The study aims to investigate the relationship between relative telomere lengths and various physiological parameters in cattle infected with bovine leukosis virus, as well as to identify the risk factors associated with cow-level bovine leukosis virus infection in dairy cattle in Türkiye. Three variables—age, parity, and body condition score—were used for risk assessment. Antibodies specific to bovine leukosis virus were detected using an enzyme-linked immunosorbent assay, and relative telomere lengths were measured by polymerase chain reaction. The highest seropositivity rate (39.13%) was observed in cattle with a body condition score of 3–3.5 and aged between 2–8 years. relative telomere length was found to be positively correlated with seropositivity, advanced age, and parity, while a negative correlation was observed between seropositivity and body condition score. Relative telomere lengths in bovine leukosis virus-infected cattle were significantly shorter (P < 0.05) than those in non-infected cattle. Among body condition score groups, the shortest relative telomere lengths were found in bovine leukosis virus-infected cattle with body condition score ≤ 2.5 and in non-infected cattle with body condition score 3–3.5. Regarding age groups, the shortest relative telomere lengths were observed in bovine leukosis virus-infected cattle older than 8 years (P < 0.05), as well as in non-infected cattle of the same age group (P < 0.05). In conclusion, relative telomere lengths in bovine leukosis virus-infected cattle were found to be significantly shorter than in non-infected cattle (P < 0.05). The shortest relative telomere lengths were recorded in both bovine leukosis virus-infected and non-infected cattle over 8 years of age (P < 0.05). Finally, the association between bovine leukosis virus infection and relative telomere lengths, along with other factors such as age, parity, and body condition score, is considered potentially useful for disease control and prevention programs.

Virucidal and antiviral activities of maca (Lepidium meyenii) root powder on three pathways of Norovirus as a foodborne pathogen; [Actividades viricidas y antivirales del polvo de raíz de maca (Lepidium meyenii) en tres vías del norovirus como patógeno transmitido por los alimentos]

Vitamin C equivalency, the cytotoxicity on murine macrophage cells, and the virucidal and antiviral activities of maca (Lepidium meyenii) powder on murine norovirus (MNV) as a norovirus surrogate for human as a foodborne pathogen were studied. The antioxidant capacity of the maca was determined by MTT assay as vitamin C equivalency (VCEAC). The 50% cytotoxic concentration (CC50) was calculated with the linearity of maca concentrations-cell viability. The activities of maca against the virus was determined as the 50% tissue culture infective dose with Spearman–Karber method. The VCEAC of maca linearly increased with the concentration. The non-cytotoxic concentrations of maca were measured on murine macrophage with a CC50 of 5.77 mg/mL and a CC80 of 1.28 mg/mL. As maca reduced the virus titration based on contact time in the virucidal assay in several conditions, it did not significantly inhibit MNV on three infectivity pathways of host cell at non-cytotoxic concentrations of maca. The result of this first study presented that despite its ineffective in the treatment and prophylaxis of norovirus infection, maca might be an alternative food additive and preservative against human norovirus as an important foodborne pathogen causing outbreaks worldwide. © Los autores. Objective: The aim of this study is to evaluate the effectiveness of three commercial nucleic acid extraction kits (kit A, B and C) in isolating SARS-CoV-2 viral RNA from wastewater samples. Method: In this study, water samples were collected in March 2021 from three wastewater treatment plants located in different parts of Istanbul, and it was confirmed that they were negative for SARS-CoV-2. Different concentrations of the SARS-CoV-2 virus, previously inactivated at the BSL-3 laboratory of the Pendik Veterinary Control Institute, were added to the wastewater samples. RNA extraction and quantification were performed using commercial nucleic acid extraction kits and and RT-qPCR kit specific to SARS-CoV-2. Results: At the end of the study, it was determined that kit C yielded the highest total RNA and produced more consistent results, significantly outperforming the other two kits in terms of RNA yield and purity. Statistical analysis revealed significant differences in RNA concentrations (p < 0.05) and gene copy numbers (p < 0.01) between the kits, and kit C demonstrated superior linearity and reproducibility. Conclusion: According to the findings, although all three evaluated kits are suitable for detecting SARS-CoV-2 RNA in wastewater samples, kit C provides the most efficient and reliable performance, especially for high-throughput studies. Additionally, this study highlights the importance of selecting appropriate nucleic acid extraction methods for wastewater surveillance, which serves as an early warning system for outbreaks that threaten public health.

A chronic microbial infection that is mediated by the host's immune system is the cause of periodontal disease. As the world's population grows, phytotherapy is a therapeutic strategy that makes use of a renewable resource that can provide less costly medicines. This study aimed to investigate the therapeutic effects of eugenol administration on periodontitis by examining the effects of eugenol at different doses biochemically, histopathologically, and radiologically in rats with experimental periodontitis. Thirty-two Sprague Dawley male rats were divided into four groups: 1. the healthy control group (CG), 2. The experimental periodontitis group (PG), 3. The experimental periodontitis+10 mg/kg eugenol group (EG10), and 4. the experimental periodontitis+20 mg/kg eugenol group (EG20). Alveolar bone loss was measured using micro-CT imaging. The TNF-α, IL-6, leptin, asprosin, and apelin levels were analyzed in the serum samples of rats. Histological and immunohistochemical analyses were performed. The data were analyzed statistically. The serum TNF-α, IL-6, leptin, asprosin, and apelin levels were found to be statistically significantly lower in the eugenol-applied groups (EG10 and EG20) than in the PG (p < 0.05). There was a statistically significant difference in the TNF-α, IL-6, leptin, asprosin, and apelin levels between the EG10 and EG20 (p < 0.05). In the measurements made using micro-CT imaging, alveolar bone loss was found to be at the lowest FR1 level, and in histopathological analysis, the least inflammation was observed in the EG20. It was concluded that the application of 20 mg/kg eugenol in rats with experimental periodontitis was a safe dose, was beneficial in alleviating bone loss, and had a positive effect on the healing process.

Investigation of the recovery efficiency of CeUF method through RT-qPCR quantification of inactivated SARS-CoV-2 in untreated wastewater

Sait, Ahmet; Korkmaz, Serol; Parmaksiz, Ayse; Bayraktar, Bulent

Wastewater-based disease monitoring is an early warning system and a surveillance tool for infectious disease outbreaks regarding pathogens with pandemic potential. This study aimed at investigating the recovery efficiency of centrifugal ultrafiltration (CeUF), which is one of the most-used virus concentration methods, for inactive severe acute respiratory syndrome coronavi-rus 2 (SARS-CoV-2) added to wastewater. Inactivated SARS-CoV-2 was inoculated into untreated wastewater at different concentrations (4 × 103, 8 × 103, 16 × 103, 24 × 103 and 32 × 103 gene copy/ µL) and concentrated through ultrafiltration with a disposable centrifugal filter device. Total nucleic acids in concentrated filtrates were extracted and isolated by an automated system. In isolates, total RNA was measured by a UV/VIS spectrophotometer, and the recovered virus was quanti-fied by RT-qPCR with two gene regions (N1 and N2). The recovery rates were between 11% and 17.8% (mean 15.1%, CV below 15%). While there were positive correlations among the inoculated virus, total RNA and recovered virus, there was no correlation and linearity between the recovery rates. Despite limited recovery rates, CeUF integrated with RT-qPCR quantification can be a valid assay for monitoring SARS-CoV-2 in wastewater, and an early warning system.
