Background. Anaerobic and aerobic power are critical factors influencing peak football performance. This study aimed to explore the differential effects of various high-intensity interval training (HIIT) protocols on these performance metrics in senior female football players. Objectives. The primary objective was to investigate the impact of linear (L) HIIT and change-of-direction (COD) HIIT protocols on key velocity at anaerobic threshold (V-AT), distance traveled in the anaerobic zone (d(AT)), and VO(2)max. Methods. Sixty senior female football players were divided into LHIIT (n=30) and COD (n=30). Both groups underwent initial testing before participating in their respective training programs for four weeks. Performance metrics were measured at the beginning and end of the training period. Results. Both groups significantly improved all measured variables (p<0.001). However, the COD group demonstrated relatively more significant enhancements in Vmax (COD=0.93; LHIIT=0.8), VAT (COD=0.94; LHIIT=0.87), and VO(2)max (COD=0.92; LHIIT=0.74). The COD group also reduced the initial difference in VO(2)max to a non-significant level. Conclusion. COD HIIT protocols appear to be slightly more effective in enhancing anaerobic power in female football players than linear HIIT protocols. Nevertheless, both HIIT programs significantly improved performance metrics, indicating that incorporating such training regimens can benefit female football training programs.

Cesaro, Angela; Broset, Esther; Salimando, Gregory J.; Oztekin, Sebahat; Mai, David; Bennett, Mariko L.; de la Fuente-nunez, Cesar
Extensive research supports a pivotal role for the gut-brain axis(i.e., the communication pathway between the bacteria in the gut and the brain) in brain health and disease. The interplay between the gut microbiota and the mammalian nervous system appears to shape both adaptive and dysfunctional neurological processes, as it directly affects neurons and neural signaling, the immune response, and metabolism via its effects on hormones, neuropeptides, and neurotransmitters. This review summarizes key findings from the recent literature focused on the interactions between neurons and gut bacteria. By focusing on synthetic biology tools currently employed to study the gut-brain axis, we can enhance our understanding of neuroactive substances released in the gut and potentially identify select molecular candidates that may facilitate the development of personalized therapies.

