Ecem Bayram, Nesrin; Celik, Saffet
Bee pollen is a bee product that is widely preferred by people for its nutritional properties as well as its health-protective properties, and is defined as a ‘superfood’. In this preliminary exploratory study, monofloral Rhododendron ponticum bee pollen was analyzed using a label-free LC-QTOF-MS proteomic approach. According to the results obtained, a total of 37 proteins from the rhododendron plant have been fully confirmed in the pollen sample. Normalized spectrum counting values (NSAF) providing information about the semi-quantitative amount of these proteins were determined to be between 120.84 and 43259.22. Based on these findings, it can be said that the proteomic approach can provide biomarkers for monitoring the botanical source and quality of monofloral bee pollen. However, since no biological or technical replicate study was conducted, the findings obtained are exploratory in nature and should be considered semi-quantitative. To our knowledge, this study is the first to report the protein profile of R. ponticum bee pollen using a proteomic approach.

Morgil, Hande; Ecem Bayram, Nesrin
The plant origin of the bee products often influences their value. Here, we investigated for the first time how the content of bee bread, a functional food with high nutritional value, differs in terms of plant and bacterial taxa in comparison with bee pollen and honey samples, using shotgun sequencing analysis. Sequencing of honey, bee pollen, and bee bread samples yielded approximately 32.74 Gbp of data. Out of a total of 268,964 abundant genes, 70,280 genes (26.13%) were assigned to a taxonomic group according to the supermajority rule. These taxonomic groups were annotated as follows: Kingdom (84.99%), phylum (75.94%), class (73.26%), order (67.21%), and family (65.11%). It was determined that honey, bee pollen, and bee bread samples consisted of large plant families Poaceae, Fabaceae and Asteraceae. This indicates that these taxa are also favored by bees for visiting, and are therefore used as food sources. These differences vary according to the environmental flora of the apiary and may be site specific. Based on these results, DNA-based next-generation sequencing techniques can be successfully used to identify the plant sources of bee products. Thus, the botanical and geographical origin of bee product samples can be more accurately, reliably and in detail identified. Furthermore, this technique can be successfully used to monitor the regional flora and food safety.

The aim of this study was to characterize Ranunculus pinardii and Ranunculus constantinopolitanus species anatomically and phytochemically. In order to contribute to the taxonomic characterization of R. pinardii and R. constantinopolitanus, anatomical structures of the stem, leaf, and petiole of species were examined with light microscopy. Total phenolic-flavonoid content, antioxidant activity, total anthocyanin, total proanthocyanidin, and individual phenolic compounds of extracts prepared with different solvents were determined. The results showed that the total phenolic contents of the species had a similar range (0.09-0.45 mg GAE/g for R. pinardii and 0.01-0.44 mg GAE/g for R. constantinopolitanus), but R. constantinopolitanus exhibited higher antioxidant activity (29.28 mg TE/g). Higher individual phenolic compound recovery was achieved from R. pinardii compared to R. constantinopolitanus with all solvents, except methanol. Both species were rich in luteolin, a natural flavonoid (163.98-34417.11 µg/kg for R. pinardii and 44.93-129009.44 µg/kg for R. constantinopolitanus). These results indicate that these plants contain bioactive ingredients that may have applications in different fields, such as medicine, cosmetics, and food. In particular, the detection of luteolin compound at high levels in these species compared to other components indicates that optimization studies aimed at the recovery of luteolin are important for future research.

