Structural and bioactive characteristics of a dextran produced by Lactobacillus kunkeei AK1

Yilmaz, Mustafa Tahsin; İspirli, Hümeyra; Taylan, Osman; Bilgrami, Anwar L.; Dertli, Enes

In this study, structural and techno-functional characteristics of an exopolysaccharide (EPS) produced by Lactobacillus kunkeei AK1 were determined. High-performance liquid chromatography (HPLC) analysis demonstrated that EPS AK1 was composed of only glucose units. 1H and 13C Nuclear magnetic resonance (NMR) analysis revealed that EPS AK1 was a dextran type EPS containing 4.78% (1 → 4)-linked α-D-glucose branches. The molecular weight of EPS AK1 was determined to be 45 kDa by Gel Permeation Chromatography (GPC) analysis. A high level of thermal stability up to 280 °C was determined for dextran AK1 detected by Differential scanning calorimetry (DSC) and Thermogravimetric analysis (TGA). Dextran AK1 appeared as regular spheres with compact morphology and as irregular particles in the solution with no clear cross-linking between the chains of the polysaccharide observed by Scanning electron microscopy (SEM) and Atomic force microscopy (AFM) analysis, respectively. X-ray diffraction analysis (XRD) analysis demonstrated that dextran AK1 had a crystalline structure. A relatively strong antioxidant activity was observed for dextran AK1 determined by 2,2-diphenyl-1-picrylhydrazyl (DPPH), 2,2′-azino-bis-3-ethylbenzothiazoline-6-sulfonic acid (ABTS) radical scavenging and cupric reducing antioxidant capacity (CUPRAC) tests. Finally, only a digestion ratio of 3.1% was observed for dextran AK1 following the in vitro simulated gastric digestion test.

    Akkoyunlu, Ebubekir; Ayazoglu, Rabil

In the present paper, we investigate the existence of multiple solutions for the nonhomogeneous fractional p-Kirchhoff equation M(∬R2N|u(x)-u(y)|p|x-y|N+psdxdy+∫RNV(x)|u|pdx)×((-Δ)psu+V(x)|u|p-2u)=f(x,u)inRN,where (-Δ)ps is the fractional p-Laplacian operator, 0 < s< 1 < p< ∞ with sp< N, M:R0+→R0+ is a nonnegative, continuous and increasing Kirchhoff function, the nonlinearity f: RN× R→ R is a Carathéodory function that obeys some conditions which will be stated later and V∈ C(RN, R+) is a non-negative potential function. We first establish the Bartsch–Pankov–Wang type compact embedding theorem for the fractional Sobolev spaces. Then multiplicity results are obtained by using the variational method, (S+) mapping theory and Krasnoselskii’s genus theory.

High-throughput phenotyping approach for screening major carotenoids of tomato by handheld raman spectroscopy using chemometric methods

Akpolat, Hacer; Barineau, Mark; Jackson, Keith A.; Akpolat, Mehmet Z.; Francis, David M.; Chen, Yu-Ju; Rodriguez-Saona, Luis E.

Our objective was to develop a rapid technique for the non-invasive profiling and quantification of major tomato carotenoids using handheld Raman spectroscopy combined with pattern recognition techniques. A total of 106 samples with varying carotenoid profiles were provided by the Ohio State University Tomato Breeding and Genetics program and Lipman Family Farms (Naples, FL, USA). Non-destructive measurement from the surface of tomatoes was performed by a handheld Raman spectrometer equipped with a 1064 nm excitation laser, and data analysis was performed using soft independent modelling of class analogy (SIMCA)), artificial neural network (ANN), and partial least squares regression (PLSR) for classification and quantification purposes. High-performance liquid chromatography (HPLC) and UV/visible spectrophotometry were used for profiling and quantification of major carotenoids. Seven groups were identified based on their carotenoid profile, and supervised classification by SIMCA and ANN clustered samples with 93% and 100% accuracy based on a validation test data, respectively. All-trans-lycopene and β-carotene levels were measured with a UV-visible spectrophotometer, and prediction models were developed using PLSR and ANN. Regression models developed with Raman spectra provided excellent prediction performance by ANN (rpre = 0.9, SEP = 1.1 mg/100 g) and PLSR (rpre = 0.87, SEP = 2.4 mg/100 g) for non-invasive determination of all-trans-lycopene in fruits. Although the number of samples were limited for β-carotene quantification, PLSR modeling showed promising results (rcv = 0.99, SECV = 0.28 mg/100 g). Non-destructive evaluation of tomato carotenoids can be useful for tomato breeders as a simple and rapid tool for developing new varieties with novel profiles and for separating orange varieties with distinct carotenoids (high in β-carotene and high in cis-lycopene).

