Çakır, Özlem; Çınar Topçu, Kübra; Sefali, Abdurrahman; Bayyiğit, İshak; İlhan, Gülçe; Singh, Jyoti
This study compared the antioxidant capacity, total phenolic content (TPC), phenolic composition, lipid oxidation level, and mineral profiles of extracts obtained from four wild hawthorn species (Crataegus tanacetifolia, C. orientalis subsp. orientalis, C. orientalis subsp. szovitsii, and C. monogyna) collected from northeastern Türkiye. Significant interspecific variations were observed in DPPH, FRAP, and CUPRAC assays. While C. monogyna had the highest reducing capacity and TPC, C. orientalis subsp. szovitsii had the highest DPPH activity. Ultra-high performance liquid chromatography coupled to tandem mass spectrometry analysis showed that catechin, epicatechin, chlorogenic acid, and quinic acid were the major phenolics, and were particularly abundant in C. monogyna. Despite having a high phenolic content, C. monogyna had the highest malondialdehyde level, indicating that phenolics alone were insufficient to fully inhibit lipid oxidation. Inductively coupled plasma-mass spectrometry mineral analysis detected high levels of K, Mg, and P in all species. Principal component analysis effectively distinguished the species based on their antioxidant, phenolic, and mineral profiles. Overall, hawthorn fruits were confirmed as valuable sources of natural antioxidants and essential minerals.

Oztekin, Sebahat; Dikmetas, Dilara Nur; Karbancıoglu-Guler, Funda
In the present study, a novel exopolysaccharide EPS-BMD26 produced by cold-adapted yeast Rhodotorula glutinis was structurally examined, along with its rheological, antioxidant, and antibiofilm properties. High-performance liquid chromatography (HPLC) analysis revealed glucose and galactose as two monosaccharide constituents. The functional groups within the EPS-BMD26 structure were detected by Fourier transform infrared (FT-IR) spectroscopy. The 1H and 13C nuclear magnetic resonance (NMR) spectra of EPS-BMD26 also confirmed its β-D-glucan type with galactose residues with an average molecular weight of 118 kDa. Differential scanning calorimetry (DSC) analysis revealed that EPS-BMD26 had a high level of thermal stability, up to 326.16 °C. Scanning electron microscopy (SEM) analysis demonstrated that EPS-BMD26 had a porous microstructure with fissures. X-ray diffraction (XRD) analysis revealed its semi-crystalline nature. EPS-BMD26 showed moderate and concentration-dependent antioxidant potential recruiting different methods, including •OH, DPPH•, CUPRAC, and ABTS radical scavenging activities. Water-soluble EPS-BMD26 exhibited high water holding capacity of 190 ± 0.22% with a water solubility index of 60.6 ± 8.3%. At 1.25 mg mL-1, EPS-BMD26 inhibited biofilm formation of Staphylococcus aureus ATCC 25923 by 79.5%. Rheological characterization revealed that the aqueous solution of EPS-BMD26 had shear thinning and pseudoplastic behavior. Given these properties, EPS-BMD26 may be a promising bio-hydrocolloid, antibiofilm agent, and food additive for industrial applications.

Chestnut (Castanea sativa) blossoms are natural resources that are not put to economic use. They are completely mixed with soil as waste. Thus, this extensive study was designed and remarkable results were found showing the potential usefulness of chestnut blossoms. In addition to the phenolic capacity and antioxidant capacity of the aqueous and ethanolic extracts of dried chestnut flowers, the anti-urease activity of these extracts was studied to demonstrate their therapeutic value. The binding interaction of phenolic substances present in chestnut blossom with urease was shown using molecular docking research. The aqueous extract, with most effect, had total phenolic content of 46.67 ± 0.37 mg GAE/g and total flavonoid content of 6.14 ± 0.40 mg QUE/g. The antioxidant activity was determined by FRAP (648.47 ± 5.27 µmol FeSO4.7H2O/g for aqueous extract and 347.53 ± 2.09 µmol FeSO4.7H2O/g for ethanolic extract) and DPPH (0.05 ± 0.01 mg/mL for SC50 of aqueous extract and 0.11 ± 0.01 mg/mL for SC50 of ethanolic extract) assays, and rutin was found to be the dominant phenolic compound according to HPLC. IC50 values for urease in aqueous and ethanolic extracts were 2.55 ± 0.09 mg/mL and 4.57 ± 0.24 mg/mL, respectively. According to the docking experiments, which were important to support the hypothesis of anti-urease activity, myricetin and luteolin showed different and effective bonding degrees to the target protein when compared with the reference molecule acetohydroxamic acid. In summary, chestnut flowers are rich in phenolic compounds which are responsible for a wide range of biological activities including antioxidant features and urease inhibition. These blossoms could be evaluated as potentially important raw materials for food.