Deep eutectic solvents (DESs) have emerged as green alternatives to conventional solvents for extracting bioactive compounds from plants. This study evaluated nine choline chloride–based DESs for recovering antioxidant polyphenols from four parts of the tea plant (Camellia sinensis): seed, bud, leaf, and flower. Total phenolic content, antioxidant activity, and individual polyphenolic profiles were determined, and the greenness of the sample preparation was assessed using AGREEprep. The highest total phenolic content (53.28 mg GAE/g) and antioxidant activity (141.17 mg TE/g) were obtained from leaves using ChCl:formic acid, outperforming ethanol and methanol. Leaves also exhibited the highest total quantified polyphenolic content (up to 95,813 mg/kg DW, corresponding to the sum of the quantified polyphenols), with epicatechin gallate and gallocatechin + epigallocatechin being the major contributors. Flowers and buds showed notable polyphenolic diversity, while seeds contained the lowest levels. Statistical analysis confirmed clear clustering patterns according to plant part and solvent type. AGREEprep evaluation yielded scores up to 0.54, highlighting the environmental benefits of DES-based extraction. We demonstrated that tea leaves were the richest source of bioactive polyphenols, and selected DESs, particularly formic acid–based and ethylene glycol–based, exhibited superior efficiency and sustainability compared to conventional solvents. These findings support the wider adoption of DESs for green, high-yield extraction of pharmacologically important compounds from tea and other medicinal plants.
- MakaleYalnız künye2026Scopus SJRQ2Optimizing microwave drying parameters for rhododendron bee pollen: effects on antioxidant potential, polyphenols, and proteins
Kutlu, Naciye; Gercek, Yusuf Can; Ecem Bayram, Nesrin
Monofloral Rhododendron spp. bee pollen is a remarkable bee product due to its functional components as well as its potential pharmacological effects. In this study, Rhododendron spp. bee pollen was dried by the microwave method as an alternative to the conventional drying method. Different combinations of microwave powers (140, 210 and 280 W) and sample weights (10, 20 and 30 g) applied during microwave drying (MWD) were determined by Central Composite Design. Drying time, total protein, total carotenoid, total phenolic-flavonoid compounds, and proanthocyanidin yields of dried pollen were determined and antioxidant activity tests (CUPRAC and DPPH•) were performed. The analysis results were optimized by response surface methodology (RSM) and the optimum drying conditions were obtained when the microwave power was 280 W and the sample weight was 30 g. The optimal conditions have been established to reduce drying time and enhance the content of bioactive compounds. As a result of optimization, the composite desirability (d) was found to be 0.8. Experimental and predicted values were close to each other. In conclusion, MWD has demonstrated efficacy as a drying technique by significantly reducing drying time while maintaining and enhancing the yields of bioactive compounds in monofloral Rhododendron spp. bee pollen. In comparison to conventional techniques, the application of MWD in the drying process of monofloral Rhododendron spp. bee pollen is demonstrably more efficient and sustainable.

Deep eutectic solvents (DESs) have been frequently preferred over the past decade as alternatives to conventional solvents for the extraction of bioactive compounds from natural products. Meanwhile, black and red goji berries are widely consumed as functional foods due to their many functional properties reported in the literature. The aim of this study is to evaluate the effectiveness of different DESs in the extraction of individual phenolic compounds from goji berry (Lycium barbarum and L. ruthenicum) fruits, to determine the most suitable DES, and to optimize the extraction conditions. The DES that provided the highest yield in the extraction of individual phenolic compounds from L. barbarum was determined to be proline:citric acid (DES6 – a total of approximately 33,545 µg/kg), while for L. ruthenicum it was choline chloride:acetic acid (DES3 – a total of approximately 37,496 µg/kg). Rutin was the predominant phenolic compound in both species. DESs provided a higher phenolic yield compared to ethanol and water. In addition, L. ruthenicum exhibited higher total phenolics, flavonoids, proanthocyanidins, and antioxidant activity compared to L. barbarum. These findings reveal that DESs are a green, efficient, and sustainable alternative for the extraction of phenolic compounds from goji berries.

This study aimed to extract bioactive compounds from goji berry (Lycium barbarum L.) fruit using the ohmic heating–assisted extraction (OHAE) method and optimize the extraction conditions. In this context, firstly, the extraction conditions to be applied for the OHAE method were determined using the response surface method (RSM)-Box Behnken experimental design. During the study, temperature, holding time, and electrical field strength were determined as independent variables, while the total phenolic compound (TPC), total flavonoid compound (TFC), and antioxidant activity (DPPH) were selected as dependent variables. As a result of the optimization, the optimum conditions were determined to be 60 °C for the temperature, 60 min for the holding time, and 45 V/cm for the electrical field strength, and the composite desirability value of the model was 0.90. The highest experimental results for TPC, TFC, and DPPH were found to be 23.83 mg GAE/g, 7.33 mg QE/g, and 24.53 mg TE/g, respectively. In addition, the polyphenolic profiles of all extracts obtained under different process conditions were determined by LC–MS/MS, and rutin was the phenolic detected at the highest rate in the extracts. In addition, the blueness and greenness of the analytical method used in this study were evaluated using the BAGI and ComplexGAPI metrics, respectively. The results indicate that OHAE is an effective and sustainable method for extracting bioactive compounds from goji berries.