- MakaleAçık Erişim2025Impact of Linear vs. Change-of-Direction HIIT on Aerobic Power
Morina, Besnik; Visic, Grgur; Antonic, Davorin; Babic, Matej; Sporis, Goran; Vlahovic, Tomislav; Franic, Miljenko; Franjko, Ivica; Tomac, Zvonimir; Akman, Onur
Background. Coaches predominantly utilize linear training protocols to enhance the aerobic power of football players. However, there is limited data on the effects of change of direction (COD) protocols on aerobic power in football. Objectives. This study aimed to determine which high-intensity interval training (HIIT) program- linear (LHIIT) or change of direction (COD)-has a more significant impact on increasing aerobic power in senior female football players. Methods. Sixty senior female football players were divided into two experimental groups: LHIIT (n=30) and COD (n=30). Both groups underwent a four-week HIIT protocol, with LHIIT involving linear and COD incorporating change-of-direction exercises. Aerobic power was measured using the 30-15 intermittent fitness test (IFT), maximal aerobic velocity (MAV), and VO2max at the beginning and end of the study to assess the differences in aerobic adaptations between the two programs. Results. The results indicated that the COD group eliminated the significant difference in VO2max that initially existed between the two groups, suggesting that COD protocols may be more effective in increasing aerobic power than linear ones. Additionally, both groups significantly improved the 30-15 IFT, MAV, and VO2max. Conclusion. Both linear and change-of-direction HIIT protocols significantly enhance aerobic performance in senior female football players. However, COD protocols may have a more significant impact on VO2max improvement. Future research should explore the potential effects of COD protocols on anaerobic power and seek to identify even more effective HIIT strategies for enhancing VO2max.
- MakaleYalnız künye2026Scopus SJRQ1Morin counteracts KBrO3-induced hepatotoxicity by reprogramming the HOTAIR/MEG3-miR-125b-5p/miR-214-5p axis and suppressing STAT1-associated RIPK3/MLKL necroptotic signaling
Özer, Muhammed Rasit; Özdemir, Selcuk; Küçükler, Sefa; Özkaraca, Mustafa; Kizil, Hamit Emre; Akaras, Nurhan; Mutlu, Huseyin; Kandemir, Fatih Mehmet
Potassium bromate (KBrO3; PB), an oxidative contaminant in ozonated water and bakery products, induces multi-mechanistic hepatotoxicity. Its epigenetic, necroptotic, and metabolic injury dimensions remain poorly characterized. Morin (MOR), a dietary flavonoid with established antioxidant and anti-inflammatory properties, has not previously been evaluated against PB-induced subacute hepatotoxicity. To characterize MOR's hepatoprotective mechanisms against PB-induced liver injury, focusing on the HOTAIR/MEG3-miRNA epigenetic axis, STAT1-associated RIPK3/MLKL necroptotic signaling, and Glut2/HPGD-AKT/mTOR metabolic signaling. Male Sprague-Dawley rats (n = 28) were assigned to Control, MOR (100 mg/kg), PB (62 mg/kg), and PB + MOR groups for 14 days. Outcomes included hepatobiliary biochemistry, oxidative stress markers, inflammatory mediators (ELISA), lncRNA/miRNA/mRNA profiling (RT-qPCR), immunohistochemistry (RIPK3, MLKL), double immunofluorescence (Glut2/AKT1; HPGD/mTOR), histopathology, and PCA. PB produced marked redox imbalance (up arrow MDA; down arrow SOD, CAT, GPx), NF-kappa B/NLRP3-associated inflammatory mediator elevation, epigenetic disruption (up arrow HOTAIR, down arrow MEG3; up arrow miR-125b-5p/down arrow BCL2L2; down arrow miR-214-5p/up arrow SMAD4), upregulation of STAT1-associated necroptosis-related transcripts and proteins (up arrow RIPK1/RIPK3/MLKL), and metabolic dysfunction (down arrow Glut2, down arrow HPGD; up arrow AKT1/mTOR). MOR restored redox balance, suppressed inflammation, and reprogrammed the miRNA axis while normalizing MLKL (transcript and immunohistochemistry) and AKT1 and partially attenuating RIPK3, mTOR, and STAT1. MOR confers multi-targeted hepatoprotection by restoring redox and metabolic homeostasis, attenuating NF-kappa B/NLRP3-associated inflammation, reprogramming HOTAIR/MEG3-miRNA epigenetics, and attenuating STAT1-associated RIPK3/MLKL signaling, supporting its potential as a dietary hepatoprotective candidate. PB is associated with engagement of the hepatic STAT1/RIPK3/MLKL axis and disrupts the HOTAIR/MEG3-miRNA network-both previously undocumented for this toxicant. MOR partially restores miR-214-5p/miR-466 expression while suppressing their downstream effector SMAD4, consistent with attenuation of pro-fibrotic signaling. Future work should employ multi-dose designs, in vitro mechanistic dissection, sex-stratified analyses, and clinical pharmacokinetic profiling to advance MOR toward translational application.

Tapkin, Serkan; Tercan, Emre; Motsa, Siphesihle Mpho; Drosopoulos, Georgios; Stavroulaki, Maria; Maravelakis, Emmanuel; Stavroulakis, Georgios
This article presents an investigation of the structural behavior of a masonry arch bridge in Turkey. An analytical study has been conducted to provide the geometry of the structure, using laser scanning. A point cloud describing the geometry was obtained and properly transformed into a format that is appropriate for structural analysis software (CAE). Then, nonlinear finite-element models were developed to simulate structural responses of the bridge. The goal of the article is to highlight the influence of both continuum and discrete approaches and related constitutive laws on the responses of the bridge. Thus, continuum damage laws and a discrete model consisting of unilateral contact-friction interfaces were developed. Different load cases were tested and a comparison between the results obtained from the different approaches was considered. The failure mechanisms and the ultimate strengths were derived, and core points of the models were highlighted. The output of this work shows how the different failure models predict the behavior of the masonry arches. It also shows that the three-hinge mechanism, which has been depicted in classical studies for single-span arch masonry bridges under a horizontal settlement of supports, may also be obtained for multiarch bridges. Similarly, downward, vertical settlement of supports may result in the development of two hinges, as in single-span arches. Finally, the beneficial influence of the backfill in limiting the failure in the arch is addressed.