Keskin, Merve; Kaya, Gülşen; Bayram, Sinan; Kurek-Górecka, Anna; Olczyk, Paweł
In this study, chestnut honey-based silver nanoparticles (CH-AgNPs) were synthesized at different temperatures (30, 60 and 90 °C) and these nanoparticles were characterized by different techniques such as UV–vis spectrophotometer, Fourier transform infrared spectroscopy (FTIR), scanning electron microscopy (SEM) and energy dispersive X-ray (EDX). The DPPH free radical scavenging assay was used to determine the antioxidant activity of the obtained nanoparticles. The inhibition effects of these nanoparticles for some clinically important enzymes such as myeloperoxidase and collagenase were investigated. In addition, the disk diffusion method (DDM), agar well diffusion (AWD), and minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) techniques were used to determine the antibacterial activity of CH-AgNPs. In honey-based silver nanoparticle production processes using green synthesis, it was determined that the nanoparticle sizes decreased from 55 to 27 nm with an increase in temperature. In addition, it was determined that the rate of inhibition of myeloperoxidase (36.4% to 34.0%) and collagenase enzymes (74.2% to 68.7%) increased with a decrease in particle size. As a result of the antibacterial activity tests, it was observed that CH-AgNPs have antibacterial activity against all target pathogens including Gram-positive and Gram-negative bacteria. The obtained results show that CH-AgNPs produced using chestnut honey have the potential to be used in fields such as medicine, pharmacy and cosmetic technology.

In recent years, the popularity of natural products and natural product formulations has increased due to their beneficial effects on health. As a part of this trend, Lamiaceae family members have attracted attention due to their rich bioactive properties. Although the Stachys rizeensis R.Bhattacharjee plant is an endemic plant species in Turkey, no studies focus on its characterization. Hence, the present study aims to demonstrate the anatomical, morphological and phytochemical characterization of S. rizeensis in Turkey. In the first stage of the study, anatomical and trichome morphological characterization was performed to contribute to the taxonomic definition of S. rizeensis was found to be anatomically different from some other previously studied Stachys sp. by showing lacunar collenchyma type, uncertain cambium at the corners, two or three rows of endodermis in certain areas in the stem and two rows of palisade parenchyma, capitate and peltate glandular trichomes in the leaf. In the second stage, the phenolic profile, total phenolic/flavonoid content and antioxidant activity of the plant extracts prepared using solvents with different polarities (ethanol, methanol, water, ethyl acetate and hexane) were tested to determine the phytochemical content of the plant. The phytochemical analysis supported by chemometric analysis (PCA and HCA) revealed that methanol was the most efficient organic solvent for the extraction of the phytochemical compounds of the plant. Additionally, it was observed that chlorogenic acid (1842.40–217,086.19 mg/kg), verbascoside (2591.43–204,429.81 mg/kg), luteolin (3579.56–88,636.14 mg/kg) and kaempferol (9706.30–50,340.93 mg/kg) compounds were present at maximum levels in this plant, in which 31 different phenolic compounds were scanned using LC-MS/MS. In addition, results obtained from the phytochemical analysis revealed that S. rizeensis is a valuable plant that can be used for medical protection/treatment and that it could also have a variety of culinary applications. The present study served as the first original report regarding the chemical profile and anatomical characteristics of S. rizeensis (Lamiaceae) from Turkey.

