In vitro inhibitory effects of palonosetron hydrochloride, bevacizumab and cyclophosphamide on purified paraoxonase-I (hPON1) from human serum

Türkeş, Cüneyt; Söyüt, Hakan; Beydemir, Şükrü

In this study, we investigated the effects of the drugs, palonosetron hydrochloride, bevacizumab and cyclophosphamide, on human serum paraoxonase-I (hPON1) enzyme activity in in vitro conditions. The enzyme was purified ~231-fold with 34.2% yield by using ammonium sulphate precipitation, DEAE-Sephadex A-50 ion-exchange chromatography and Sephadex G-200 gel-filtration chromatography from human serum. hPON1 exhibited a single protein band on the SDS polyacrylamide gel electrophoresis. The inhibition studies were performed on paraoxonase activity of palonosetron hydrochloride, bevacizumab and cyclophosphamide. Ki constants were found as 0.033 ± 0.001, 0.054 ± 0.003 mM and 3.419 ± 0.518 mM, respectively. Compared to the inhibition rates of the drugs, palonosetron hydrochloride has the maximum inhibition rate. However, inhibition mechanisms of the drugs were determined as noncompetitive by Lineweaver-Burk curves.

Study of phylogenetic relationship of Turkish species of Matthiola (Brassicaceae) based on ISSR amplification

Doğan, Bekir; Çelik, Mustafa; Ünal, Murat; Sefali, Abdurrahman; Martin, Esra; Kaya, Ayla

Matthiola W.T.Aiton is a taxonomically complex genus in which there are many problems, mostly with Matthiola longipetala and M. odoratissima. Matthiola species native to Turkey were collected from various locations in Anatolia, and their DNA was isolated. Revision studies performed on the basis of molecular data obtained from studies conducted in recent years have made the phylogenetic relationships and systematic positions of the taxa more apparent and reliable. Consequently, the remaining taxonomic problems among the species have been resolved through the use of DNA-based molecular analysis methods, which, unlike phenotype studies, are not affected by environmental factors. The inter-simple sequence repeat (ISSR) fingerprinting method was used in the study because its properties were considered to be more reliable and consistent than those of the randomly amplified polymorphic DNA method. DNA fragments were amplified through the use of ISSR primers. The phylogenetic relationships among the taxa were represe ted on a dendrogram constructed through means of NTSYSpc 2.02 software. The infrageneric and intergeneric phylogenetic relationships between Matthiola and other related genera were also characterized. It was determined that the taxa Matthiola odoratissima and M. ovatifolia are separate but closely related. Moreover, it was observed that the Matthiola longipetala complex forms a separate group within the genera. Clearly, the genera Matthiola, Sterigmostemum, Strigosella, Malcolmia, and Chorispora are phylogenetically differentiated on the dendrogram.

Effect of cold-press carrot seed oil on the performance, carcass characteristics, and shelf life of broiler chickens

    Ürüşan, H.; Erhan, M.K.; Bölükbaşı, S.C.

Present study aims to assess the effects of cold-press carrot seed oil (CSO) on performance, gut microflora, some serum biochemical parameters, meat color and meat shelf life of broiler chickens by experimenting with young broiler chicks. Accordingly, hundred fifty day-old Ross PM 308 broiler chicks were randomly given three different dietary treatments supplemented with 0 (control), 100mL/kg or 200 mL/kg (CSO). Five replicates of 10 birds each. It was observed that the supplementation of CSO led to an increase in weight gain, hot carcass and carcass yield on day 42. Addition of CSO supplement in the basal diet had no effect on serumbiochemical parameters. The number of lactic acid bacteria was higher in the group of chicks fed on the diet supplemented with 200 ml/kg CSO than in the other groups. Compared to the control group, dietary CSO supplement reduced the MDA values in leg muscles. L* (lightness) and b* (yellowness) values of the breast muscles increased in the chicks fed with 200 ml/kg CSO. Dietary 100 ml/kg CSO supplementation decreased a* (redness) values of the breast and leg muscles. In conclusion, supplementation of CSO had resulted in positive changes on weight gain, carcas yield, lactic acid bacteria count and breast tissue shelf life. As a result, this study suggests that cold-press carrot seed oil can be used as a dietary source of natural antioxidant for broilers.

