Silver nanoparticles (AgNPs) are known for their significant anticancer and antimicrobial activities in the field of medicine. In the present study, AgNPs were synthesized using a microwave-assisted green synthesis process, in which Ag(I) ions were reduced by organic compounds extracted from the leaves and stems of Phlomis linearis, an endemic plant which acted as both reducing and stabilizing agents. Although various species of the Phlomis genus have been studied in the literature, the synthesis of the nano-sized biogenic silver with reducing Ag(I) in the extract of the endemic plant P. linearis is reported here for the first study time, along with the evaluations of its bioactive properties. The obtained AgNPs were characterized by optical, structural, morphological, etc. techniques such as Ultraviolet–visible spectroscopy (UV–Vis.), Fourier-Transform Infrared Spectroscopy (FT-IR), X-ray diffraction (XRD), scanning electron microscopy (SEM), energy dispersive X-ray (EDX), transmission electron microscopy (TEM), dynamic light scattering (DLS), and photoluminescence (PL). Gas chromatography − mass spectrometry (GC–MS) and FT-IR techniques were used in order to evaluate the biochemical-reduction mechanism. When their antimicrobial activities were determined by the well diffusion method on six bacterial strains, the results showed that P. linearis exhibited antimicrobial activity against all tested bacteria. Additionally, the investigation of the anticancer activity of the particles by using the MTS method on a breast cancer cell line (MCF-7) and a healthy human cell population (human umbilical vein endothelial cells [HUVEC]) revealed that our AgNPs were more cytotoxic toward the cancerous cells compared to the non-cancerous ones (p < 0.001).

    Sait, A.; Bayraktar, B.; Korkmaz, S.; Genç, M.; Bayraktar, S.; Kotan, G. C.; Aydemir, S.; Takma, C.; Bülbül, A. S.; Aksakal, V

Apelin is a hormone with lymphangiogenesis potential, associated with tumor growth and lymph node metastasis, and is reported to stimulate tumor vascularization. This study aimed to investigate the response of the apelin hormone in cattle with and without Enzootic Bovine Leukosis (EBL) infection, based on physiological parameters such as breed, age, and body condition score (BCS). A total of 120 Simmental and Montafon cattle of varying ages (<2, 2-8, and >8) and BCS categories (<2, 3-3.5, and >= 4) were included in the study. Serum apelin hormone levels were measured using the ELISA technique in blood samples collected from the neck vein ( Vena jugularis) of the animals. The analysis of mean serum apelin (ng/mL) values revealed that, among EBL-positive cattle, the highest value was 2.77 ng/mL in Simmental cattle aged 2-8 years with a BCS of 3-3.5, while a value of 2.69 ng/mL was observed in Montafon cattle aged >8 years with a BCS of 3-3.5. For EBL-negative cattle, the highest mean serum apelin value was 2.24 ng/mL, recorded in Simmental cattle aged 2-8 years with a BCS of 3-3.5, whereas it was 1.45 ng/mL in Montafon cattle aged >8 years with the same BCS. In conclusion, this study demonstrated for the first time the effect of apelin hormone levels in cattle with or without EBL, depending on physiological parameters such as age, breed, and BCS. It is suggested that the apelin hormone, which is involved in neoplastic lymphocyte cell infiltration in the lymphoreticular tissues of cattle, and plays a role in regulating tumor growth in EBL, a malignant tumor disease, may contribute to eradication programs. Moreover, the evaluated parameters may prove valuable for disease prevention and control strategies.

