Purpose: The aim of this study is to evaluate the impact of gossypin on the expression level of heat shock proteins (HSPs) genes in different cancer cells. Materials and Methods: Cells were grown under standard culture conditions. Cancer cells were treated with different concentrations (5-100 mu g/ml) of gossypin and cisplatin (50 mu M) as positive control. Cell viability and effective dose range (5-100 mu g/ml) of gossypin were determined by MTT at 24, 48 and 72 hours. After RNA isolation and cDNA synthesis, HSP60 and HSP70 gene expression levels were analyzed using RT-PCR. For gene expression analysis, the 2-(Delta Delta ct) method was used. Results: According to the Mil results, 25-50-100 mu g/ml of gossypin doses were found effective on HSP60 and HSP70 gene expression levels in the cancer cell lines. Gossypin affected with dose-dependently the expression of HSP60 and HSP70 in the three cell lines. In the three cell lines, 50 mu g/ml and 100 mu g/ml of gossypin doses significantly reduced the expression of HSP60 and HSP70 compared to control group. Conclusion: Our results strongly supported the anticarcinogenic effect of gossypin at various doses in different cell lines. However, we believe that further in vivo research and human studies are needed. Our findings suggest that gossypin could be suitable candidate agent for further investigation to develop new strategies for the prevention and/or treatment of different cancer types.

Ustabulut, Mete Yusuf; Boylu, Emrah; Basar, Umut
Turkiye's migrant integration policy, which is one of the most migration-receiving countries in the Middle East owing to regional conflicts during the 2000s, is assessed alongside that of other receiving nations in the Migrant Integration Policy Index (MIPEX). These index ranks nations in eight key categories: labor market mobility, family reunion, education, health, political participation, permanent residence, access to nationality, and anti-discrimination. This study examines Turkiye's change in migration policy ratings by comparing the index 2015 and 2020 with six European countries (Germany, Belgium, France, Norway, Finland, Sweden). It is found that there was a significant disparity in index scores between Turkiye and these European countries in the 2015 MIPEX report; however, this difference has narrowed in the 2020 report. Turkiye's migrant integration policy rankings have improved substantially (+17), according to the 2020 report. As a result, Turkiye's policy indicator has increased from slightly unfavourable to halfway favourable . Although this score and increase are not at the desired level, they may indicate comprehensive improvements in Turkiye's integration policies in the following report.

- MakaleAçık Erişim2023Prioritising and selecting the railway routes planned in the Eastern Black Sea Region with the AHP and TOPSIS methods
Boz, Ediz; Türk, Ersin; Togan, Vedat
The construction of the railway, which is needed for the development of the Eastern Black Sea Region, is an issue that has been discussed and unresolved for about 110 years. The geomorphological structure of the region and the developments in the highway played a role in the delay of the railway construction. However, the developing technology in the railway, Turkey's being a part of the Iron Silk Road Project, and the agreements made with international connections have brought the railway construction to the region again. In the Environmental Plan (EP) approved and in force by the Ministry of Environment, Urbanization and Climate Change, four railway routes are planned to integrate the Eastern Black Sea region with the national/international railway network. These routes are; Samsun-Sarp, Trabzon-Erzincan, Of -Erzurum and Tirebolu-Bayburt. However, some of these routes are alternatives to each other and are not feasible and rational since the implementation of all planned routes requires great costs. The aim of this study is to develop a method for prioritizing and selecting the railway routes planned in the EP. Within the scope of this purpose, by using Analytical Hierarchy Process (AHP) and TOPSIS techniques, which are multi-criteria decision making methods, the routes were listed according to 4 criteria and 10 sub-criteria determined with the help of literature and expert team. As a result of the study; In both techniques, the order was the same and it was determined that the Samsun-Sarp railway was the most priority railway route. Trabzon-Erzincan came second, Of-Erzurum came third and Tirebolu-Bayburt came fourth.