Uzundumlu, Ahmet Semih; Oksuz, Muhammet Emre; Kurtoglu, Seval

Fig is a fruit that grows in warm and humid climates; therefore, it is widely cultivated in the countries of Mediterranean climate zone. It has an important role in human nutrition with the minerals and Vitamins it contains. While fresh figs are offered for direct consumption, dried figs are used as the main or auxiliary ingredient in many desserts. Located in the Mediterranean climate zone, Turkey is an important fig producer and exporter with a share of 23% in world fig production. Although fig production is realized in 60 out of 81 provinces in Turkey, it is mostly produced in Aydin, Izmir, Bursa, Mersin, and Hatay provinces, which provide some 86 percent of the total production. The aim of this study is to predict the fig production trend of Turkey for the next 9 years from 2017 to 2025 using the fig production data from the period between 1991 and 2016 in order to enlighten the policy and decision makers regarding fig, an important export product of Turkey. The ARIMA model has been used to make the estimations in the study. According to the findings, it has been forecasted that Turkey’s fig production will decrease in the next ten years from 2017 to 2025 and the total share of five leading provinces will increase by 1 percent in fig production. It has also been estimated that fig production will increase in leading fresh fig producer provinces, whereas it will decrease in leading dried fig producer provinces in Turkey. It has been concluded that there is a need to create fresh fig demand in other countries for new exporting possibilities.

Energy saving potential of heat insulation solar glass: Key results from laboratory and in-situ testing

Cuce, Erdem; Cuce, Pinar Mert; Young, Chin-Huai

HISG (heat insulation solar glass) is a recently developed multi-functional glazing technology to mitigate energy consumption of buildings. HISG can generate electricity similar to conventional PV (photovoltaic) glazing products when exposed to sunlight, however it differs from them by having some extraordinary characteristic features such as thermal insulation, which is competitive with Argon filled triple glazed windows, acoustic comfort, remarkable energy saving potential and self-cleaning ability owing to TiO2 nano coating. Within the scope of this research, latest results from laboratory and in-situ testing of HISG are presented in terms of its key role in mitigating heating and cooling demand of buildings as well as clean energy generation. Lighting and thermal comfort related parameters such as shading coefficient, UV, IR and visible light intensity are also investigated through the tests conducted in real operating conditions. It is achieved from the results that instant electricity generation of HISG is 16% higher than that of standard PV glazing owing to its nano layer reflective film. Shading coefficient of HISG is only 0.136, which provides almost 80% reduction in solar heat gain compared to ordinary glazing. Indoor air temperature measured from HISG test house in summer time is very close to the ambient temperature, whereas it is found to be 14.7 °C higher in ordinary glass test house due to greenhouse effect. Annual heating and cooling demand tests indicate that HISG provides 38 and 48% energy saving in heating and cooling season, respectively.

Optimum Design of Braced Steel Space Frames including Soil-Structure Interaction via Teaching-Learning-Based Optimization and Harmony Search Algorithms

Daloglu, Ayse T.; Artar, Musa; Ozgan, Korhan; Karakas, Ali I.

Optimum design of braced steel space frames including soil-structure interaction is studied by using harmony search (HS) and teaching-learning-based optimization (TLBO) algorithms. A three-parameter elastic foundation model is used to incorporate the soil-structure interaction effect. A 10-storey braced steel space frame example taken from literature is investigated according to four different bracing types for the cases with/without soil-structure interaction. X, V, Z, and eccentric V-shaped bracing types are considered in the study. Optimum solutions of examples are carried out by a computer program coded in MATLAB interacting with SAP2000-OAPI for two-way data exchange. The stress constraints according to AISC-ASD (American Institute of Steel Construction-Allowable Stress Design), maximum lateral displacement constraints, interstorey drift constraints, and beam-to-column connection constraints are taken into consideration in the optimum design process. The parameters of the foundation model are calculated depending on soil surface displacements by using an iterative approach. The results obtained in the study show that bracing types and soil-structure interaction play very important roles in the optimum design of steel space frames. Finally, the techniques used in the optimum design seem to be quite suitable for practical applications.