Deep eutectic solvents (DESs) have been widely used in food processing in recent years, owing to their remarkable advantages. The aim of this study was to evaluate the potential of DESs for the recovery of bioactive components from chokeberry fruit. First, among the 10 different DESs, the DES that extracted the highest total individual phenolic content (TIPC) from chokeberry was determined. Then, ultrasonically assisted extraction conditions (ultrasonic amplitude, time, and temperature) were optimized by response surface methodology to obtain the maximum amount of TIPC using this DES. According to the results obtained, the natural deep eutectic solvent (NaDES) choline chloride: acetic acid (1:2, 30% water content) provided the highest yield for TIPC extraction and the optimum conditions for maximum TIPC extraction were determined as 100% for ultrasonic amplitude, 50 °C for temperature and 60 min for sonication time. The TIPC content of the extracts prepared with this NaDES was determined between 1740.26 and 3703.14 mg/kg. In addition, the individual organic acid and individual anthocyanin contents of the NaDES extracts, as well as their antioxidant and antimicrobial activities, were determined. Chlorogenic acid (1392.351–2361.48 mg/kg) among phenolic compounds, pyruvic acid (9361.35–47398.04 mg/kg) among organic acids and kuromanin chloride (69748.13–134668.24 mg/kg) among anthocyanins were the dominant compounds in NaDES extracts. However, the extracts showed varying levels of antimicrobial activity against Gram (+) bacteria, Gram (−) bacteria, and yeast-like fungi (Candida strains). Finally, the Blue Applicability Grade Index (BAGI) was used to evaluate the practicality/blueness potential of the chromatographic method used to determine the anthocyanin composition of the DES extracts. In conclusion, NaDES (choline chloride:acetic acid) can be an alternative solvent for the recovery of some individual phenolics, organic acids and anthocyanins under different extraction conditions. In addition, the use of NaDES in the recovery of various functional compounds needed in food, cosmetics, pharmaceuticals, etc., is important for the promotion of sustainability and green technology.

- MakaleAçık Erişim2025Scopus SJRQ2Extraction of Phenolic Compounds From Lavender (Lavandula angustifolia L.) Plant by Natural Deep Eutectic Solvent and Greenness Assessment of Analytical Method With Modified Green Analytical Procedure Index
Ecem Bayram, Nesrin; Yenigün, Baran; Gerçek, Yusuf Can
In recent years, it has been important in terms of green technology and sustainability to promote the use of natural deep eutectic solvents (NADESs), especially those based on natural products, for the extraction of bioactive components with different functional properties from foods. Therefore, in this study, the effects of green extraction technology (ultrasonic-assisted extraction) and green solvents (NADESs) on the recovery of bioactive components from lavender (Lavandula angustifolia L.) were investigated and evaluated. In addition, the degree of greenness of the analytical method used during the study was determined using the modified green analytical procedure index (MoGAPI). In this context, the total phenolic content (TPC), total antioxidant capacity, and individual polyphenolic profiles of lavender extracts prepared with two different NADES were investigated by liquid chromatography-tandem mass spectrophotometer. Based on the results obtained, it was determined that NADES formed with choline chloride:formic acid (ChCl:FA) had a higher potential to extract bioactive components from lavender than NADES formed with ChCl:ethylene glycol (ChCl:EG). The TPC, antioxidant capacity, and total individual polyphenolic compound content of lavender extracted with these NADES were determined as 9.76 mg gallic acid equivalent/g, 20.83–104.39 mg Trolox equivalent/g, and 200.835 mg/kg, respectively. However, the degree of greenness determined by the MoGAPI tool of the analytical method used in this study was 78% and classified as ‘excellent green’. As a result, it can be concluded that NADES formed with ChCl:FA can be an alternative to conventional solvents for the extraction of bioactive components from lavender, as it provides a remarkable recovery of bioactive substances.

