A natural nephroprotective adjuvant for cancer chemotherapy: Rosmarinic acid disrupts IL-17 A-Ferroptosis coupling in Ifosfamide-induced renal injury

Süzen Celbek, Büşra; Şimşek, Hasan; Akaras, Nurhan; Kandemir, Özge; Kızıl, Hamit Emre; Mutlu, Hüseyin; Kandemir, Fatih Mehmet

Rosmarinic acid (RA) is a natural polyphenol with established pleiotropic protective effects. Ifosfamide (IFA) is a potent antineoplastic agent whose clinical utility is severely limited by dose-dependent nephrotoxicity, primarily mediated by its metabolite, chloroacetaldehyde (CAA). This study aimed to investigate whether RA protects against IFA-induced nephrotoxicity and to elucidate the underlying molecular mechanisms. Male Wistar albino rats (n = 7 per group) were allocated into four groups: Control, RA-only (50 mg/kg, p.o., 2 days), IFA-only (a single 500 mg/kg, i.p. dose), and IFA + RA. Serum biochemical markers (urea, creatinine), renal oxidative stress parameters (MDA, GSH, SOD, CAT, GPx), gene expression levels (NF-κB, TNF-α, IL-1β, IL-17 A, ACT1, TRAF6, Caspase-3, Bax, Bcl-2, PTGS2, GPX4, TfR1), and histopathological/immunohistochemical analyses (KIM-1, Nephrin, Caspase-3) were performed 24 h post-administration. IFA induced severe renal dysfunction, marked oxidative stress, and extensive histopathological damage. Mechanistically, IFA initiated a pathogenic cascade activating intrinsic apoptosis and ferroptosis, driven by a self-sustaining IL-17 A-TRAF6-NF-κB inflammatory loop. RA co-treatment (50 mg/kg) significantly reversed all functional, biochemical, and histological damage by strategically breaking this crosstalk, restoring redox homeostasis, and simultaneously restraining both cell death programs. In conclusion, RA protects against IFA nephrotoxicity by targeting the critical inflammation-ferroptosis coupling, positioning it as a highly promising adjuvant candidate for clinical use to mitigate IFA-induced renal injury.

Effects of Moringa oleifera Oil on Adipokine Responses, Inflammation, Oxidative Stress, Bacterial Burden, and Early Survival in CLP-Induced Septic Rats

Ülker, Ufuk; Bayraktar, Bülent; Alr, Mehmet Eray; Kızıl, Sibel; Karan, Tünay; Kotan, Gökşad Cemil; Yeni, Mustafa

Moringa oleifera has anti-inflammatory and antioxidant properties, but its effects in polymicrobial sepsis are incompletely defined. We evaluated M. oleifera oil (MOO) in female Wistar rats subjected to cecal ligation and puncture (CLP). Forty rats were randomized to Control, Sham, CLP, CLP + MOO 100 mg/kg, or CLP + MOO 200 mg/kg (n = 8/group). At 24 h, survival, clinical scores, serum apelin, omentin-1, interleukin-6 (IL-6), tumor necrosis factor-α (TNF-α), malondialdehyde (MDA), recoverable bacterial burden, and histopathology were assessed. CLP decreased omentin-1 and increased IL-6, TNF-α, MDA, recoverable Staphylococcus aureus and Escherichia coli, clinical severity, and multi-organ injury. At 200 mg/kg, apelin and omentin-1 were 1.15 ± 0.12 and 28.4 ± 3.1 ng/mL, while IL-6, TNF-α, and MDA decreased to 18.4 ± 1.8 pg/mL, 24.1 ± 2.8 pg/mL, and 2.5 ± 0.3 nmol/mL, respectively. Neither organism was recovered above the detection limit. Survival was 62.5% in CLP, 87.5% with 100 mg/kg, and 100% with 200 mg/kg MOO. Within this 24 h model, a single post-CLP oral dose of MOO was associated with lower inflammatory cytokine concentrations and MDA, reduced recoverable bacterial burden, improved clinical and histopathological findings, and higher descriptive survival. Because MDA was the sole oxidative endpoint and no non-septic MOO-only group or molecular redox assays were included, these findings support the attenuation of sepsis-associated lipid peroxidation but do not establish a direct antioxidant mechanism.