- MakaleAçık Erişim2023Scopus SJRQ1Structure-function-guided design of synthetic peptides with anti-infective activity derived from wasp venom
Boaro, Andreia; Ageitos, Lucia; Torres, Marcelo Der Torossian; Blasco, Esther Broset; Oztekin, Sebahat; de la Fuente-nunez, Cesar
Antimicrobial peptides (AMPs) derived from natural toxins and venoms offer a promising alternative source of antibiotics. Here, through structure-function-guided design, we convert two natural AMPs derived from the venom of the solitary eumenine wasp Eu-menes micado into a-helical AMPs with reduced toxicity that kill Gram-negative bacteria in vitro and in a preclinical mouse model. To identify the sequence determinants conferring antimicrobial ac-tivity, an alanine scan screen and strategic single lysine substitutions are made to the amino acid sequence of these natural peptides. These efforts yield a total of 34 synthetic derivatives, including alanine substituted and lysine-substituted sequences with stabilized a-helical structures and increased net positive charge. The resulting lead synthetic peptides kill the Gram-negative pathogens Escheri-chia coli and Pseudomonas aeruginosa (PAO1 and PA14) by rapidly permeabilizing both their outer and cytoplasmic membranes, exhibit anti-infective efficacy in a mouse model by reducing bacte-rial loads by up to three orders of magnitude, and do not readily select for bacterial resistance.

Liu, Fei; Saricaoglu, Furkan Tuerker; Avena-Bustillos, Roberto J.; Bridges, David F.; Takeoka, Gary R.; Wu, Vivian C. H.; Chiou, Bor-Sen; Wood, Delilah F.; McHugh, Tara H.; Zhong, Fang
Carvacrol is a volatile monoterpenic phenol and main component of oregano essential oil that shows nonspecific antimicrobial activity against foodborne pathogenic bacteria. Fish-skin gelatin (FSG) nanofibers encapsulating carvacrol (15%, 20%, 25%, and 30%, w/w FSG) were successfully prepared via solution blow-spinning (SBS) technique using lecithin (2.475% wb) as the surfactant. FSG emulsions with lower carvacrol ratios (5% and 10%) showed higher values in particle size and surface tension as well as lower values in viscosity and modulus, which led to failure of maintaining nanofibers shape. The formed carvacrol-FSG nanofibers showed round and smooth morphologies with average fiber diameters ranging from 103.2 to 138.1 nm as the carvacrol ratio increased from 15% to 30%. Carvacrol was evenly dispersed within the interior of nanofiber matrix. All carvacrol-FSG nanofibers showed inhibitive effects against the growth of Escherichia coli, Salmonella enterica, and Listeria monocytogenes. Moreover, nanofibers with lower carvacrol ratios showed bigger inhibition zones for E. coli and L. monocytogenes (20 mm compared with 12.5 mm for lowest to highest carvacrol ratios, respectively). Nanofibers stored at 20 degrees C (51% RH) showed better retention (40% to 60%) for carvacrol during the first 4 weeks of storage, while nanofibers stored at 2 degrees C (70% RH) showed better retention (10% to 30%) at the end of storage.

