Electrospinning Encapsulation of Chlorophylls Microwave-Extracted from Spinach Waste and Utilization as Coloring Agent in Yogurt

Isik, Beyza Sukran; Bayram, Sinan; Altay, Filiz

Natural colorants have become a developing market because of consumer preferences, and more cost-effective and stable production is necessary. In the present study, a natural green colorant was obtained via microwave-assisted extraction from spinach waste to produce affordable, environmentally-friendly, and innovative colorants. The extracts were encapsulated with zein via uniaxial and coaxial electrospinning techniques to overcome the stability issues associated with their natural green color. The encapsulation efficiencies of uniaxially encapsulated chlorophyll extracted from spinach waste by microwave were 63.50 and 57.18 for chlorophyll a and b, respectively. The efficiencies of chlorophyll a and b by coaxial electrospinning were found 49.53% and 88.58%, respectively. The microwave-assisted extraction and the addition of eumelanin may contribute positively to the antioxidant activity and thermal stability of electrospun samples. The stability test against light revealed that 61% of chlorophyll a could be preserved by uniaxial electrospinning. In addition, when chlorophyll-loaded nanofibers were incorporated into yogurt, a more stable green color was achieved during storage.

Karakas, Ümıt; Bayram, Sinan; Kizil, Hamit Emre; Öztürk, Serdar

Melanins, which are divided into five different subgroups as eumelanin, pheomelanin, allomelanin, neuromelanin and pyomelanin, are a heterogeneous and amor- phous biopolymer family. Although melanin is produced in many different ways today, it is more practical, and more economical to produce melanin using bacterial strains. The eumelanin pigment, which is the subject of this study, was obtained using Streptomyces parvus BSB49 strain, and its antiproliferative effect at various concentrations against small cell (DMS-114) and non-small cell (H-460) lung cancer was determined. According to our results, the synthesized eumelanin molecule was statistically cytotoxic on non-small cell lung cancer cell line (H-460) at concentrations of 3.125 and 1.56 μg/mL after 24 hours of treatment (IC50=8 μg/mL) and only at a concentration of 3.125 μg/mL after 48 hours of treatment (IC50=8.4 μg/mL). In the small cell lung cancer cell line (DMS-114), no statistically cytotoxic concentration was detected (p<0.05).

Kantar, Naciye Kutlu; Bayram, Sinan; Çelik, Saffet; Ecem Bayram, Nesrin; Gerçek, Yusuf Can; Baştürk, Fatma Nur; Yildirim, Nazlican

The role of eumelanin as a natural pigment in protecting human skin from ultraviolet (UV) light has drawn vast interest in the research and industrial community. Encapsulation of the compound by various shell materials has been extensively studied to optimize and prolong its shielding efficiency from UV penetration through the skin. Polylactide (PLA)-based copolymers have been widely used in the encapsulation of various active compounds due to their biocompatibility and biodegradability that facilitate sustained release of the active compounds. In this work, stereocomplex PLA (sc-PLA) derived from mixtures of poly(D-lactide-caprolactone-D-lactide), P(DLA-b-CL-b-DLA), a triblock copolymer with linear poly(L-lactide), and PLLA are employed to encapsulate eumelanin by an oil-in-water emulsion (O/W) technique. The effect of eumelanin distribution in PLA's enantiomers and ultrasonication on the physicochemical properties, encapsulation efficiency, and release behavior of the nano/microparticles were evaluated. The potential application of the resulting particles for sunscreen products was assessed in terms of UV absorbance and in vitro sun protection factor (SPF). The nano/microparticles show a hollow spherical structure, whose size can be controlled by ultrasonication. The distribution of eumelanin and the ultrasonication process play a key role in the growth of sc-PLA and the crystalline structure of the particles. The highest encapsulation efficiency of 46.6% was achieved for sc-PLA2U particles. The high content of eumelanin and the hollow structure with a large surface area lead to improvement in the UV absorbance and sunscreen performance of the particles, as revealed by the increase in the SPF value from 9.7 to 16.5. The materials show high potential for various applications, especially in cosmetic and pharmaceutical fields, as UV-shielding products.