Effect of doping concentration on the structural and optical properties of nanostructured Cu-doped Mn3O4 films obtained by SILAR technique

Bayram, Ozkan; Guney, Harun; Ertargin, Mehmet Emrah; Igman, Erdal; Simsek, Onder

In this study, the effect of Cu doping on the optical, structural and wettability properties of the nanostructured Cu-doped Mn3O4 (CMO) thin films obtained by using successive ionic layer adsorption and reaction technique was systematically investigated. The optical, morphological, structural and wettability properties of the nanostructured CMO films were determined using scanning electron microscopy (SEM), X-ray diffraction (XRD), UV–Vis spectroscopy, and water contact angle (WCA) measurements. There were significant differences in the crystal structure of the nanostructured CMO thin films. The Cu doping disrupted this structure of pure (undoped) Mn3O4 films and caused the formation of the porous structure. Optical properties such as extinction coefficient, refractive index, and dielectric constants and optical band gap, were determined for nanostructured CMO films. Compared with pure films, the band gap of Cu-doped films decreased from 2.06 to 1.71 eV with increasing Cu concentration. The dielectric constants of the films showed that the nanostructured CMO films have a transparent structure. At 300 nm wavelength, the dielectric constants of pure Mn3O4 films were 2.22 while the calculated dielectric constants at increasing doping concentrations were 3.55, 2.41 and 2.25. It was also found from the WCA measurements that the thin films were hydrophilic in character and the WCA values for the nanostructured CMO thin films were measured as 24, 29 and 40 of degree.

Yudar, H. Hakan; Pat, Suat; Özen, Soner; Şenay, Volkan; Korkmaz, Şadan; Pat, Zerrin

In this paper, the effect of RF power on LiCoO2 thin films was investigated using X-ray diffractometer (XRD), atomic force microscopy, UV–Vis spectrophotometer, and potentiostat. The microstructural, surface, optical and electrochemical impedance measurements were performed to LiCoO2 thin films, are used to for the fully solid-state battery cathode material. According to obtained results, the relative intensities of the Li (002) crystal phase in XRD patterns of deposited LiCoO2 thin films were increased by increasing applied RF power, for the first time. The intensity of the LiCoO2 (104) plane is nearly invariant. The relative intensities of the LiCoO2 (113) plane were decreased by increasing RF power. The peak locations of the Li (002) and LiCoO2 (104) were not changed. It was found that Li (002) relative intensities affect the all investigated parameters for the LiCoO2 thin films. Especially, transmittance value increased about 20%. The band gap of the deposited film was changed 100–300 meV drastically. Deposited samples are shown high transparency in the visible region. Randles circuit was used for the equivalent circuit model. Nyquist plots, fitting parameters values and value of the circuit elements were estimated by ZSim software.

Use of meta-heuristic algorithms in analysis of structures is much more recent thantheir use in design of structures. Effectively, while design optimization makes use ofgenetic algorithms, simulated annealing, swarm optimization, etc. since about half acentury with an increasing interest, these techniques are being used in structural analysissince a decade only.Total Potential Optimization using Meta-heuristic Algorithms (TPO/MA), thoughbeing an emerging method at the moment, has proved to be very efficient in solvingnonlinear problems, involving both material and geometric nonlinearities. Thecapabilities of the method in treating under-constrained problems and problems with nonuniquesolutions are much more advanced than the classical methods including the wellknownFinite Element Method.

Performance investigation of a jet loop membrane bioreactor for the treatment of an actual olive mill wastewater

Değermenci, Nejdet; Cengiz, İbrahim; Yildiz, Ergun; Nuhoglu, Alper

In this study, following the pre-treatment of olive mill wastewater (OMW), its treatment in a jet loop membrane bioreactor (JLMBR) was investigated. Among the pre-treatment options, the configuration composed of physical settling, cartridge filter and ceramic membrane showed the best performance in terms of investigated parameters. For the JLMBR that was fed by pretreated OMW, up to 93 and 87% removal efficiencies were achieved for the chemical oxygen demand (COD) and total phenol, respectively, at volumetric organic load (VOL) of 17.8 kg COD/m3 day. The calculated specific oxygen uptake rate (SOUR) values were in the range 7.7–34.7 g O2/kg MLVSS h. When even hydraulic retention times (HRT) values decreased by a factor of 1:24, system performance in terms of COD and total phenol removal remained almost stable. Decreasing the sludge retention time (SRT) to three days made considerable perturbations for the system performance, increasing the effluent COD and total phenol values in 900 and 80 mg/L, respectively. The JLMBR showed a high overall performance for the treatment of an actual OMW under the evaluated operational conditions.