Orthorexia nervosa (ON) is a type of eating disorder marked by an excessive preoccupation with eating healthily to preserve and promote health, which surpasses normal levels of concern. This study aims to investigate the correlation between orthorexic tendencies, anthropometric measurements, and several other characteristics among university students. A cross-sectional study was done with a sample size of 307 university students from various departments, consisting of 228 females and 79 males. The data-gathering process consisted of administering a demographic information form and the ORTO-11 questionnaire. Anthropometric measurements, such as weight, height, waist circumference, hip circumference, and chest circumference, were measured. In addition, Body Mass Index (BMI) was calculated. The TANITA BC-418 equipment was used to measure the weight of the participants, while a flexible tape measure was used to measure their height, waist circumference, hip circumference, and chest circumference. The BMI was calculated by the individual's weight (in kilograms) by the square of their height (in meters). The data analysis encompassed the utilization of descriptive statistics, t-tests, variance analysis, and Pearson correlation analysis. A score of 25 or lower on the ORTO-11 scale was categorized as orthorexic, whereas a score higher than 25 was deemed normal. The average age and orthorexia scores of the subjects were 20.81±1.86 and 27.76±4.42 for females, and 21.38±4.14 and 28.11±4.37 for males, respectively. It was determined that 29.3% of the participants were found to display orthorexic tendencies, while the remaining 70.7% did not. There were no statistically significant variations in age, hip circumference, and orthorexia scores across genders (p>0.05). No statistically significant relationship was found between the total orthorexia scores and BMIs of the individuals (r=-.051, p>0.05). ON is associated with psychological and social complications, which in turn contribute to the development of disordered eating patterns. The results of this study indicate that determining the frequency of orthorexic tendencies among university students may contribute to the development of strategies for its prevention.

Effect of Different Rates of Rambutan Peel Extract on Visfatin and Cardiac Troponi N I Response in Type 2 Diabetic Rats Induced with Streptozotocin

    Çelikel Taşci, S.; Bayraktar, B.; Bülbül, A.S.; Sait, A.; Korkmaz, S.

Background: The aim of this study was to investigate the effects of STZ (Streptozotocin) and Rambutan peel extract (RPE) on serum visfatin and cardiac response in experimental Type 2 diabetic rats. Methods: In this study, 64 adult male Wistar albino rats, aged between 8-10 weeks, were used. 8 groups were randomly selected, each containing 8 rats as Control (C), R100 (RPE 100 mg/kg group given with oral gavage route of administration (OGRA), R200 (RPE 200 mg/kg group given with OGRA), R300 (RPE 300 mg/kg group given with OGRA); Diabetes Control (DC): STZ 50 mg/kg i.p administered group, DR100 (D+100 mg/kg JPE), DR200 (D+200 mg/kg JPE), DR300 (D+300 mg/kg JPE). The study lasted a total of 31 days, including adaptation (7 days), induction of diabetes (3 days) and trial period (21 days). Blood samples were taken from the tail vein (vena caudalis) of all subjects on days 0 and 21 of the study. Visfatin and cTnI levels in the serum samples were measured and evaluated by ELISA method. Result: While the increase in mean serum visfatin and cTnI levels in diabetic groups was seen mostly in DC groups, the most significant decrease in mean serum visfatin level due to the addition of RPE was determined in DR100 groups (p<0.01). As a result, it was concluded that the administration of 100 mg/kg RPE in rats had no adverse effects. It was concluded that RPE, which has a cardioprotective effect by regulating the visfatin response that plays a role in the development of diabetes, may be useful in supporting the impaired cardiovascular system function that occurs in diabetes.

Taş, Recep; Köroğlu, Ebru; Karakuş, Ahmet; Bülbül, Ali Savaş; Taş, Nilay Akkuş

Metal nanoparticles that are widely studied in optoelectronics, catalysis, medicine, and sensors offer remarkable optical and electronic properties. To address the cost and environmental concerns associated with their synthesis, this study employs an environmentally friendly method using Laureus nobilis extract to produce silver and zinc nanoparticles, which are prominent in nanotechnology. This study includes investigations of factors such as reaction time, AgNO3/laurel ratio, Zn(Ac)2 H2O/laurel ratio and temperature in nanoparticle biosynthesis to optimize the process. The next stage was set to evaluate the photocatalytic performance of these nanoparticles, specifically against the methylene blue dye under dark and UV light conditions. Parameters such as pollutant decomposition, degradation rate, catalyst stability, and nanoparticle recovery were analysed. Structural characterization of the obtained nanoparticles was performed using UV-Vis, FTIR, SEM, and XRD techniques. The photocatalytic results showed significant degradation percentages for LB-AgNP (silver nanoparticles synthesized with Laureus nobilis extract) (97.5%) and LB-ZnNP (zinc nanoparticles synthesized with Laureus nobilis extract) (90.9%). LB-ZnNP showed superior performance. Therefore, LB-AgNP and LB-ZnNP are promising photocatalysts for water purification and the elimination of toxic organic pollutants.