Biocatalytic asymmetric synthesis of (S)-1-indanol using Lactobacillus paracasei BD71

Kalay, Erbay; Dertli, Enes; Şahin, Engin

Enantiopure benzo-fused cyclic alcohols have been used as a building block of a drug for Parkinson’s disease. Biocatalytic reduction of ketones is one of the most promising and significant routes to prepare optically active alcohols. In this study, the reductive capacity of seven lactic acid bacteria (LAB) strains were investigated as whole-cell biocatalyst in the enantioselective reduction of 1-indanone (1). Lactobacillus paracasei BD71 was found to have the best reductive capacity. Effects of different parameters such as pH, incubation time, agitation speed and temperature, on enantiomeric excess (ee) and conversion were investigated in a bioconversion. (S)-1-indanol ((S)-2) could be used as precursor for the synthesis of rasagiline mesylate TVP1012 for the therapy of Parkinson’s illness. It was produced in gram-scale (5.24 g), high yield (93%) and enantiomerically pure form using L. paracasei BD71 whole-cell biocatalysts. Also, to our knowledge, this is the first report on production of (S)-2 using whole-cell catalyst in enantiopure form, excellent yield, conversion and gram scale. This is a cheap, clean and eco-friendly process for production of (S)-2 compared to chemical processes.

Municipal solid waste-based district heating and electricity production: A case study

Tozlu, Alperen; Abusoglu, Aysegul; Ozahi, Emrah; Anvari-Moghaddam, Amjad

In this paper, municipal solid waste (MSW) based electricity production and district heating (DH) potential of Turkey are considered. Three MSW based waste-to-energy (WtE) scenarios is developed: (i) Scenario-I, a DH system integrated into a gas turbine power plant (GTPP), (ii) Scenario-II, a DH system integrated into an organic Rankine cycle (ORC), and (iii) Scenario-III, which is based solely on a DH system. As a result of the thermodynamic and thermoeconomic analyzes of these developed scenarios using an existing MSW-based cogeneration facility's actual operating data, the system with the most extended payback period (about 5 years) is found as the GTPP-DH system developed in Scenario-I, which also has the highest investment cost. On the other hand, the system with the shortest payback period (about 2 years) is found as the DH system developed in Scenario-III, which also has the lowest investment cost. Overall exergy efficiencies of the GTTP-DH, ORC-DH, and DH systems are found to be 41.86%, 16.15%, and 31.87%, respectively. When the developed WtE scenarios adapted to the pilot provinces selected from each geographical region of Turkey, it is found that the GTPP system developed in Scenario-I can increase the power generation capacity of MSW plants for each province by about 20%.

Multicultural education in Turkish social studies curriculum and textbooks

    Yılmaz, Sevda

Multicultural education is an educational approach that is guided by cultural components and is based on cultural diversity. Students are expected to gain skills and values in the education process with the social studies course, such as accepting cultural diversity, respecting human rights, and assessing different perspectives and ideas, all of which are crucial in multicultural education. From this perspective, it is clear that this notion of education is intertwined with the social studies course. Integrating a multicultural education approach into the social studies curriculum and textbooks is viewed as critical. This paper aims to determine the status of the multicultural education approach in both social studies curriculum and textbooks. The study employed the document analysis approach, which is one of the qualitative research methodologies. As data sources, the research used the 2018 social studies course curriculum as well as social studies textbooks for 4th, 5th, 6th, and 7th grade. The paper attempted to establish the status of how multicultural education is included in the curriculum and social studies textbooks. The results of the study demonstrate that the multicultural education approach is represented in the curriculum, that the multicultural education approach in textbook texts is consistent with acquisitions, but that the activities are lacking.

