Lactic acid bacteria can synthesize exopolysaccharides (EPSs), and some of these EPSs have pharmaceutical or medicinal properties. However, the wound healing capacities of EPSs have rarely been reported. In this study, EPS extracted from Weissella cibaria MED17 was used to investigate wound healing capacity. MED17 EPS was previously characterized and antioxidant capacity was also demonstrated. The application dose of the compound was determined by the MTT assay. The comet assay was applied to reveal the free radical scavenging activity and protection capacity of MED 17 against DNA damage by inducing oxidative stress and DNA damage with Mitomycin-C (MMC) in the cells. The comet assay showed that 15 mg/ml MED 17 EPS could ameliorate DNA damage by 50%. Moreover, the scratch test investigated the wound-healing effect of MED 17 EPS on HT-29 colon cells. MED 17 EPS could stimulate the migration of HT-29 and ameliorate MMC-induced DNA damage. However, further studies need to be conducted to clarify the mechanisms of the wound-healing process of MED 17.

An exopolysaccharide (EPS) produced by bee pollen isolate Lactobacillus kunkeei AP-27 was characterised in this study. EPS monomeric composition of strain AP-27 was characterised by High-performance liquid chromatography (HPLC) analysis and glucose was the only sugar in the EPS repeating unit. Structural analysis of glucan AP27 by 1H and 13C Nuclear magnetic resonance (NMR) analysis demonstrated that this glucan was a dextran ((1→6)-linked α-D-glucose units as the main body) consisting very low level of (1 → 4)-linked α-D-glucose units (∼ 0.5 %). Dextran AP-27 had a degradation temperature of 308 °C that revealed its superior thermal strength identified by Thermogravimetric (TGA) and Differential scanning calorimetry (DSC) analysis. The molecular weight of dextran AP-27 was determined to be 25 kDa by Gel Permeation Chromatography (GPC) analysis. Crystallographic properties of dextran AP-27 was determined by X-ray diffraction (XRD) analysis that divulged its amorphous feature. Scanning Electron Microscopy (SEM) analysis was applied for microstructural characterisation of dextran AP-27 which revealed its regular sphere like characteristics. These findings suggest that Lactic Acid Bacteria (LAB) strains from different environments such as bee products can be the source for distinct EPS producer strains.

In this study, structural and techno-functional characteristics of an exopolysaccharide (EPS) produced by Lactobacillus kunkeei AK1 were determined. High-performance liquid chromatography (HPLC) analysis demonstrated that EPS AK1 was composed of only glucose units. 1H and 13C Nuclear magnetic resonance (NMR) analysis revealed that EPS AK1 was a dextran type EPS containing 4.78% (1 → 4)-linked α-D-glucose branches. The molecular weight of EPS AK1 was determined to be 45 kDa by Gel Permeation Chromatography (GPC) analysis. A high level of thermal stability up to 280 °C was determined for dextran AK1 detected by Differential scanning calorimetry (DSC) and Thermogravimetric analysis (TGA). Dextran AK1 appeared as regular spheres with compact morphology and as irregular particles in the solution with no clear cross-linking between the chains of the polysaccharide observed by Scanning electron microscopy (SEM) and Atomic force microscopy (AFM) analysis, respectively. X-ray diffraction analysis (XRD) analysis demonstrated that dextran AK1 had a crystalline structure. A relatively strong antioxidant activity was observed for dextran AK1 determined by 2,2-diphenyl-1-picrylhydrazyl (DPPH), 2,2′-azino-bis-3-ethylbenzothiazoline-6-sulfonic acid (ABTS) radical scavenging and cupric reducing antioxidant capacity (CUPRAC) tests. Finally, only a digestion ratio of 3.1% was observed for dextran AK1 following the in vitro simulated gastric digestion test.

An exopolysaccharide (EPS) producer slimy-mucoid type colony was isolated from sourdough and identified as Weissella cibaria MED17. The 1H and 13C NMR spectra of EPS MED17 demonstrated that this EPS was a dextran type glucan ((1 → 6)-linked α-D-glucose core structure) containing (1 → 3)-linked α-D-glucose branches and proportion of (1 → 6)-linked α-D-glucose units to (1 → 3)-linked α-D-glucose units was 94.3:5.7%. The FTIR analysis also confirmed the (1 → 6)-linked α-D-glucose linkage. A high level of thermal stability was observed for glucan MED17 as no degradation up to 300 °C was observed by TGA and DSC analysis. The XRD analysis of glucan MED17 showed its semi- crystalline nature and its compact sheet-like morphology was observed by SEM analysis. Finally, antioxidant characteristics of glucan MED17 were determined by ABTS and DPPH radical scavenging activity tests that revealed a moderate antioxidant activity of glucan MED17. These findings show potential techno-functional characteristics of glucan MED17.