Kaya, H.; Karaalp, M.; Kaynar, Ö.; Tekçe, E.; Aksakal, A.; Bayram, B.
The study was conducted to determine the effects of stocking density (SD, 5 or 7 layers/cage) and tarragon (Artemisia dracunculus L.) diet supplementation at four levels (0, 1.2, 6 and 12 g/kg feed) on performance, certain egg characteristics, serum, liver, egg yolk and small intestine bacteria parameters in laying hens. The experiment was carried out over a period of 8 weeks, with 192 Lohman Brown commercial hybrids at 50-w-age. The results showed that an increased SD reduced feed intake (FI; p<0.01) and egg production (p<0.05), but had no effect on the weight gain, feed conversion ratio, damaged egg ratio, egg weight and egg quality (p>0.05) parameters. The supplementation of tarragon to the diet reduced the FI and damaged egg ratio (p<0.01), and improved egg production and FCR (p<0.01). While an increased SD reduced serum total antioxidants (p<0.05), it elevated corticosterone (CORT) and total oxidant serum (TOS) (p<0.05). Tarragon was found to enhance total immunoglobulin (p<0.05), but to decreased the CORT and TOS of the serum (p<0.05). An increased SD raised the malondialdehyde (MDA) in the serum (p<0.001), liver (p<0.05) and yolk (p<0.001). Tarragon supplementation reduced MDA of the serum (p<0.05), liver (p<0.001) and yolk (p<0.001). E. coli and total Mesophilic Aerobic Bacteria counts in the small intestine were raised (p<0.001) with increased SD. Tarragon decreased (p<0.05) mesophilic aerobic bacteria. It was thus found that, tarragon supplementation can be considered generally effective in improving performance parameters, alleviating stress-induced negativities, reducing lipid peroxidation, regulating the immune system and controlling some intestinal microorganisms.

İncele - Derleme MakaleAçık Erişim2020Scopus SJRQ1Effects of umbelliferone isolated from the Ferulago pauciradiata Boiss. & Heldr. Plant on cecal ligation and puncture-induced sepsis model in rats
Kutlu, Z.; Celik, M.; Bilen, A.; Halıcı, Z.; Yıldırım, S.; Karabulut, S.; Karakaya, S.; Bostanlık, Delimustafaoğlu F.; Aydın, P.
Sepsis is a pathophysiological event involving systemic inflammatory response syndrome, multiple organ failure syndromes, and tissue damage. Overproduction of free radicals as a result of tissue damage during sepsis contributes to cellular toxicity, organ failure, and even mortality. Antioxidants, which scavenge free radicals, play a protective role against various diseases. Previous studies have shown that umbelliferone (UF) has antioxidant and anti-inflammatory effects. Since oxidative stress is naturally associated with sepsis-induced organ dysfunction, the application of antioxidant compounds could potentially illuminate the pathophysiology of sepsis, which does not yet have an effective treatment. The sepsis model induced by cecal ligation and puncture (CLP) was applied to rats. Different doses of UF (10░mg/kg, 20░mg/kg, and 40░mg/kg) on oxidant-antioxidant in septic rats, mRNA of inflammatory mediators such as tumor necrosis factor- α (TNF-α) and interleukin (IL)-1 its effects on expression levels were evaluated in lung, kidney, and liver tissues. When the lung, kidney, and liver tissues of septic rats were compared with those of the control group, it was found that UF administration increased dose-dependent superoxide dismutase activity and glutathione levels and significantly decreased malondialdehyde levels. The effects of UF administration on oxidative parameters were dose-dependent. The 40░mg/kg UF dose showed greater anti-oxidative properties than the 20░mg/kg and 10░mg/kg doses for all the evaluated parameters. Further, the TNF- α mRNA expression of the CLP +40░mg/kg group was reduced to a level comparable to that of the control group. UF has been found to be an effective molecule in reducing oxidative stress by supporting endogenous antioxidants and enhancing the scavenging effects of free radicals. The potent antioxidant property of UF may also be related to the suppression of the cytokine cascade during sepsis. The results suggest that UF administration may represent a new treatment for the prevention of lung, kidney and liver damage caused by septic conditions.

Anzer honey is produced on the Anzer plateau, known for its rich flora, in the Eastern Black Sea region of Turkey. It is well known across the world, and is believed to be of high medical value. This study is the first detailed research in which the polyphenolic profiles responsible for the bioactive properties of Anzer honey was determined, melissopalynological analysis was performed, and in which total phenolic contents (TPC), total flavonoid contents (TFC), and total antioxidant activities were identified. The ferric (III) reducing antioxidant power (FRAP) assay and 1,1-diphenyl-2- picrylhydrasil (DPPH) free radical scavenging test were used to determine antioxidant activity. The total mean phenolic content of Anzer honey was 26.92 GAE/100 g, and the total flavonoid content was 2.79 mgQE/100g. Anzer honeys' mean FRAP and DPPH values were 110.11 μmoLTrolox/100g and 49,12 mg/mL, respectively. The phenolic acids and flavonoids of Anzer honey were determined using highperformance liquid chromatography (HPLC). Of the 19 standard compounds used in the analysis, pinocembrin, hesperidin, chrysin, protocatechuic acid, p-coumaric acid, catechin, caffeic acid phenyl ester (CAPE), p-OH benzoic acid, and caffeic acid as major compounds, while myricetin, luteolin, rutin, resveratrol, epicatechin, t-cinnamic acid, ferulic acid, and gallic acid were identified as minor compounds. Daidzein and syringic acid were not detected. Based on these findings, Anzer is a flower honey noteworthy for the rich variety of its polyphenols.