Multi-Pathway Renoprotection by Betanin Against Docetaxel Nephrotoxicity: Modulation of Inflammation, ER Stress, Apoptosis, and Ferroptosis

Süzen Celbek, Büşra; Akaras, Nurhan; Şimşek, Hasan; Mutlu, Hüseyin; Kızıl, Hamit Emre; Kandemir, Özge; Aygörmez, Serpil; Kandemir, Fatih Mehmet

Docetaxel (DTX), a cornerstone chemotherapeutic agent, has its clinical utility significantly limited by dose-dependent nephrotoxicity. This study investigated the multi-pathway renoprotective mechanisms of betanin (BTN), a natural antioxidant derived from beetroot, against DTX-induced kidney injury. Male Wistar rats received DTX (30 mg/kg, i.p., single dose) with or without BTN (100 mg/kg/day, p.o.) for 7 days. DTX induced severe renal dysfunction with elevated serum urea and creatinine, and significant oxidative stress characterized by increased MDA and decreased GSH, GPx, SOD, and CAT. Molecular analysis demonstrated widespread upregulation of pro-inflammatory (NF-κB, TNF-α, IL-1β), endoplasmic reticulum stress (PERK, ATF6), pro-apoptotic (Caspase-3, Bax, Apaf-1), and pro-ferroptotic (PTGS2, TfR1) mediators, with concurrent downregulation of protective Bcl-2 and GPx4. Histopathological examination revealed extensive tubular injury, while immunohistochemical analysis showed markedly increased kidney injury marker KIM-1 and decreased aquaporin-1 expression. Remarkably, BTN co-administration significantly attenuated all biochemical, molecular, and structural derangements, restored tissue architecture, and normalized KIM-1 and AQP-1 expression. In conclusion, betanin provides robust, multi-pathway renoprotection against docetaxel-induced nephrotoxicity by concurrently targeting oxidative stress, inflammation, ER stress, apoptosis, and ferroptosis.

Dietary Carvacrol Attenuates Cyclophosphamide-Induced Neurotoxicity: Implications for Food-Derived Neuroprotection and Molecular Mechanisms

    Kızıl, Hamit Emre

Carvacrol (CRV) is a phenolic monoterpene abundant in culinary herbs such as oregano and thyme and is well known for its potent antioxidant, anti-inflammatory, and neuroprotective properties. This study investigated the ability of CRV to counteract neurotoxicity induced by cyclophosphamide (CP), a widely used antineoplastic agent. Male Wistar albino rats were divided into five groups and received CP and/or CRV treatments. Neurotoxicity and neuroprotection were evaluated through biochemical assays, real-time PCR, histopathological and immunohistochemical analyses, and behavioral testing (Morris Water Maze). CP administration led to significant increases in oxidative stress markers, disruption of antioxidant enzyme activities, upregulation of inflammatory mediators (NF-κB, TNF-α, iNOS), dysregulation of apoptotic regulators (increased Bax and Casp-3, decreased Bcl-2), alterations in autophagy markers (Beclin-1, LC3A, LC3B), and suppression of Notch1/Hes1 signaling. Histopathological analyses revealed neuronal degeneration, vascular hyperemia, and increased GFAP and 8-OHdG expression in brain tissue. CRV treatment, particularly at higher doses, effectively mitigated these biochemical, molecular, and histological alterations. Notably, CRV administration preserved spatial learning and memory function in CP-treated rats, as demonstrated by the Morris Water Maze test, indicating functional neuroprotection. These findings highlight the multifaceted neuroprotective mechanisms of CRV and suggest its potential as a food-derived bioactive compound for development into functional foods or dietary supplements to improve the quality of life for cancer patients undergoing chemotherapy.

