The design and synthesis of hydrazone derivatives are increasing in popularity day by day due to the significant biological activities of this scaffold. In the present study, twelve novel isonicotinic hydrazide-hydrazone analogues were synthesized by the condensation reaction of isonicotinic hydrazide with benzaldehyde possessing sulfonate moiety. The structures of the novel compounds have been characterized in detail using spectroscopic techniques. All compounds have shown inhibitory effects against the AChE enzyme at rates ranging from 21.00 to 59.48%. Among them, compound 5 has exhibited the best inhibitory effect of 59.48% against AChE at a concentration of 0.1 mM. Furthermore, to determine how effective the novel compounds are as antioxidants, FRAP and DPPH studies were also carried out. FRAP values in compounds 1–12 were found to range from 26.989–3415.556 µmol FeSO4.7H2O/mg. They also displayed moderate antioxidant potential in the range of SC50=0.03–87.32 mg/mL compared with the control Trolox (SC50 =0.004) in DPPH radical scavenging activities. It was seen that the AChE inhibition percentages of the compounds were in the range of 23.04–58.10% at 0.1 mM concentration. This is the first research on the synthesis, antioxidant and enzyme inhibition properties of these compounds.

İncele - MakaleAçık Erişim2022Scopus SJRQ2What Should Be the Ideal Solvent Percentage and Solvent-Propolis ratio in the Preparation of Ethanolic Propolis Extract?
Kara, Yakup; Can, Zehra; Kolaylı, Sevgi
The biological activity of propolis is mostly due to its polyphenols. Raw propolis is consumed after being extracted in various solvents, ethanol being the most commonly used. However, the ideal percentage and ratio of ethanol for use in the preparation of propolis extracts are uncertain. The aim of this study is to compare the phenolic compositions and antioxidant activities of propolis extracts extracted with different ethanol concentrations and extraction conditions. Two different extraction methods were used, normal and ultrasonic extraction. The propolis extracts were evaluated in terms of total phenolic content, total flavonoid content, ferric reducing/antioxidant power,1,1-diphenyl-2-picrylhydrazyl radical scavenging activity, and phenolic compositions using RP-HPLC–PDA. The propolis extracts with the highest activity were determined beforehand by preparing ethanol–water mixtures at different percentages, ranging from 0 to 99.9%. The most suitable propolis-to-solvent ratio was then determined. The optimum ethanolic percentage varied between 60 and 70%, the ultrasonic extraction process increased the efficiency, and the ideal solvent-propolis ratio (w/v) ranged between 1:5 and 1:10.

Gurgen, Ayşenur; Serencam, Huseyin; Kara, Yakup; Can, Zehra; Ylldlz, Sibel
In this study, the optimum values of propolis ethanol extracts parameters were determined with the use of single and multi-objective optimization procedures. The euclidean distance approach was used in the multi-objective optimization process. Firstly, propolis was extracted using water with ethanol contents 40, 50, 60, 70 and 80% for 8, 10, 12, 16, 20 and 24 h. Then, total phenolic content (TPC) and ferric reducing antioxidant power (FRAP) activities of all extracts were determined. With the obtained data a prediction model was produced with the use of artificial neural networks (ANN), and optimization was performed using a moth-flame (MFO) algorithm. The best prediction models for the TPC and FRAP were observed in 2-5-1 and 2-5-1 network architecture with the mean absolute percentage error (MAPE) values, 5.126 and 2.451%, respectively. For maximum TPC, the extraction parameters were determined as ethanol content 57.5% and extraction time 13.56 h. To maximize FRAP, the optimized extraction parameters were ethanol content 72.03% and extraction time 18.04 h. The optimum extraction conditions for both maximum values of the studied assays were ethanol content 70.03% and extraction time 16.93 h. The study concluded that the integrated ANN and MFO algorithm system can be used in single and multi-objective optimization of extraction parameters. The established optimization model can save time, money, labor and energy.

HMF (5-hydroxymethylfurfural) is one of the major intermediate products in the Maillard reaction that can lead to quality reduction in heated honey. In this study, the effect of heating on the antioxidant activity, and colour values as Maillard reaction indicators of three different botanical honeys were investigated; the HMF formation was also determined. Temperatures of 50, 70 and 80 °C were applied on the honeys between 0 and 48 h. Total phenolic content, ferric reducing/antioxidant power and scavenging of 2,2-diphenyl-1-picrylhydrazyl free radical assays are used to determine the antioxidant capacity. Results showed that the formation of HMF and the antioxidant properties of honeys were significantly increased during the heating process. Pure HMF compound also showed lower antioxidant activity. The formation of HMF has higher degree of linearity in the fit of the zero order reaction and also it was the highest in the chestnut honey. Furthermore, it was found that as the biochemical value of the honeys increased, the HMF formation decelerated.