Active vibration control of a blade element with uncertainty modeling in PZT actuator force

Sivrioglu, Selim; Bolat, Fevzi Cakmak; Erturk, Ercan

The aim of this research is to attenuate the vibrations of a blade structure with an attached piezoelectric actuator using robust multi-objective control. The force obtained from a piezoelectric patch loading has uncertainties due to the complicated shape (airfoil) of the blade element. A parameter-dependent model of the force equation is developed to understand the possible variation range of the actuation force. The modal analysis of the blade is performed to find vibration mode frequencies, and an aerodynamic load is generated experimentally to create steady-state vibration on the blade. A state-space model is obtained by considering certain vibration modes and the parameter-dependent part of the force in the input vector is taken outside of the plant model. The robust stability filter is modified with parameter dependency to have a cluster of the filter. Two different multi-objective controllers are designed with different design objectives. The designed controllers are implemented in experiments and performances of the controllers are compared using frequency and time domain responses. It is shown that the flexible blade vibrations are successfully suppressed with the proposed mixed norm robust controllers under the effect of steady-state aerodynamic disturbance with different air speeds. It is observed in experimental results that the performances of the H∞/H2 controller are better than the H2/H∞ controller.

Existence of multiple solutions of Schrödinger-Kirchhoff-type equations involving the p(.) -Laplacian in ℝN

Ayazoglu, Rabil; Akbulut, Sezgin; Akkoyunlu, Ebubekir

In this paper, we prove the existence of multiple solutions for the nonhomogeneous Schrödinger-Kirchhoff-type problem involving the p(.)-Laplacian (Formula presented.) where b≥0 is a constant, N ≥2, Δp(.)u:=div(|∇u|p(.)−2∇u) is the p(.)-Laplacian operator, (Formula presented.) is Lipschitz continuous, (Formula presented.) is a coercive type potential, (Formula presented.) and (Formula presented.) functions verifying suitable conditions. We propose different assumptions on the nonlinear term (Formula presented.) to yield bounded Palais-Smale sequences and then prove that the special sequences we found converge to critical points, respectively. The solutions are obtained by the Mountain Pass Theorem, Ekeland variational principle, and Krasnoselskii genus theory.

Pb (II) biosorption from the aqueous solutions by raw and modified tea factory waste (TFW)

    Nuhoğlu, Y.; Ekmekyapar Kul, Z.; Kul, S.; Nuhoğlu, Ç.; Ekmekyapar Torun, F.

The study is aimed to determine Pb (II) biosorption from synthetic wastewaters using natural and modified tea factory waste as biosorbent material. This process is carried out with this waste matter, which is found in large quantities, so cheap and has high metal adsorption ability. The sorption kinetic studies were determined as variations of initial pH value, particle size, initial Pb (II) and tea factory waste concentrations, stirring speed and temperature. Biosorption reached the equilibrium in 15 min. Zeta potential values of the biosorbent were defined in the range of pH 2 to 4. The adsorption isotherm models give the experimental values, and the thermodynamic characteristics of the patterns generally give insight about the sorption mechanism depending on the surface characteristics and affinity of the biosorbent. The biosorption mechanism was searched considering to Freundlich, Langmuir, BET and Temkin isotherm model, and the experimental adsorption values were conformed to Freundlich adsorption model. The optimum experimental conditions were investigated as pH 3.5, particle size of 0.125–0.25 mm, stirring speed of 200 rpm, biosorbent concentration of 7 g L−1 and 25 °C. In these conditions, the highest efficiency of biosorption was obtained as 94.07% and after the activating process the efficiency was reached to 97.73% at 70 mg L−1. The maximum Pb (II) removal capacity of raw tea factory waste was found to be 22.111 mg g−1 at 200 mg L−1. The biosorption kinetics has complied with the pseudo-second-order model. Thermodynamic studies stated that this biosorption phenomenon was an endothermic process.

Optimization of asymmetric reduction conditions of 1-(benzo [d] [1,3] dioxol-5-yl) ethanone by Lactobacillus fermentum P1 using D-optimal experimental design-based model

