In this study, the utilization of LAB (Lactiplantibacillus plantarum, Levilactobacillus brevis, Furfurilactobacillus rossiae, Weissella cibaria) cultures with selected flours of different immature cereal grains (wheat, barley, oat and rye) on sourdough and sourdough bread was investigated. In total, 16 different sourdough breads were produced and microbiological, pH, TTA and rheological properties of sourdoughs at 0, 8 and 24 h were determined. At the beginning of the fermentation, while sourdoughs showed elasticity, they showed viscoelastic properties after 24 h of fermentation. The sugar profile of cereal grains at the stage of milk formation and the sugar groups presented in EPSs isolated from sourdoughs were revealed by HPLC analysis. The sugar groups in sourdoughs were glucose, xylose and arabinose, while the sugars in the immature cereal grains were glucose, fructose and mannose. The highest EPS production level (1882.69 ± 8.16) was observed in immature barley flour and Lpb. plantarum containing sourdough and all sourdoughs were found to contain glucan type EPSs. It was observed that the amount of EPS produced by the related species differed significantly under different grain conditions. Besides, it was observed that the sourdough breads produced were harder than the control bread. Novelty Impact Statement: Wheat, barley, oat and rye immature grains and four distinct LAB strains were used during sourdough fermentation. Immature grain type and LAB strain used were determinants for in situ exopolysaccharide (EPS) production. Viscoelastic properties of sourdough and textural properties of sourdough bread were affected by grains and LAB strain utilized.

In this study, structural and techno-functional characteristics of an exopolysaccharide (EPS) produced by Lactobacillus kunkeei AK1 were determined. High-performance liquid chromatography (HPLC) analysis demonstrated that EPS AK1 was composed of only glucose units. 1H and 13C Nuclear magnetic resonance (NMR) analysis revealed that EPS AK1 was a dextran type EPS containing 4.78% (1 → 4)-linked α-D-glucose branches. The molecular weight of EPS AK1 was determined to be 45 kDa by Gel Permeation Chromatography (GPC) analysis. A high level of thermal stability up to 280 °C was determined for dextran AK1 detected by Differential scanning calorimetry (DSC) and Thermogravimetric analysis (TGA). Dextran AK1 appeared as regular spheres with compact morphology and as irregular particles in the solution with no clear cross-linking between the chains of the polysaccharide observed by Scanning electron microscopy (SEM) and Atomic force microscopy (AFM) analysis, respectively. X-ray diffraction analysis (XRD) analysis demonstrated that dextran AK1 had a crystalline structure. A relatively strong antioxidant activity was observed for dextran AK1 determined by 2,2-diphenyl-1-picrylhydrazyl (DPPH), 2,2′-azino-bis-3-ethylbenzothiazoline-6-sulfonic acid (ABTS) radical scavenging and cupric reducing antioxidant capacity (CUPRAC) tests. Finally, only a digestion ratio of 3.1% was observed for dextran AK1 following the in vitro simulated gastric digestion test.

Recently, isolation of Lactic Acid Bacteria (LAB) strains with important technological functions from plant-based products has gained special interests related with the increment of consumer preferences to the plant-based fermented products. In this context, various cereals and legumes that were underwent to germination or not were selected for the isolation of exopolysaccharide (EPS) producer LAB strains in this study. Following genotypic discrimination, 17 distinct strains belonging to Weissella confusa, Weissella cibaria, Lactococcus lactis subsp. hordniae, Leuconostoc lactis, Lactiplantibacillus plantarum and Leuconostoc mesenteroides were identified from pre-ferment liquids. Two strains Weissella confusa W-16 and Leuconostoc lactis GW-6 were the highest EPS producers, and their EPSs were further characterized. The 1H and 13C NMR results revealed that both strains were dextran producers containing (1 → 3)-linked α-D-glucose units as branches with 9.2% and 6.9% levels for Weissella confusa W-16 and Leuconostoc lactis GW-6, respectively. FTIR analysis also supported the dextran structure and demonstrated the functional groups within the dextrans. A high level of thermal stability was observed in both dextrans detected by TGA and DSC analysis. Both dextrans were in amorphous characteristics determined by XRD analysis. These findings demonstrate that pre-fermented liquids of different cereals and legumes can be source of technologically important LAB strains with EPS production characteristics.