Green synthesis of (S)-1-(furan-2-yl)propan-1-ol from asymmetric bioreduction of 1-(furan-2-yl)propan-1-one using whole-cell of Lactobacillus paracasei BD101

    Bülbül, Ali Savaş; Şahin, Engin

Chiral heterocyclic alcohols are important precursors for production of pharmaceutical medicines and natural products. (S)-1-(furan-2-yl)propan-1-ol ((S)-2) can be used production of pyranone, which can be used in the synthesis of sugar analogues, antibiotics, tirantamycines, and anticancer drugs. The synthetic approaches for (S)-2, however, have substantial difficulties in terms of inadequate enantiomeric excess (ee) and gram scale synthesis. Moreover, the biocatalytic synthesis of (S)-2 is unknown until now. In this study, the synthesis of (S)-2 was carried out by performing the asymmetric bioreduction of 1-(furan-2-yl)propan-1-one (1) using the Lactobacillus paracasei BD101 biocatalyst obtained from boza, a grain-based fermented beverage. (S)-2 was obtained with >99% conversion, >99% ee, and 96% yield under the optimized conditions. Furthermore, in 50 h, 8.37 g of 1 was entirely transformed into (S)-2 on gram scale (96% isolated yield, 8.11 g). This is the first report on the high-gram scale biocatalyzed synthesis of enantiopure (S)-2. These data suggest that L. paracasei BD101 can be used to bioreduction of 1 in gram scale and efficiently produce (S)-2. Furthermore, these findings laid the base for future study into the biocatalytic production of (S)-2. It was particularly notable as it was the highest known to date optical purity of (S)-2 generated by asymmetric reduction using a biocatalyst. This work offers a productive environmentally friendly method for producing (S)-2 using biocatalysts.

Bülbül, Ali Savaş; Sen, Fatih; Bekmezci, Muhammed; Karakaya, Funda

In this study, R. aculeatus plant extract and biogenically formed AgNPs were investigated for their potential antibacterial, antibiofilm and anticancer abilities. AgNPs were characterised using scanning electron microscopy (SEM), Fourier transform infrared spectroscopy (FT-IR), X-ray diffraction (XRD) and UV-vis spectroscopy (UV-VIS). According to the Debye Scherrer formula, the particle size was found to be 32.56 nm. Disc diffusion and microdilution methods were used to investigate the antibacterial activity. In the disc diffusion study, the best results were obtained from the extract and AgNP. In the tests using plant extracts, Staphylococcus aureus ATCC 25923 showed the lowest antibiofilm activity, while Bacillus subtilis and Enterobacter aerogenes ATCC 13048 showed the highest activity. Salmonella infantis was most affected by AgNP, while Escherichia coli CFAI ATCC 25922 was least affected. Biogenically synthesised AgNPs were also investigated in cytotoxic activity studies. It was found to have the lowest concentration value on MCF-7 and HUVEC cell lines at the determined concentrations. The extract did not have any cytotoxic effect on MCF-7 cell line. HUVEC cell line showed the lowest cytotoxic activity of 10⁻³ g/mL. The antibacterial, antibiofilm and anticancer properties of R. aculeatus plant extract and biogenically produced AgNPs have been the subject of an important study. Furthermore, the comparison of the effects of plant extract and AgNPs on breast cancer cell lines and healthy cell lines provides a rich scientific material.