The goal of this research was to investigate the effects of the ozone treatment (OT) to raw cream on the physicochemical, microbiological, thermal, and oxidative properties of butter. For this purpose, butter samples were produced from raw cream, which was ozone-treated for 5 (OT-5), 15 (OT-15), 30 (OT-30), and 60 (OT-60) min. The control sample was also produced from non-ozonated raw cream. OT increased firmness, consistency, and fat particle size values of cream. The firmness of butter was between 97 and 380 N. Average fat particle size of samples (D[3.2]) ranged from 22.1 to 30.2 mu m, in which OT decreased it. A monomodal fat particle size distribution was determined in all-butter samples. OT decreased b value (yellowness) of butter, and the lowest value was determined in OT-60. The induction time of butter changed from 1.04 (OT-60) - 32.9 h (control). Two melting peaks were determined in butter samples at 14.2-14.6 and 32.3-33 degrees C. Ozonation for 60 min caused 2.01 log reduction in Staphylococci. Above 15 min, OT completely inactivated Salmonella and yeast-mould. Coliform was not detected in OT-30 and OT-60. The results showed that OT resulted in higher microbiological quality in butter from raw cream.

İncele - MakaleYayıncıda açık erişim2025Answer-based and reference-based BERT models for automatic scoring of Turkish short answers: The decisive role of task complexity
Kara, Abdulkadir; Kara, Zeynep Avinc; Yildirim, Serkan
In measurement and evaluation processes, natural language responses are often avoided due to time, workload, and reliability concerns. However, the increasing popularity of automatic short-answer grading studies for natural language responses means such answers can now be measured more quickly and reliably. This study aims to build models for predicting automatic short answer scores using the pre-trained BERT deep learning language model and to reveal their effectiveness. For this purpose, two different score prediction models were created using an answerbased approach that aligns student answers with expert judgements and a referencebased approach that matches student answers with reference answers. The dataset includes answers from 246 Physics department students responding to 4 physicsrelated questions. The performance of these models was evaluated on four physics questions representing varying levels of cognitive complexity, using Cohen's Kappa for statistical comparison of agreement with expert scores. Our findings reveal a clear interaction between model architecture and task complexity. The answer-based model was unequivocally superior for the most complex, multi-class task, effectively capturing diverse, nuanced responses. Conversely, the reference-based model demonstrated a statistically significant advantage for a well-defined, mediumcomplexity binary task. This study concludes that the optimal model for ASAG in Turkish is contingent on the cognitive demands of the assessment task, suggesting that a onesize-fits-all solution may not be the most effective approach. This provides a critical framework for practitioners, demonstrating not only that effective models are feasible for complex languages, but that their selection must be guided by task complexity.

İncele - MakaleAçık Erişim2021Expression level of the angiotensin 2 (ACE2) gene in patients with COVID-19
Dirican, Ebubekir; Aydin, Ismail Erkan; Savrun, Seyda
Purpose: Coronavirus disease-19 (COVID-19) is caused by the Severe Acute Respiratory Syndrome Coronavirus 2 (SARSCoV2). The angiotensin converting enzyme 2 (ACE2) can play a vital role in regulating the pathological changes of various diseases, including COVID-19. The aim of this study was to analyze the expression of the ACE2 gene in Turkish COVID-19 patients. Materials and Methods: Eighty COVID-19 PCR test positive patients and 80 PCR test negative healthy volunteers were included in the study. RNA isolation was carried out from all samples. The RT-PCR device was used to analyze the expression of the ACE2 gene. Results: A significant difference was found in the age, Lactate dehydrogenase (LDH), ferritin and C-reactive protein (CRP) levels of the COVID-19 patients compared to the healthy volunteers. Based on the gene expression results, it was found that ACE2 gene expression was significantly higher in COVID-19 patients compared to healthy volunteers. In patients with COVID-19, a significant difference was found between the expression of the ACE2 gene and the levels of Blood urea nitrogen (BUN), Hemoglobin (HGB), Hematocrit (HCT) and CRP. Conclusion: ACE2 gene expression was high in the COVID-19 patients and showed differences based on clinical data. Thus, the expression of ACE2 can have paradoxical effects that support the pathogenicity of SARSCoV2 but, conversely, limit viral infection. The availability of ACE2 receptors may increase the susceptibility and / or disease course of COVID-19.