Çelik, Saffet; Gerçek, Yusuf Can; Kutlu, Naciye; Bayram, Sinan; Kırkıncı, Seran; Ecem Bayram, Nesrin
In this study, different deep eutectic solvents (DESs) were used for vitamin extraction from bee pollen. The DES combination with the highest total amount of individual vitamins was choline chloride: ethylene glycol (molar ratio 1:2). The response surface method was then used to optimize the extraction conditions. The independent variables used for optimization were ultrasonic power, time, and temperature, while the dependent variables were the total individual amino acids, total individual phenolic compounds, and total individual vitamin contents. Spectrophotometric analyses and antimicrobial activity were also determined in the extracts. In addition, AGREEprep, ComplexGAPI and BAGI tools were used to evaluate the greenness of the method. The results obtained with DES were compared with water, ethanol, and methanol extracts, which were considered control groups. As a result of optimization, the optimum conditions were found to be 75 % for ultrasonic power, 71.5 min for sonication time, and 40 °C for ultrasonic bath temperature. The model's composite desirability value (d) was found to be 0.81. The predicted value and the experimental values were close to each other. Furthermore, the most dominant vitamin in DES extracts was ascorbic acid, the most dominant amino acid was proline, and the most dominant phenolic compound was myricetin. The amounts of these compounds were higher than those obtained from water, ethanol, and methanol extracts. The spectrophotometric analysis results obtained higher yields with DES compared to the control groups. Antimicrobial activity analysis showed that pollen extracts prepared using choline chloride: ethylene glycol had a weak inhibitory effect for Gram-positive and Gram-negative bacteria but not yeast-like fungal strains. In conclusion, DES has been successfully used to extract vitamins and other bioactive compounds from bee pollen. The results obtained showed that DESs can be an alternative to organic solvents for the extraction of vitamins, which are among the minor components of bee pollen. Besides, the potential of the application different DESs for more efficient extraction of vitamins and components with important pharmaceutical properties from bee pollen needs to be evaluated in detail.

Kutlu, Naciye; Gerçek, Yusuf Can; Çelik, Saffet; Bayram, Sinan; Ecem Bayram, Nesrin
In this study, the aim was to optimize the extraction conditions (molar ratio, time, temperature) in order to maximize the total individual amino acid (TAA) yield from bee bread, which is a fermented bee product, using choline chloride-acetic acid deep eutectic solvent (DES). In addition to the phenolic profile and amino acid profile investigated by LC-MS/MS, some spectrophotometric (total phenolic content, total flavonoid content, total proanthocyanidin content) and bioactivity (antioxidant/antimicrobial activity) tests were carried out on the extracts. Optimum conditions were determined as 1:2 for molar ratio, 15 min for time and 10 °C for temperature with optimization based on the TAA values of the different extracts (d: 0.99). Molar ratio and temperature were found to have statistically significant effects on TAA values. At optimum conditions, the amount of TAA obtained with DES (29.678 g/kg) was found to be approximately 35.2% and 54.4% higher than that obtained with ethanol and methanol extracts, respectively. All these results indicate that choline chloride-acetic acid DES can be chosen when preparing bee bread extracts, which are evaluated for different areas of use such as food and health due to active ingredient content. Higher active ingredients can be extracted by trying different DES combinations.

Bee bread is one of the basic nutrients that bees use as a protein source and is formed as a result of the fermentation of bee pollen. In the last few years, this product has become increasingly recognized and consumed among bee products due to its functional properties. However, deep eutectic solvents (DES) have attracted great interest in recent years as promising green alternatives to replace traditional solvents due to their variable viscosity, low vapor pressure, application-specific adjustability, non-flammability and chemical stability. In this study, bee bread was extracted in 12 different DES with different hydrogen bond acceptor (HBA):hydrogen bond donor (HBD) combinations. The DES combination with the highest total protein content was determined to be choline chloride (ChCl)-urea. Then, ultrasonic extraction conditions were optimized for maximum bioactive extraction in extracts prepared with ChCl-urea. The optimum conditions were found as 31.3 %, 25 min, 35 °C for ultrasonic power, extraction time and temperature, respectively. The desirability value (d) of the model was 0.83. The extracts were analyzed for total phenolic content, total protein content, individual polyphenolics, individual amino acids and individual vitamins as well as antimicrobial and antioxidant activity tests. According to the results, total protein, total individual amino acids and total individual phenolics were higher in the extracts obtained with ChCl-urea compared to the extract prepared with conventional solvent (ethanol). In conclusion, all these findings showed that ChCl-urea combination can be an alternative to ethanol depending on the structural characteristics of the target compounds to be extracted from bee bread.

- MakaleYalnız künye2023Scopus SJRQ1Nectar honey from Turkey: crystallization and physicochemical profile
Ecem Bayram, Nesrin
Honey is a traditional food used by humans due to many therapeutic properties. Turkey is in a good position for the production of bee products, and honey is the most popular bee product in the country. In this study, the aim was to evaluate the quality parameters of 40 honey samples based on physicochemical parameters and to gain information about speed of crystallization. For this purpose, moisture (%), fructose (%), glucose (%), fructose + glucose (F + G, %), fructose/glucose (F/G), glucose/moisture (G/M), and proline (mg/kg) contents were determined in honey samples and cluster analysis was performed. When the sugar profile of honey samples is examined, F + G was 61.63–78.03%, F/G was 0.99–1.25, moisture content was 14.9–19.1%, and proline content was 384.41–1271.56 mg/kg. These results comply with national standards. Honey samples tend to crystallize quickly when their crystallization properties are evaluated based on F/G and G/M ratios. Consequently, the quality of honey differs according to the region from which it is produced and it is natural for honey to crystallize.