Evaluating STC-1 and STC-2 mRNA expressions in Schizophrenia patients with increased oxidative stress; [Évaluation des expressions d'ARNm de STC-1 et STC-2 chez des patients schizophrènes présentant un stress oxydatif accru]

Karabulut Uzunçakmak, Sevgi; Özcan, Halil; Dirican, Ebubekir; Tavacı Özçelik, Ayşegül

Introduction: Schizophrenia is a chronic psychiatric disorder characterized by cognitive impairment. Oxidative stress is associated with disease progression in patients with schizophrenia. Stanniocalcin (STC)-1 and STC-2 are two proteins commonly expressed in mammals belonging to the stanniocalcin family and they are involved in oxidative stress. The aim of this study is to investigate STC-1 and STC-2 mRNA expressions in schizophrenia patients with altered oxidative stress parameters. Methods: For this purpose, 70 patients with schizophrenia and 40 healthy individuals were recruited for the study. Peripheral blood samples were obtained from all participants. Glutathione (GSH) activities, superoxide dismutase (SOD) activities, and malondialdehyde (MDA) levels were measured. STC-1 and STC-2 expressions were assessed by real-time polymerase chain reactions. Results: SOD activity levels were lower in patients than in healthy individuals (p = 0.0309), while the patients’ MDA levels were higher (p = 0.039). STC-1 and STC-2 expressions were lower in patients than in healthy individuals (p = 0.1049 and p < 0.0001, respectively). Furthermore, these two genes had a positive correlation among the patients (r = 0.435, p = 0.0025). According to area under the curve (AUC) values, STC-2 (AUC = 0.8332, p < 0.0001) had better diagnostic power than STC-1 (AUC = 0.6167, p = 0.1037). Conclusions: The expression of stanniocalcins in schizophrenia was investigated here for the first time. Decreased STC-2 expression in patients with schizophrenia with increased oxidative stress parameters may guide the understanding of the pathophysiological mechanisms caused by oxidative stress, which may be increased in cognitive diseases such as schizophrenia, and it also has the potential to be a prognostic factor that can be used in the diagnosis of schizophrenia.

Morin ameliorates methotrexate-induced hepatotoxicity via targeting Nrf2/HO-1 and Bax/Bcl2/Caspase-3 signaling pathways

Kızıl, Hamit Emre; Caglayan, Cuneyt; Darendelioğlu, Ekrem; Ayna, Adnan; Gür, Cihan; Kandemir, Fatih Mehmet; Küçükler, Sefa

Background: Organ toxicity limits the therapeutic efficacy of methotrexate (MTX), an anti-metabolite therapeutic that is frequently used as an anti-cancer and immunosuppressive medicine. Hepatocellular toxicity is among the most severe side effects of long-term MTX use. The present study unveils new confirmations as regards the remedial effects of morin on MTX-induced hepatocellular injury through regulation of oxidative stress, apoptosis and MAPK signaling. Methods and results: Rats were subjected to oral treatment of morin (50 and 100 mg/kg body weight) for 10 days. Hepatotoxicity was induced by single intraperitoneal injection of MTX (20 mg/kg body weight) on the 5th day. MTX related hepatic injury was associated with increased MDA while decreased GSH levels, the activities of endogen antioxidants (glutathione peroxidase, superoxide dismutase and catalase) and mRNA levels of HO-1 and Nrf2 in the hepatic tissue. MTX treatment also resulted in apoptosis in the liver tissue via increasing mRNA transcript levels of Bax, caspase-3, Apaf-1 and downregulation of Bcl-2. Conversely, treatment with morin at different doses (50 and 100 mg/kg) considerably mitigated MTX-induced oxidative stress and apoptosis in the liver tissue. Morin also mitigated MTX-induced increases of ALT, ALP and AST levels, downregulated mRNA expressions of matrix metalloproteinases (MMP-2 and MMP-9), MAPK14 and MAPK15, JNK, Akt2 and FOXO1 genes. Conclusion: According to the findings of this study, morin may be a potential way to shield the liver tissue from the oxidative damage and apoptosis.

Suberosin Alleviates Sepsis-Induced Lung Injury in A Rat Model of Cecal Ligation and Puncture

Karabulut Uzunçakmak, Sevgi; Halıcı, Zekai; Karakaya, Songül; Kutlu, Zerrin; Sağlam, Yavuz Selim; Bolat, İsmail; Aydın, Pelin; Kılıç, Ceyda Sibel