Gökçe, H. S.; Tuyan, M.; Ramyar, K.; Nehdi, M. L.
The eco-efficiency and economy of geopolymer composites largely depend on their alkaline activator dosage. In this study, the effect of applying a pre-setting pressure and variation of the aggregate-to-fly ash ratio on the alkaline solution dosage and mechanical strength of fly ash-based geopolymer composites was explored. It is shown that through control of the aggregate-to-ash ratio and application of pre-setting pressure, compressive strength could be increased by 102% and 86% for Class F fly ash-based geopolymer and alkali-activated Class C fly ash-based mixture, respectively. The total alkaline solution consumption could be reduced from 718 to 188 kg/m(3) and from 769 to 262 kg/m3 for Class F fly ash-based geopolymer and alkali-activated Class C fly ash-based mixture, respectively. The proposed method reduced the alkaline solution consumption per compressive strength (6.2 kg.m(-3).MPa-1) by up to 85% compared to that of the reference manually consolidated control geopolymer. The findings demonstrate that fly ash-based geopolymers could be made more sustainable and eco-efficient through tailored production techniques. (C) 2019 American Society of Civil Engineers.

Emsen, Bugrahan; Togar, Basak; Turkez, Hasan; Aslan, Ali
The present study aims at assessing the efficacies of olivetoric acid (OA) and physodic acid (PA) isolated from Pseudevernia furfuracea (L.) Zopf (Parmeliaceae) in human lymphocytes (HLs) in vitro. 3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide and lactate dehydrogenase assays were performed to establish cytotoxicity in HLs. Besides, oxidative stress and genotoxicity were monitored by estimating the changes of total oxidative stress (TOS) and 8-hydroxy-2'-deoxyguanosine (8-OH-dG) levels, respectively, in HLs. At the same time, OA- and PA-induced total antioxidant capacity (TAC) levels in HLs were determined. Although especially low concentrations of OA (IC50 = 109.94 mg/L) and PA (IC50 = 665.49 mg/L) did not show cytotoxic effect at high levels in HLs, it was revealed that cytotoxicity was significantly (p < 0.05) associated with oxidative stress and genotoxicity via correlation analysis. While TOS level in HLs did not statistically (p > 0.05) increase in the presence of all treatments (0.5-100 mg/L) of PA, TAC level was increased by PA applications in certain concentrations (0.5-10 mg/L). Overall, the obtained data indicate that OA and especially PA as lichen compounds that do not cause oxidative stress can be a new resource of therapeutics as recognized in the present study with their high antioxidant features.

- BildiriAçık Erişim2013Scopus SJRQ2Fuzzy weighted association rule based solution approaches to facility layout problem in cellular manufacturing system
Altuntas, S.; Dereli, T.; Selim, H.
This study aims to propose new solution approaches based on fuzzy weighted association rules for facility layout problem in a cellular manufacturing system. The facility layout problem under concern has two important aspects, namely; fuzzy product route and fuzzy machine weight. Number of machine types as well as the number of machines in each type is determined in order to define a fuzzy product route. A fuzzy set is then assigned for each machine type to match them with appropriate products. Alternative routes for each product and machine weights are defined by fuzzy functions. Market demand is also used to assign weights to the machines. Three solution approaches, namely fuzzy weighted association rule mining (FWARM), Gyenesei's weighted quantitative association rules (WQAR) and normalised weighted association rule mining (NWARM) based approaches, are proposed in this study. To the authors' best knowledge, this is the first attempt which considers fuzzy weighted association rule-based data mining approaches to the facility layout problem. An illustrative example is presented to demonstrate the usefulness of the proposed approaches.

Carbonic anhydrase is an enzyme that takes responsibility in inhalation function but, until today, carbonic anhydrase is not examined if it is present in the bovine brain membrane or not. The enzyme carbonic anhydrase was purified and separately characterized according to the bonding forms in 4 steps such as outer peripheral, cytosolic, inner peripheral and integral. Affinity chromatography was used for purification of the enzyme in all four steps. The affinity column was prepared with sepharose-4B-L-tyrosine-sulphanilamide. Purified carbonic anhydrase was obtained at each step. Enzyme activity was measured by CO2 hydratase activity and esterase activity methods. Optimum pH and optimum temperature were defined for purified enzymes at each step. Morover molecular weight and purity were detected by gel filtration and SDS-PAGE electrophorose. In addition, the enzyme's Km and nmax values were found with the Lineweaver- Burk method. The obtained results are discussed in comparison with other mammalian carbonic anhydrases. Carbonic anhydrase was shown to be exist in bovine brain membrane and this enzyme was optimized.