Quality evaluation and pollen profile of honey samples from different locations

    Bayram, Nesrin Ecem

In this study, it is aimed to determine some physicochemical properties and pollen types of honey samples collected from different regions. Pollen profile, moisture content, HMF, proline, frutose/glucose and electrical conductivity were the parameters analysed in each honey sample. According to the results obtained, all honey samples obtained from different geographic origins were found to be consistent, in terms of the parameters analyzed, with the values given in the Turkish Food Codex Honey Communiqué (2012). It was found that only one honey sample did not comply with the values given in the codex in terms of frutose/glucose ratio. As a result of the melissopalynological experiment, three honey samples were found to be unifloral (two Castanea sativa and one Astragalus spp.) and the others were found to be polyfloral honey. The results obtained indicate that physicochemical properties of the honey samples produced at different points in Turkey differ greatly in accordance with the diversity of the flora of the region.

Taguchi Approach for Optimization of Parameters that Effect Grain Size of Cast A357 Alloy

    Çolak, M.; Dlşplnar, D.

Grain refining and modification are common foundry practice for improving properties of cast Al-Si alloys. In general, these types of treatments provide better fluidity, decreased porosity, higher yield strength and ductility. However, in practice, there are still some discrepancies on the reproducibility of the results from grain refining and effect of the refiner's additions. Several factors include the fading effect of grain refinement and modifiers, inhomogeneous dendritic structure and non-uniform eutectic modification. In this study, standard ALCAN test was used by considering Taguchi's experimental design techniques to evaluate grain refinement and modification efficiency. The effects of five casting parameters on the grain size have been investigated for A357 casting alloy. The results showed that the addition of the grain refiner was the most effective factor on the grain size. It was found that holding time, casting temperature, alloy type and modification with Sr were less effective over grain refinement.

A causal and effect model for knowledge flow analysis among firms

    Dereli, Türkay; Altuntas, Serkan

Examination of the relations among firms with respect to knowledge flows is essential for sustainable development and creative economy in global competition as well as for governments, investors and decision makers. Knowing the causal relations among firms provides answers to questions such as: (1) which firms have strong associations? (2) which firms are in the cause group and (3) which firms are in the effect group? The knowledge flow among firms shows the relations among firms directly. Patent citation analysis is extensively conducted to measure the knowledge flow in the literature. The decision making trial and evaluation laboratory (DEMATEL) method is also extensively used to construct causal relations among variables. In this study, the DEMATEL method based patent citation analysis is utilized to construct causal relations among firms. An illustrative example is presented to find relations among firms which operate business method technologies. The results of the example show that the DEMATEL based patent citation analysis can be performed to construct causal relations among firms.

Determination of the effect of electric fence system on productivity and behaviour of honeybees housed in different beehive types (Apis mellifera L.)

    Erdoğan, Yaşar

This study was conducted in 2018 in order to determine the effects of an electric fence system on honeybee colonies housed in hives made from different materials. The experiment was carried out with honeybee colonies of 36 Caucasian races (Apis mellifera Caucasica) in full-size Langstroth hive. In terms of mean values of the aggressive behaviour of honeybee colonies were found to be the highest (13.57 number/colony) application group where the conductor wires of electric fence system are within a 1 m distance from hives and the lowest (2.39 number/colony) were obtained from control application group. The highest mean values adult bee development (23.76 ± 12.34 number/colony), development of brood area (5421.25 ± 1432.75 cm 2 /colony), nectar flow period weight gain (46.42 ± 3.61 kg/colony), flight activity (107.38 ± 11.74 number/colony) and honey yield (21.55 ± 1.54 kg/colony) were determined in control groups. When the average values of honeybees robbing behaviour were examined, the highest value (3.33 ± 0.32 number/colony) was determined in the control groups. This experiment shows that the electromagnetic and electric fields produced by the electric fence system have negative effects on honeybees. In order to minimise the negative effects of the electric fence system on the bees, hives should be covered with a metal screen or should be placed long distance (minimum 2 m) from the electric fences.Highlights The electric fence system is the most effective method of keeping honeybee colonies. At the same time, the electric fence system disturbs the honeybees. All negativity can be completely eliminated by simple measures.