Background/aims: Sepsis is one of the major problems encountered in intensive care units, causing organ damage and increasing mortality. Suberosin (SBR) is a type of coumarin with antioxidant and anti-inflammatory activities. The goal of this study is to explore the protective effects of SBR on the lungs in a rat model of sepsis. Methods: Male Wistar rats were utilized in this study. A cecal ligation and puncture (CLP) model was applied to induce sepsis. Rats were separated into six groups with nine animals in each group, including healthy control, SBR, CLP, and CLP + SBR (5, 10, and 20 mg/kg) groups. Superoxide dismutase (SOD), glutathione (GSH) enzyme activities, and malondialdehyde (MDA) level were measured via enzyme-linked immunosorbent assay (ELISA). The messenger RNA (mRNA) expressions of tumor necrosis factor α (TNF-α) and interleukin 1β (IL-1β) were evaluated by real-time polymerase chain reaction (RT-PCR). Histopathological changes in the lungs were investigated with hematoxylin and eosin (H&E). Results: MDA levels and GSH and SOD enzyme activities were negatively affected in the CLP group, but SBR treatment ameliorated these oxidative stress parameters in the SBR1-3 groups (p< 0.05). The mRNA expressions of TNF-α and IL-1β were increased in the CLP group, and SBR treatment decreased those expression levels in a dose-dependent manner (p < 0.05). Organ damage and necrosis were seen in the CLP group and were alleviated in the SBR3 group. Immunohistochemical (IHC) analysis of lung tissues demonstrated decreased TNF-α and IL-1β immunopositivity in the SBR1-3 groups (p< 0.05). Conclusions: SBR ameliorated sepsis-related lung injury in a dose-dependent manner. This compound has significant potential as a future agent in the treatment of sepsis.

Thiol disulfide homeostasis in primary dysmenorrhea

Savrun, Atakan; Savrun, Şeyda Tuba; Dirican, Ebubekir; Gökçen, Emre; Arıcı, Yeliz Kasko; Aygün, Ali; Korkmaz, Vakkas; Neşelioğlu, Salim; Erel, Özcan; Kıran, Tuğba Raika

Oxidative stress is defined as a result of the loss of balance between the production of free radical or reactive oxygen species and the antioxidant system. This study aimed to determine the level of thiol-disulfide homeostasis (TDH) in the serum of women with primary dysmenorrhea. The study group consisted of 42 subjects with primary dysmenorrhea, and the control group consisted of 30 volunteer women with demographic characteristics similar to the study group. Native thiol (SH), total thiol (TSH), disulphide (SS), and SH/SS parameters were measured for TDH of the subjects. The SH (p = 0.038) and SH/TSH (p = 0.046) levels were significantly higher while SS (p = 0.013), SS/SH (p = 0.042) and SS/TSH (p = 0.046) levels were lower in the study group than in the healthy control group. The SS cut-off value was determined as 17.85 in the study group (sensitivity = 61.9%, specificity = 43.3%). Therefore, the probability of dysmenorrhea may increase significantly when SS levels fall below this value. In subjects with dysmenorrhea, there was a decrease in SS levels and an increase in SH levels in order to protect the cells and tissues from the harmful effects of free radicals.

Eumelanin protects the liver against diethylnitrosamine-induced liver injury

Altındağ, Fikret; Boğokşayan, Seda; Bayram, Sinan

This study aims to evaluate in vivo protective effects of eumelanin (EU) on diethylnitrosamine (DEN)-induced liver injury. Wistar albino male rats were divided into 6 groups (n = 6), Control, DMSO, DEN, DEN + EU10, DEN + EU15, and DEN + EU20. Animals in the DEN group were injected i.p a single dose of 200 mg/kg DEN, DEN + EU10 group was given 10 mg/kg EU, DEN + EU15 group was given 15 mg/kg, DEN + EU20 group was given 20 mg/kg EU for a week. The results showed that there was no significant difference in vessel volume density between the groups. Inflammatory cell infiltration, hydropic degeneration, and necrotic cells were observed in the DEN group, and these histopathological changes were significantly reduced in all treatment groups. Although there was a low intensity of PAS-positive staining in the DEN groups, moderate staining was observed in the treatment groups. While Caspase-3, PCNA, TNF-α, and IL-6 expressions increased in the DEN group, their expressions decreased in the EU-treated groups. DEN increased AST, ALT, and MDA levels and decreased CAT levels. In particular, the EU10 dose significantly improved these parameters. The present study revealed that eumelanin has protective effects against DEN-induced liver injury.