Özdemir, Akın; Dertli, Enes; Şahin, Engin

The biocatalytic asymmetric reduction of prochiral ketones is a significant transformation in organic chemistry as chiral carbinols are biologically active molecules and may be used as precursors of many drugs. In this study, the bioreduction of 1-(benzo [d] [1,3] dioxol-5-yl) ethanone for the production of enantiomerically pure (S)-1-(1,3-benzodioxal-5-yl) ethanol was investigated using freeze-dried whole-cell of Lactobacillus fermentum P1 and the reduction conditions was optimized with a D-optimal experimental design-based optimization methodology. This is the first study using this optimization methodology in a biocatalytic asymmetric reduction. Using D-optimal experimental design-based optimization, optimum reaction conditions were predicted as pH 6.20, temperature 30 °C, incubation time 30 h, and agitation speed 193 rpm. For these operating conditions, it was estimated that the product could be obtained with 94% enantiomeric excess (ee) and 95% conversion rate (cr). Besides, the actual ee and cr were found to be 99% tested under optimized reaction conditions. These findings demonstrated that L. fermentum P1 as an effective biocatalyst to obtain (S)-1-(1,3-benzodioxal-5-yl) ethanol and with the D-optimal experimental design-based optimization, this product could be obtained with the 99% ee and 99% cr. Finally, the proposed mathematical optimization technique showed the applicability of the obtained results for asymmetric reduction reactions.

Green synthesis of chiral aromatic alcohols with Lactobacillus kefiri P2 as a novel biocatalyst

Baydaş, Yasemin; Dertli, Enes; Şahin, Engin

Biocatalytic reduction is a very important field of research in synthetic organic chemistry. Herein, three different Lactic Acid Bacteria (LAB) strains were evaluated for their bioreduction potential using acetophenone as a model substrate. Among these strains, Lactobacillus kefiri P2 strain was determined as the best asymmetric reduction biocatalyst. Reaction optimization parameters such as reaction time, temperature, agitation speed and pH were systematically optimized using Lactobacillus kefiri P2 strain and model substrate acetophenone. Under these optimized reaction conditions, secondary chiral alcohols were obtained by bioreduction of various prochiral ketones with results up to 99% enantiomeric excess. In addition, the steric and electronic effects of substituents on enantioselectivity and conversion were evaluated. It has been shown that Lactobacillus kefiri P2 biocatalyst was an effective catalyst for asymmetric reduction. This method provides an environmentally friendly method for the synthesis of optically pure alcohols and an alternative approach to chemical catalysts.

Evaluation of Social Security Cooperation between Turkey and Central Asian Turkish Republics; [Orta Asya Türk Cumhuriyetleri ile Türkiye Arasında Sosyal Güvence İş Birliklerinin Değerlendirilmesi]

    Bulut, Mehmet

One of the most important problems of international migration for immigrants is the issue of social security. As a matter of fact, the people who went to work abroad from their countries either remained completely out of the system or became a part of both countries with the effect of strict rules in social security legislation. In order to solve this problem, countries are introducing bilateral social security agreements as well as domestic legal systems. The human mobility occurred between the Central Asian Turkish Republics and Turkey have increased cumulatively at over the years. As a result of this, the social security cooperation to be realized is not yet sufficient and necessary steps should be taken. Currently, there are bilateral social security agreements that have been ratified with Azerbaijan and Kyrgyzstan. However, there are problems at the point of application.

EFFECTS OF MOLAR RATIO, FREQUENCY, WATER CONTENT AND TEMPERATURE ON DIELECTRIC PROPERTIES OF DEEP EUTECTIC SOLVENTS

    Kutlu, N.; Yilmaz, M.S.; Erdem, G.M.; Sakiyan, O.; Isci, A.

The aim of the study is to determine the effect of process parameters on dielectric propertiesof deep eutectic solvents (DESs), which have beenwidely used in recent years. In this study, DESs wereprepared using choline-chloride as the hydrogenbondacceptor and glycerol, formic acid and aceticacid as the hydrogen-bond donor. The effect of differentprocess parameters such as molar ratio (1:2, 1:3and 1:4), water content (15%, 30% and 45%), temperature(25, 50 and 75 °C) and frequency on dielectricproperties of the DESs were examined. The highestdielectric constant values were found to be38.13±0.54, 44.96±0.07 and 46.74±0.02 for glycerol, formic acid and acetic acid, respectively for the processconditions of 25 °C temperature, 45% water contentand 1:4 molar ratio (at 2450 MHz). Moreover, forall DESs, it was found that a decrease in water contentresulted in a decrease in both dielectric constant andloss factor values. This can be explained by the absenceof free water molecules which are responsiblefrom dipole rotation mechanism. In light of the results,if DES will be used in microwave processes, formicor acetic acid containing DESs might give moresuccessful results compared to the ones with glycerol