İspirli, Hümeyra; Dertli, Enes
Bioactive oligosaccharides are important compounds for development of functional foods and ingredients. In this study, lactose derivative hetero-oligosaccharides were produced by acceptor reaction of glucansucrase E81 with whey. The synthesis of oligosaccharides was monitored by Thin Layer Chromatography (TLC) analysis followed by 1H and 13C Nuclear Magnetic Resonance (NMR) spectroscopy analysis which demonstrated the glycosylation of lactose with α-(1 → 2), α-(1 → 3) and α-(1 → 4) linkages. Liquid chromatography–mass spectrometry (LC-MS) analysis demonstrated the formation of DP 7 oligosaccharides from whey. The synthesized oligosaccharides were not digested under in vitro human gastric juice and α-amylase conditions. The prebiotic effect of lactose derivative hetero-oligosaccharides was tested and no growth in pathogenic strains was observed whereas certain probiotic strains utilised lactose derivative hetero-oligosaccharides as Carbon source and their growth was promoted. These findings can be important to reveal the potential of glucansucrases to produce functional oligosaccharides from different sources including waste materials.

Glucansucrases can perform acceptor reactions with different sugars to produce distinct oligosaccharides which can potentially act as prebiotics depending on their characteristics. Using cheap substrate sucrose and under mild and environmentally friendly conditions, glucansucrase-derived oligosaccharides can be produced with high yields. In this study, raffinose-derived oligosaccharides up to degree of polymerization (DP) 7 produced by glucansucrase E81 were characterized and their prebiotic functions were evaluated. The main product was a DP 4 oligosaccharide containing two products formed by the transfer of the glucose to raffinose at either the 3- (81%) or 4-O (19%) position of its galactose residue confirmed by NMR spectroscopy and LC-MS-LTQ analysis. The purified DP 4 raffinose-derived oligosaccharides were subjected to in vitro human gastric fluid and α-amylase digestion tests and no digestion was observed under both cases. Finally, prebiotic potential of DP 4 raffinose-derived oligosaccharides was tested with probiotic and pathogenic strains and only growth of Bifidobacteria promoted, thus supported the Bifidogenic activity of DP 4 raffinose-derived oligosaccharides.

Cellobiose-derived oligosaccharides were produced from acceptor reactions with glucansucrase E81 and their prebiotic potentials were determined in this study. The major product was a mixture of trisaccharides formed by the addition of an α-linked glucose to cellobiose at either the 2-O position of the reducing end (86%) glucose or the 6-O position of the non-reducing end glucose (11%) demonstrated by NMR spectroscopy and LC-MS-LTQ analysis. The purified DP 3 cellobiose-derived oligosaccharides were then tested for in vitro human gastric juice and α-amylase digestion and no digestion was observed during the 6 h digestion period. Importantly, DP 3 cellobiose-derived oligosaccharides promoted the growth of Bifidobacteria at high levels demonstrating a Bifidogenic effect and a mild level of proliferation was observed for Lactobacilli suggesting the prebiotic roles of cellobiose-derived oligosaccharides.

An exopolysaccharide (EPS) producer slimy-mucoid type colony was isolated from sourdough and identified as Weissella cibaria MED17. The 1H and 13C NMR spectra of EPS MED17 demonstrated that this EPS was a dextran type glucan ((1 → 6)-linked α-D-glucose core structure) containing (1 → 3)-linked α-D-glucose branches and proportion of (1 → 6)-linked α-D-glucose units to (1 → 3)-linked α-D-glucose units was 94.3:5.7%. The FTIR analysis also confirmed the (1 → 6)-linked α-D-glucose linkage. A high level of thermal stability was observed for glucan MED17 as no degradation up to 300 °C was observed by TGA and DSC analysis. The XRD analysis of glucan MED17 showed its semi- crystalline nature and its compact sheet-like morphology was observed by SEM analysis. Finally, antioxidant characteristics of glucan MED17 were determined by ABTS and DPPH radical scavenging activity tests that revealed a moderate antioxidant activity of glucan MED17. These findings show potential techno-functional characteristics of glucan MED17.