Effects of processing methods and extraction solvents on the chemical content and bioactive properties of propolis

Ecem Bayram, Nesrin; Gerçek, Yusuf Can; Bayram, Sinan; Toğar, Başak

The aim of this study was to evaluate the bioactive properties of propolis extract prepared using different solvents and different extraction methods. The extracts were analyzed by gas chromatography–mass spectrometry (GC–MS) and the differences between their antibacterial activities were evaluated by disc diffusion method. At the same time, bioactive properties of different concentrations of propolis extracts were investigated on human umbilical vein endothelial cells (HUVEC). The proliferative effects and cytotoxic effects of the extracts were determined by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) and lactate dehydrogenase (LDH) analyzes, respectively. Total antioxidant capacity (TAC) and total oxidative status (TOS) parameters were used in assessing biochemical effects in the HUVEC cell line. The DNA damage was also analyzed by 8-oxo-2-deoxyguanosine (8-OHdG) level as indicators of genotoxicity. As a result of the MTT analysis conducted within the scope of the present study, the extracts tested were sorted as 95% ethanol extract of propolis (PEE95) > ultrasonic ethanol extract of propolis (PUEE) > 50% ethanol extract of propolis (PEE50) > ultrasonic water extract of propolis (PUWE) in terms of the effectiveness of their cell viabilities. It was observed that high concentrations of PEE95 induced LDH release. In addition to this, our findings have shown that PEE50, PUEE and PUWE increased oxidative stress at high concentrations. According to 8-OH-dG analysis, all tested extracts were found to be non-genotoxic. The results obtained from antibacterial activity and minimum inhibition concentration tests showed that PUEE and PEE95 had stronger antibacterial effects than PEE50 and PUWE. All these results indicated that propolis has beneficial effects for human health and therefore it is a valuable product which can be used as a food supplement.

Effects of oregano essential oil (Origanum syriacum l.) on performance, egg quality, intestinal morphology and oxidative stress in laying hens; [Einfluss von oregano-öl (Origanum syriacum l.) auf leistung, eiqualität, darmmorphologie und oxidativen stress bei legehennen]

    Gul, M.; Yilmaz, E.; Yildirim, B.A.; Sezmis, G.; Kaya, A.; Timurkaan, S.; Onel, S.E.; Tekce, E.

This study aimed to investigate the effects that including different levels of oregano essential oil (Origanum syriacum L.) (200, 400, and 600 mg/kg) in the diets of laying hens had on performance, egg quality, and intestinal morphology, as well as egg malondialdehyde (MDA) levels, blood lipid peroxidation, and oxidative stress parameters. The study used a total of 120 Lohmann white laying hens aged 20 to 22 weeks. The experiment comprised the following three treatment groups, based on the dosages of oregano essential oil: 200 mg/kg (A), 400 mg/kg (B), 600 mg/kg (C); and a control group (K) that did not use oregano essential oil. Each group included 30 hens and each was divided into six subgroups with five hens each. In this study, feed intake and egg weight were significantly higher in the treatment groups than in the control group (P=0.046; P< 0.001). There was no difference among the groups in terms of eggshell strength, egg shape index, eggshell thickness, albumin index, or yolk index. However, there was a significant difference in eggshell weight, yolk color and Haugh unit in the experimental groups (P=0.015; P=0.031; P=0.042). The MDA levels in the yolk were lowest in group A (P< 0.05). In all groups with the exception of group A, yolk and serum MDA levels increased while superoxide dismutase, glutathione peroxidase, and catalase decreased as the doses of oregano essential oil increased (P< 0.001; P< 0.001; P< 0.01; P< 0.001). There was no significant serum glutathione level. In the histological analysis of the intestine, the villus height was higher in group C than in the control group (P=0.010). Based on these results, we concluded that 200 mg/kg was the most effective dose of oregano essential oil for improving performance, egg quality, and intestinal morphology while reducing oxidative stress in laying hens.