Traditional kefir grains were collected from distinct parts of Turkey, and their microbial profile was determined. A wide bacterial biota was observed formed by distinct lactic acid bacteria (LAB) in which Lactococcus lactis strains appeared to be dominant. Yeast species were also identified in kefir grains. Significant levels of antifungal and antibacterial activities were monitored in kefir isolates. All tested LAB produced an exopolysaccharide (EPS) containing glucose and galactose, and some strains formed a fructan-type EPS. Importantly, low levels of antibiotic resistance were observed among the kefir isolates.

Cereal-associated Lactic Acid Bacteria (LAB) are well known for homopolymeric exopolysaccharide (EPS) production. Herein, the structure of an EPS isolated from sourdough isolate Lactobacillus brevis E25 was determined. A modified BHI medium was used for production of EPS-E25 in order to eliminate potential contaminants. Analysis of sugar monomers in EPS revealed that glucose was the only sugar present. Structural characterisation of EPS by NMR and methylation analysis revealed that E25 produced a highly branched α-glucan with (α1 → 3) and (α1 → 6) glycosidic linkages, and was similar in structure to a previously reported EPS from Lactobacillus reuteri 180. The 1H and 13C NMR data were contrasted with newly recorded data for known polysaccharides (alternan, commercial dextran) which also contain α-(1,3,6)Glc branch points. It was found in both E25 EPS and alternan that NMR parameters could be used to distinguish glucose residues that had the same substitution pattern but occupied different positions in the structure.

In this study, effect of in situ exopolysaccharide (EPS) production on sensory properties of Turkish-type fermented sausage (sucuk) was determined. Sausage mixes were prepared as four groups: (a) Control group, control sausage samples produced without LAB addition, with natural flora, (b) Strain 1 group, sausage samples produced by using Strain 1 (EPS+ Lactobacillus plantarum 162 R strain), (c) Strain 2 group: sausage samples produced by using Strain 2 (EPS+ Leuconostoc mesenteroides N6) and (d) Mixture group, sausage samples produced by using mixture of Strain 1 and Strain 2. These mixes were fermented at 14, 16 and 18 degrees C for 8, 12 and 16 days. Fermentation conditions remarkably affected the sensory properties of the sausage using different EPS producing EPS. These results of this study demonstrated the importance of in situ EPS production on final sensory properties of sausage.

In this work, the role of in situ exopolysaccharide (EPS) production under different fermentation conditions on physicochemical, microbiological, textural and microstructural properties of sucuk was determined. For this purpose, the effect of EPS producing strains (control, strain 1, strain 2 and mixture) and fermentation conditions (fermentation temperature; 14, 16 and 18 °C and time; 8, 12 and 16 days) on physicochemical, microbiological, textural and microstructural properties were investigated using response surface methodology. In situ EPS production was observed to remarkably affect these properties while fermentation conditions were also seen to dominantly influence the physicochemical properties of sucuk, revealing that the ripening temperature appeared to be more determinant factor. EPS producing cultures enhanced the textural properties of sucuk which became harder, less adhesive and tougher. The microstructural analysis revealed the formation of web-like structure by in situ EPS production in sucuk mix during fermentation process. This study revealed the importance of in situ EPS production on final technological properties of sucuk.

Production of exopolysaccharides (EPS) by starter Lactic acid bacteria (LAB) gained special interest due to technological role of these natural polymers. Yogurt samples were collected from different households in Turkey and bacterial strains were isolated and evaluated for EPS production. Strains potentially producing EPS were then identified to species level with 16S rRNA sequencing. An EPS producer Streptecoccus thermophilus strain was identified from yogurt samples. EPS was extracted from culture supernatants of S. thermophilus and partially purified and structural analysis of the crude EPS by FTIR spectroscopy revealed the presence of typical functional groups related to exopolysaccharides. This study explains the identification of a potential starter culture for yogurt production with EPS production.

- MakaleAçık Erişim2015Scopus SJRQ1Impact of the exopolysaccharide layer on biofilms, adhesion and resistance to stress in Lactobacillus johnsonii FI9785
Dertli, Enes; Mayer, Melinda J.; Narbad, Arjan
Background: The bacterial cell surface is a crucial factor in cell-cell and cell-host interactions. Lactobacillus johnsonii FI9785 produces an exopolysaccharide (EPS) layer whose quantity and composition is altered in mutants that harbour genetic changes in their eps gene clusters. We have assessed the effect of changes in EPS production on cell surface characteristics that may affect the ability of L. johnsonii to colonise the poultry host and exclude pathogens. Results: Analysis of physicochemical cell surface characteristics reflected by Zeta potential and adhesion to hexadecane showed that an increase in EPS gave a less negative, more hydrophilic surface and reduced autoaggregation. Autoaggregation was significantly higher in mutants that have reduced EPS, indicating that EPS can mask surface structures responsible for cell-cell interactions. EPS also affected biofilm formation, but here the quantity of EPS produced was not the only determinant. A reduction in EPS production increased bacterial adhesion to chicken gut explants, but made the bacteria less able to survive some stresses. Conclusions: This study showed that manipulation of EPS production in L. johnsonii FI9785 can affect properties which may improve its performance as a competitive exclusion agent, but that positive changes in adhesion may be compromised by a reduction in the ability to survive stress.