Oligosaccharides are one of the functional ingredients to be used in food technology. In this study, by using an active glucansucrase GTFA-ΔN E81 in the acceptor reaction of mannose, mannose-containing oligosaccharides were produced and their functionality was tested. The formation of the oligosaccharides were visualised by TLC analysis and mannose-containing oligosaccharides up to DP 7 were determined by LC-MS analysis. The presence of the (1,6)Glc and (1,3)Glc units within the oligosaccharides were determined by NMR analysis. The in vitro immune-modulatory functions of the mannose-containing oligosaccharides were determined but no induction in IL-4, IL-10, IL-12 and TNF-α cytokine levels were detected. Importantly this oligosaccharide mixture showed prebiotic effect by triggering the growth of tested probiotics and not affecting the growth of pathogen strains. Our findings reveals the potential of the role of glucansucrases for the production of functional oligosaccharides.

In this study, technological role of α-glucan with (α1-3) and (α1-6) glycosidic linkages produced by Lactobacillus reuteri E81 on sourdough rheology and bread texture was investigated. To achieve this, a glucan deficient ΔgtfA mutant was produced by the deletion of the glucansucrase gene from strain E81. By comparison of glucan-positive and glucan-negative strains for rheological properties of sourdough revealed that in situ α-glucan production resulted in higher elasticity in sourdough. However, no additional effect of in situ glucan on textural characteristics of bread was observed although addition of glucan ex situ at 0.09% level significantly decreased bread hardness. Scanning Electron Microscopy (SEM) images of lyophilized sourdoughs confirmed association of glucan and starch granules forming a film like structure in sourdough.

Glucansucrases are responsible for the production of α-glucans using sucrose as the substrate. Glucansucrases may also produce different oligosaccharides by transferring the glucose moiety from sucrose to a variety carbohydrates acting as acceptor nucleophile. In this study, melibiose-derived oligosaccharides were produced by the glucansucrase from Lactobacillus reuteri E81 expressed without the N-terminal region (GtfA-ΔN). The reaction products were characterized by TLC, LC-MS and NMR analysis and it was found that GtfA-ΔN synthesized melibiose-derived oligosaccharides (DP3 and DP4) by adding glucose units through alpha 1->3 or 1->6 glycosidic bond. The functional characteristics of these melibiose-derived oligosaccharides were determined by testing the immune-modulatory functions in HT-29 cells and testing their growth promoting effects for important probiotic and pathogenic strains. The melibiose-derived oligosaccharides triggered the production of anti-inflammatory cytokine IL-10 and pro-inflammatory cytokine TNF-α depending on their concentrations. Finally, melibiose-derived oligosaccharides showed bifidogenic effect as potential prebiotics.

- MakaleAçık Erişim2020Scopus SJRQ3Determining the optimum model parameters for oligosaccharide production efficiency using response surface integrated particle swarm optimization method: an experimental validation study
Şensoy, Abdullah Tahir; İspirli, Hümeyra; Dertli, Enes
Glucansucrases (GTFs) catalyzes the synthesis of α-glucans from sucrose and oligosaccharides in the presence of an acceptor sugar by transferring glucosyl units to the acceptor molecule with different linkages. The acceptor reactions can be affected by several parameters and this study aimed to determine the optimal reaction parameters for the production of glucansucrase-based oligosaccharides using sucrose and maltose as the donor and acceptor sugars, respectively via a hybrid technique of Response Surface Method (RSM) and Particle Swarm Optimization (PSO). The experimental design was performed using Central Composite Design and the tested parameters were enzyme concentration, acceptor:donor ratio and the reaction period. The optimization studies showed that enzyme concentration was the most effective parameter for the final oligosaccharides yields. The optimal values of the significant parameters determined for enzyme concentration and acceptor:donor ratio were 3.45 U and 0.62, respectively. Even the response surface plots for input parameters verified the PSO results, an experimental validation study was performed for the reverification. The experimental verification results obtained were also consistent with the PSO results. These findings will help our understanding in the role of different parameters for the production of oligosaccharides in the acceptor reactions of GTFs.