Leuconostoc mesenteroides S81 was isolated from traditional sourdough as an exopolysaccharide (EPS) producer strain. The monosaccharide composition of the EPS from strain S81 was characterized by HPLC analysis and only fructose was found in the repeating unit structure. The NMR spectroscopy analysis revealed that EPS was a levan type EPS as a β-(2 → 6)-linked fructan. The FTIR analysis further confirmed the presence of the furanoid rings in the EPS structure. The levan S81 showed high level of thermal stability determined by DSC and TGA analysis. The lyophilised levan S81 showed a sheet-like compact morphology and its aqueous solution formed spheroidal lumps with a compact structure detected by SEM and AFM analysis, respectively. Importantly the levan S81 showed a high level of immunomodulatory role, induced the anti-inflammatory cytokine IL-4, and exhibited a strong antioxidant capacity with EC50 value 1.7 mg mL−1 obtained by hydroxyl radical scavenging activity test under in vitro conditions. These findings reveal potential of levan S81 for technological purposes and as a potential natural immunomodulatory and antioxidant.

The purpose of the study was to analyze the po-tential health-pro mo ting components of some edible forest species of fruits. These are Service tree (Sor-bus domestica), Black rosehip (Rosa pimpinelli-folia), Red rosehip (Rosa carina), Bay fruit {Lawns nobilis), Cornelian cherry (Cornus mas), Autumn-olive (Elaeagrtus umbellate), and two different blue-berry fruits (Vaccinium arctostaplylos and Vaccin-ium myrtillus). In the present study, chemical and an-tioxidant activities, phenolic compounds as well as, inhibition effects on important enzymes (acetylcho-linesterase, xanthine oxidase and urease) of fruits were studied. Total polyphenol, flavonoid content, and the in-vitro antioxidant activity were evaluated by DPPH and FRAP. The sugar contents and phe-nolic compounds of the fruits were analyzed by HPLC-RJD and HPLC-UV. Fruits was determined the total polyphenol content (TPC) 1.10 to 19.87 mgGAE/g, total flavonoid content (TFC) 0.01 to 0.80 mgQE/g, DPPH values 0.22 to 6.03 mg/mL and FRAP values 4.24 to 168. 02 umol FeSO47H2O/g. All samples were determined various rate fructose and glucose. HPLC-UV revealed fruits contained different phenolic compounds. Inhibition values of the enzymes were expressed as inhibition concentra-tion (IC50: Mg/mL or ug/mL). It can be concluded that edible forest fruits are a potential source of anti-oxidants with therapeutic importance as well a natu-ral enzyme inhibitor.

This study aimed in sedentary subjected to progressive relaxation training (PRT) for determination of the effect of this exercise on MDA, OHdG, and NO levels and SOD and GPx activities. Experimental design was used in this research. Seven healthy, sedentary volunteers were included in the study. All subjects were male and within 15 to 18 years of age. Jacabson's Progressive Muscle Relaxation Training was applied to the volunteers for 6 six weeks. Before and after the chronic and acute exercise, blood samples were taken from the subjects, and K-Related Samples Friedman test was used for the analysis of repeated measurements and for the comparison of pre- and post-exercise data and chronic and acute mode data. In addition, 2-Related Samples Wilcoxon test was used in the evaluation of the groups as pairs. Our findings showed increases in MDA PRT, and no statistically significant increase in NO levels after PRT in sedentary volunteers. When the effect of relaxation exercise on the antioxidant system is considered, both SOD and GPx activities increased. The effect of PRT on 8-OHdG has been found to exhibit significant increase after PRT. It can easily be said that PRT may be helpful in patients with chronic disease, especially for obtaining a healing and a high quality sleep. In addition, in order for the athletes to feel well and to use their muscles actively, PRT may be helpful. We can conclude that, with respect to human health and physiology, no controversy may be seen in progressive muscle relaxation training.
