- MakaleAçık Erişim2022Scopus SJRQ2What Should Be the Ideal Solvent Percentage and Solvent-Propolis ratio in the Preparation of Ethanolic Propolis Extract?
Kara, Yakup; Can, Zehra; Kolaylı, Sevgi
The biological activity of propolis is mostly due to its polyphenols. Raw propolis is consumed after being extracted in various solvents, ethanol being the most commonly used. However, the ideal percentage and ratio of ethanol for use in the preparation of propolis extracts are uncertain. The aim of this study is to compare the phenolic compositions and antioxidant activities of propolis extracts extracted with different ethanol concentrations and extraction conditions. Two different extraction methods were used, normal and ultrasonic extraction. The propolis extracts were evaluated in terms of total phenolic content, total flavonoid content, ferric reducing/antioxidant power,1,1-diphenyl-2-picrylhydrazyl radical scavenging activity, and phenolic compositions using RP-HPLC–PDA. The propolis extracts with the highest activity were determined beforehand by preparing ethanol–water mixtures at different percentages, ranging from 0 to 99.9%. The most suitable propolis-to-solvent ratio was then determined. The optimum ethanolic percentage varied between 60 and 70%, the ultrasonic extraction process increased the efficiency, and the ideal solvent-propolis ratio (w/v) ranged between 1:5 and 1:10.

In this work, a reverse phase high-performance liquid chromatographic method (RP-HPLC) with photo diode array (PDA) at 4 different wavelengths (250, 280, 320 and 360) for the determination of some polyphenols in some bee products (honey, pollen, and propolis) is developed. The analyses were carried out on C18 column (250 mm x 4.6 mm, 5 μm; GL Sciences), and the gradient program was used with a mobile phase A reservoir with 70% acetonitrile and the B reservoir with 2% acetic acid. The HPLC method was founded that limit of detection was in the range 0.022-0.0908 μg/mL; the limit of quantification was in the range 0.074-0.3027 μg/mL, all calibration curves were linear R2>0.995 within the range, the recovery range was 91.43-111.37% for 10 ppm and 98.44-101.68% for 40 ppm and relative error levels 0.0330-0.0290 respectively. The developed method was applied to some bee products available on the Turkey market. The study aimed at phenolic profiles of the bee products extracts were revealed by using 25 phenolic standards. The proposed method was optimized, quickly, and simple validated by evaluating the linear range, the limits of detection and quantification, the accuracy, the precious, the repeatability, and recoveries suitable for the phenolic analysis. It is concluded that evaluated and the quantitative determination of the bee products can be made quickly and reliably with the optimized method.

Anzer honey is produced on the Anzer plateau, known for its rich flora, in the Eastern Black Sea region of Turkey. It is well known across the world, and is believed to be of high medical value. This study is the first detailed research in which the polyphenolic profiles responsible for the bioactive properties of Anzer honey was determined, melissopalynological analysis was performed, and in which total phenolic contents (TPC), total flavonoid contents (TFC), and total antioxidant activities were identified. The ferric (III) reducing antioxidant power (FRAP) assay and 1,1-diphenyl-2- picrylhydrasil (DPPH) free radical scavenging test were used to determine antioxidant activity. The total mean phenolic content of Anzer honey was 26.92 GAE/100 g, and the total flavonoid content was 2.79 mgQE/100g. Anzer honeys' mean FRAP and DPPH values were 110.11 μmoLTrolox/100g and 49,12 mg/mL, respectively. The phenolic acids and flavonoids of Anzer honey were determined using highperformance liquid chromatography (HPLC). Of the 19 standard compounds used in the analysis, pinocembrin, hesperidin, chrysin, protocatechuic acid, p-coumaric acid, catechin, caffeic acid phenyl ester (CAPE), p-OH benzoic acid, and caffeic acid as major compounds, while myricetin, luteolin, rutin, resveratrol, epicatechin, t-cinnamic acid, ferulic acid, and gallic acid were identified as minor compounds. Daidzein and syringic acid were not detected. Based on these findings, Anzer is a flower honey noteworthy for the rich variety of its polyphenols.

İncele - MakaleYalnız künye2017Scopus SJRQ4Bioactive content of Rosa canina biotypes from Turkey
Beyhan, O.; Koc, A.; Ercisli, S.; Jurikova, T.; Cakir, O.
Rose hips have been used as medicinal and aromatic plants for a long time throughout Turkey. The plant has been the focus of many recent studies due to its potential effects on treatment and prevention of several diseases. However, there are few in vitro studies concerning its composition and antioxidant capacity. The aim of the present study was to determine bioactive content including ascorbic acid, total phenolics, total flavonoids, total carotenoids and antioxidant activity of Rosa canina biotypes growing wild in Sivas province located eastern Anatolia. The results revealed that fruits of Rosa canina biotypes rich in terms of Vitamin C which ranged from 360 to 482 mg/100 g fresh weight base. The total phenolic content varied from 340 to 464 mg/100 g fresh weight. The biotype S-07 had the highest total flavonoids content (241 mg quercetin equivalent/100 g fresh weight) and the lowest value attributed to biotype S-01 as 151 mg/quercetin equivalent/100 g fresh weight. The antioxidant activity of the biotypes was between 14.2-30.7 μg Trolox/ml sample. The results revealed that there was enough diversity among Rosa canina biotypes for bioactive content and promising biotypes with high bioactive content are ready material for future breeding activities.

A rose hip is the fruit of a rose plant and mostly belongs to Rosa canina and Rosa dumalis. These species are genuine found as wild in natural conditions, while in some countries are cultivated for their hips that are picked and used in a wide variety of preparations. Because they contain a variety of antioxidants, carotenoids, flavonoids, polyphenols, leucoanthocyanins and catechins, rose hips are considered to be a good cancer preventative. The Rosa genus has been the focus of many recent studies due to its potential benefic effects on treatment and prevention of several diseases. However, there are few in vitro studies concerning its composition and antioxidant capacity. The aim of the present study was to determine bioactive content, including ascorbic acid, total phenolics, total flavonoids, total carotenoids and antioxidant activity of Rosa dumalis genotypes growing wild in Erzurum province located eastern Anatolia. The results revealed that fruits of Rosa dumalis different genotypes were rich rich in terms of vitamin C, which ranged from 402 to 511 mg/100 g fresh weight base. The total phenolic content varied from 297 to 403 mg/100 g fresh weight. The genotype 'E-09' had the highest total flavonoids content (229 mg quercetin equivalent/100 g fresh weight) and the lowest value was attributed to genotype 'E-04' with 143 mg/ quercetin equivalent/100 g fresh weight. The antioxidant activity of the genotypes was between 12.9-28.6 μg Trolox/ml samples. The results revealed that there was enough diversity among Rosa dumalis genotypes for bioactive content and promising genotypes, with high bioactive content, were determined, which can become study material for future breeding activities.