Optimisation of exopolysaccharides (EPS) production in Lactic Acid Bacteria (LAB) is an important task as EPS production can be affected by different parameters. In this respect, this study aimed to characterise the structure of an EPS from Leuconstoc mesenteroides DL1 strain and to optimise the EPS production by determination of the effects of incubation time, sucrose concentration, incubation temperature and initial levan concentration (input parameters) using integrated ANNs (Artificial neural networks) and fuzzy modelling approaches. The characterisation of the EPS monomeric composition by HPLC analysis revealed that EPS DL1 was composed of glucose and fructose. The 1H and 13C NMR spectra of EPS DL1 also confirmed the glucan and fructan production. The effects of the input parameters on glucan and fructan production levels as output parameters by DL1 were optimised using neural network and fuzzy modelling tools. The fuzzy model was developed based on the recognition of basic elements of input-output parameters, and the power of ANNs used for system identification. A structural analysis was carried out to improve the flexibility of fuzzy model, and to design the unknown mappings of the input and output parameters more robustly. The parameters then were fine-tuned by qualitative reasoning to establish the relations of input output parameters using membership functions (MFs) and their intervals determination. A hybrid training algorithm was employed for parameter identification, MFs and their interval determination to obtain the fuzzy model. The model can predict the outcome parameters; glucan and fructan with high accuracy for the predetermined input parameters.

İspirli, Hümeyra; Şimşek, Ömer; Skory, Christopher; Sağdıç, Osman; Dertli, Enes
A wide number of Lactic Acid Bacteria (LAB) species produce α-glucans with their ability to synthesize glucansucrases (GS) which use sucrose as substrate for the glucan production. Recently another group of enzymes in LAB gained special interest for their ability to produce α-glucans targeting the substrates containing α1-4-linkages and synthesizing new (α1-6) or (α1-3)–linkages as α‑glucanotransferases. In this study, a putative 4,6‑α‑glucanotransferase (GTFB) from sourdough isolate Lactobacillus reuteri E81 was identified and expressed in Escherichia coli. The biochemical characterization of the GTFB-E81 confirmed its function as it cleaved the α1-4-linkages in different substrates and produced new gluco-oligomers/polymers containing α1-6 linkages together with the α1-4-linkages detected by NMR analysis. GTFB-E81 produced malto-oligosaccharides targeting maltose and maltoheptaose as substrates with up to DP 8 detected by TLC and ESI-MS/MS analysis. The functional roles of these malto-oligosaccharides were determined by testing their immune-modulatory functions in HT29 cells and they triggered the production of anti-inflammatory 1L-4 and pro-inflammatory IL-12 cytokines.

Glucansucrases, which can be produced by different Lactic Acid Bacteria (LAB), catalyze the synthesis of α-glucans with different structures and properties using sucrose as substrate. In this study, a novel glucansucrase (GTFA) from Lactobacillus reuteri E81 was identified and heterologously expressed. Alignments of GTFA with other glucansucrases revealed its novelty and a putative 3D model structure was obtained. The biochemical properties of the truncated enzyme without the N-terminal variable region, GTFA-ΔN, was characterized. The Km and Vmax were found to be 7.5 mM and 1.49 IU/mg, respectively, and it showed optimum activities at pH 7 and at 50 °C. The GTFA-ΔN produced in vitro an α-glucan with (α1 → 3) and (α1 → 6) glycosidic linkages using sucrose as the substrate. Importantly, GTFA-ΔN synthesized DP = 9 oligosaccharides using sucrose and maltose as the donor and acceptor sugars, respectively, as detected by TLC, HPLC, LC-MS and NMR analysis.