THE EFFECT OF PROGRESIVE RELAXATION TRAINING ON FREE RADICALS, ANTIOXIDANT SYSTEM AND OXYDATIVE DNA DAMAGE

Aggon, Eser; Ozturk, Nurinnisa; Alp, Hamit Hakan; Siktar, Elif; Agirbas, Ozturk; Bakan, Ebubekir

This study aimed in sedentary subjected to progressive relaxation training (PRT) for determination of the effect of this exercise on MDA, OHdG, and NO levels and SOD and GPx activities. Experimental design was used in this research. Seven healthy, sedentary volunteers were included in the study. All subjects were male and within 15 to 18 years of age. Jacabson's Progressive Muscle Relaxation Training was applied to the volunteers for 6 six weeks. Before and after the chronic and acute exercise, blood samples were taken from the subjects, and K-Related Samples Friedman test was used for the analysis of repeated measurements and for the comparison of pre- and post-exercise data and chronic and acute mode data. In addition, 2-Related Samples Wilcoxon test was used in the evaluation of the groups as pairs. Our findings showed increases in MDA PRT, and no statistically significant increase in NO levels after PRT in sedentary volunteers. When the effect of relaxation exercise on the antioxidant system is considered, both SOD and GPx activities increased. The effect of PRT on 8-OHdG has been found to exhibit significant increase after PRT. It can easily be said that PRT may be helpful in patients with chronic disease, especially for obtaining a healing and a high quality sleep. In addition, in order for the athletes to feel well and to use their muscles actively, PRT may be helpful. We can conclude that, with respect to human health and physiology, no controversy may be seen in progressive muscle relaxation training.

Effects of two lichen acids isolated from Pseudevernia furfuracea (L.) Zopf in cultured human lymphocytes

Emsen, Bugrahan; Togar, Basak; Turkez, Hasan; Aslan, Ali

The present study aims at assessing the efficacies of olivetoric acid (OA) and physodic acid (PA) isolated from Pseudevernia furfuracea (L.) Zopf (Parmeliaceae) in human lymphocytes (HLs) in vitro. 3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide and lactate dehydrogenase assays were performed to establish cytotoxicity in HLs. Besides, oxidative stress and genotoxicity were monitored by estimating the changes of total oxidative stress (TOS) and 8-hydroxy-2'-deoxyguanosine (8-OH-dG) levels, respectively, in HLs. At the same time, OA- and PA-induced total antioxidant capacity (TAC) levels in HLs were determined. Although especially low concentrations of OA (IC50 = 109.94 mg/L) and PA (IC50 = 665.49 mg/L) did not show cytotoxic effect at high levels in HLs, it was revealed that cytotoxicity was significantly (p < 0.05) associated with oxidative stress and genotoxicity via correlation analysis. While TOS level in HLs did not statistically (p > 0.05) increase in the presence of all treatments (0.5-100 mg/L) of PA, TAC level was increased by PA applications in certain concentrations (0.5-10 mg/L). Overall, the obtained data indicate that OA and especially PA as lichen compounds that do not cause oxidative stress can be a new resource of therapeutics as recognized in the present study with their high antioxidant features.

Endurance training attenuates the oxidative stress due to acute exhaustive exercise in rat liver

    Taysi, S.; Oztasan, N.; Efe, H.; Polat, M.F.; Gumustekin, K.; Siktar, E.; Canakci, E.; Akcay, F.; Dane, S.; Gul, M.

The aim of this study was to investigate whether an 8-week treadmill training attenuates exerciseinduced oxidative stress in rat liver. Male rats were divided into untrained and trained groups. Endurance training consisted of treadmill running at a speed of 2.1 km/h, 1.5 h/day, 5 days a week for 8 weeks. To see the effects of endurance training on acute exhaustive exercise induced oxidative stress, untrained and trained rats were further devided into two groups: animals killed at rest and those killed after acute exhaustive exercise, in which the rats run at 2.1 km/h (10% uphill) until exhaustion. Acute exhaustive exercise increased malondialdehyde level in untrained but not in trained rats. It decreased the activity of glutathione peroxidase and total (enzymatic plus non-enzymatic) superoxide scavenger activity in untrained rats and catalase activity in trained rats. However, it did not affect glutathione S-transferase, glutathione reductase, superoxide dismutase and non-enzymatic superoxide radical scavenger activities in both trained and untrained rats. On the other hand, endurance training decreased glutathione peroxidase and glutathione S-transferase activities. The results suggested that endurance training attenuated exercise-induced oxidative stress in liver, probably by preventing the decreases in glutathione peroxidase and total superoxide scavenger activities during exercise.