- MakaleAçık Erişim2023Scopus SJRQ2Utilization of Response Surface Methodology in Optimization of Proline Extraction from Castanea sativa Honey
Ecem Bayram, Nesrin; Kutlu, Naciye; Can Gercek, Yusuf
Proline constitutes approximately 85 % of the amino acid composition of honey. Therefore, the quantitative determination of this amino acid in honey samples is used by many national/international authorities to evaluate the quality of honey types. In this study, it was aimed to achieve maximum proline amino acid extraction from honey samples whose botanical origins were confirmed by melissopalynological analysis. For this reason, based on three different spectrophotometric methods used in the literature for proline analysis, proline extraction was optimized with the Response Surface Method (RSM) and Box-Behnken experimental design. Three independent variables were determined as treatment time (2, 6, and 10 min), treatment temperature (22, 46, and 70 °C), and cooling time (5, 25, and 45 min). As a result of the optimization, it was seen that only significantly effective independent variable on the proline content of honey was the processing temperature. The optimum conditions obtained as a result of the RSM were found to be 2 min for the treatment time, 70 °C for the treatment temperature and 45 min for the cooling time. The composite desirability of the optimum conditions (R2) was found to be 1.00. It was determined that the method proposed by International Honey Commission (IHC) is efficient for proline analysis, but it provides more proline extraction by reducing of time from 10 min to 2 min in hold time in boiling water bath only during the extraction step. As a result, the conditions to be used in order to achieve maximum proline extraction with different spectrophotometric methods were determined and optimum values were determined. In addition, since the botanical origin of honey samples significantly affects the proline content of honey, it can be suggested that this study be optimized for different monofloral honey samples as well.

Honey is one of the important natural products used for apitherapeutic purposes as well as food consumption. The authenticity of the products to be used for apitherapy is very important. Botanical origin is one of the most critical factors affecting the characterization of bee products, especially honey. Therefore, melissopalynological analysis is essential in honey samples. In this study, the pollen morphologies of four different Rhododendron taxa, the origin of the monofloral "Mad honey" used for apitherapeutic purposes in Turkey, were examined. Five different measurement parameters have been used to investigate the pollen morphology of the plants. As a result, it can be said that pollen morphology has an important place in confirming the botanical origin of “Mad honey”.

In recent years, the popularity of natural products and natural product formulations has increased due to their beneficial effects on health. As a part of this trend, Lamiaceae family members have attracted attention due to their rich bioactive properties. Although the Stachys rizeensis R.Bhattacharjee plant is an endemic plant species in Turkey, no studies focus on its characterization. Hence, the present study aims to demonstrate the anatomical, morphological and phytochemical characterization of S. rizeensis in Turkey. In the first stage of the study, anatomical and trichome morphological characterization was performed to contribute to the taxonomic definition of S. rizeensis was found to be anatomically different from some other previously studied Stachys sp. by showing lacunar collenchyma type, uncertain cambium at the corners, two or three rows of endodermis in certain areas in the stem and two rows of palisade parenchyma, capitate and peltate glandular trichomes in the leaf. In the second stage, the phenolic profile, total phenolic/flavonoid content and antioxidant activity of the plant extracts prepared using solvents with different polarities (ethanol, methanol, water, ethyl acetate and hexane) were tested to determine the phytochemical content of the plant. The phytochemical analysis supported by chemometric analysis (PCA and HCA) revealed that methanol was the most efficient organic solvent for the extraction of the phytochemical compounds of the plant. Additionally, it was observed that chlorogenic acid (1842.40–217,086.19 mg/kg), verbascoside (2591.43–204,429.81 mg/kg), luteolin (3579.56–88,636.14 mg/kg) and kaempferol (9706.30–50,340.93 mg/kg) compounds were present at maximum levels in this plant, in which 31 different phenolic compounds were scanned using LC-MS/MS. In addition, results obtained from the phytochemical analysis revealed that S. rizeensis is a valuable plant that can be used for medical protection/treatment and that it could also have a variety of culinary applications. The present study served as the first original report regarding the chemical profile and anatomical characteristics of S. rizeensis (Lamiaceae) from Turkey.
