean particle size (D [3.2]) values of TS samples were lower than the R and P samples. Thermosonication improved spreadability compared with that of butter from pasteurised cream. Oxidative stability (induction time) of TS samples was lower than R, but higher than P. Two melting peaks were determined; in all samples, the Tonset of the first peak ranged from 11.39 to 11.93 °C, and the Tpeak was between 14.01 and 14.14 °C. Thermosonication significantly reduced total bacteria count and can improve spreadability, oxidative stability, and microbial safety of butter samples.

Technological properties of autochthonous Lactobacillus plantarum strains isolated from sucuk (Turkish dry-fermented sausage)

Kamiloğlu, Aybike; Kaban, Güzin; Kaya, Mükerrem

Purpose: Five Lactobacillus strains isolated from sucuk (Turkish dry-fermented sausage) were studied for their genetic and technological properties. Methods: For genotypic identification, strains 16S rRNA gene sequences were used. To determine the antimicrobial activity of strains, seven foodborne pathogens were tested. Strains technological properties were characterized. Results: These strains were identified as Lactobacillus plantarum by 16S rRNA gene sequence analysis and the phylogenetic tree obtained by neighbor-joining method allowed grouping of these strains into three subgroups. L. plantarum strains showed antagonistic activities against Staphylococcus aureus, Listeria monocytogenes, Bacillus cereus, and Micrococcus luteus strains. PCR assay, using specific primers, showed the presence of bacteriocin (plantaricin) encoding genes in all L. plantarum strains tested. Antimicrobial metabolite production of these strains started at log phase and reached the maximum level at the end of the stationary phase. Regarding their technological properties, better growth was observed at 25 °C compared with 15 °C and 45 °C. The isolates which grown well within the pH scale pH 4.5–6.5 range additionally showed a decent growth at 6.5% salt concentration. It has been found that strains do not exhibit lipolytic and proteolytic activities nor have lysine, ornithine, and arginine decarboxylase activity. On the other hand, one strain showed weak nitrate reductase activity, and four strains produced acetoin from glucose. In addition, all strains were DL-lactic acid producers. Consequently, L. plantarum strains isolated exhibited some biochemical properties required for a starter culture in sucuk and similar products. Conclusions: All identified strains may be a protective culture in the production of fermented meat products. In particular, L. plantarum S51 was distinguished from other isolates due to the inability to form acetoin from glucose. Further work will be needed to characterize L. plantarum strains as starter culture.

Impact of glucan type exopolysaccharide (EPS) production on technological characteristics of sourdough bread

İspirli, Hümeyra; Özmen, D.; Yılmaz, Mustafa Tahsin; Sağdıç, O.; Dertli, Enes

In this study, technological role of α-glucan with (α1-3) and (α1-6) glycosidic linkages produced by Lactobacillus reuteri E81 on sourdough rheology and bread texture was investigated. To achieve this, a glucan deficient ΔgtfA mutant was produced by the deletion of the glucansucrase gene from strain E81. By comparison of glucan-positive and glucan-negative strains for rheological properties of sourdough revealed that in situ α-glucan production resulted in higher elasticity in sourdough. However, no additional effect of in situ glucan on textural characteristics of bread was observed although addition of glucan ex situ at 0.09% level significantly decreased bread hardness. Scanning Electron Microscopy (SEM) images of lyophilized sourdoughs confirmed association of glucan and starch granules forming a film like structure in sourdough.