Contribution of Oxidative Stress, Apoptosis, Endoplasmic Reticulum Stress and Autophagy Pathways to the Ameliorative Effects of Hesperidin in NaF-Induced Testicular Toxicity

Emre Kızıl, Hamit; Gür, Cihan; Ayna, Adnan; Darendelioğlu, Ekrem; Küçükler, Sefa; Sağ, Sevda

The ameliorative effects of hesperidin (HES) on the toxicities created by sodium fluoride (NaF) in the testes tissue of rats were studied via oxidative stress, apoptosis and endoplasmic reticulum (ER) stress pathways. The animals were divided into five distinct groups (7 rats in each group). Group 1 was control group, group 2 received NaF-only (600 ppm), group 3 received HES-only (200 mg/kg bw); group 4 received NaF (600 ppm)+HES (100 mg/kg bw) and group 5 received NaF (600 ppm)+HES (200 mg/kg bw) for 14 days. NaF-induced testes tissue damage by reducing activities of superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GPx) and levels of glutathione (GSH), and increasing lipid peroxidation levels. NaF treatment significantly downregulated the mRNA levels of SOD1, CAT and GPx. NaF supplementation caused apoptosis in the testes by upregulating p53, NFkB, caspase-3, caspase-6, caspase-9, and Bax and downregulating Bcl-2. Furthermore, NaF caused ER stress via increasing mRNA transcript levels of PERK, IRE1, ATF-6 and GRP78. NaF treatment led to autophagy via upregulation of Beclin1, LC3A, LC3B and AKT2. In testes tissue, however, co-treatment with HES at doses of 100 and 200 mg/kg significantly reduced oxidative stress, apoptosis, autophagy and ER stress. Overall, the findings of this study suggest that HES may help to reduce testes damage caused by NaF toxicity.

Pseudomonas aeruginosa and Pyoverdines: A Hidden Threat in Apitherapy Applications

    Bayram, Sinan

Pseudomonas aeruginosa, a Gram-negative rod-shape bacterium, is one of the most common pathogens in nosocomial infections and this pathogenic species draws attention with its high prevalence rate in patients with septicemia. Pyoverdines are fluorescent siderophore produced by some Pseudomonas species such as P. aeruginosa and Pseudomonas fluorescens. These siderophores support biofilm production in these microorganisms and also plays a role as a virulence factor. In this study, anti-Pseudomonas activity of 10 different honey samples was evaluated by agar well diffusion (AWD) method. In addition, minimum inhibition concentration (MIC) and minimum bactericidal concentration (MBC) values of these honey samples were determined via microbroth dilution method. The obtained results showed that only oak honey and chestnut honey have negligible scale low anti-Pseudomonas activity. In addition, it was observed that other honey samples did not have an inhibitory effect against these pathogens. Finally, the promoting property of pyoverdine siderophore production of these honey samples was evaluated and it was concluded that all honey samples stimulated pyoverdin production at a concentration ½ w/v.

Searching for Versatile Polysaccharide-Degrading Alkali-tolerant or Alkaliphilic Bacillus Strains

    Aydogan, Mehmet Nuri; Bayram, Sinan

The plant-derived polysaccharides (Cellulose, xylose and amylose, etc.) are the most abundant renewable raw materials in nature. Cellulose and xylose are the predominant carbohydrate polymer components of the plant cell walls and the most abundant biopolymers in the world. Another plant-derived polysaccharide, starch is found in plant tubers, roots and seed endosperms as a major carbohydrate reserve. In this study, it was aimed to find multi-enzyme producer bacteria strains in terms of industrially important enzymes such as cellulase, xylanase and amylase. For this purpose, isolated Bacillus strains from different samples were qualitatively evaluated for cellulase, xylanase and amylase enzyme production potentials. The isolates that have the highest enzyme activity were selected for biochemical tests, molecular and phenotypic characterization. As a result of these characterization process, SB57, SB104, SB155, SB178, SB197 and SB199 strains were identified as Bacillus pumilus and SB118, SB138 strains were identified as Bacillus safensis. In addition to these strains, SB120 and SB147 strains were identified as Bacillus aerius and Bacillus licheniformis respectively. 16S rDNA sequence analysis results of these Bacillus strains were deposited in NCBI GenBank® under accession number KT371465 - KT371474 respectively.

Ultrasonicate-assisted preparation of eumelanin-loaded nano/microparticles based on polylactide stereocomplex

Putri, Oceu Dwi; Petchsuk, Atitsa; Bayram, Sinan; Opaprakasit, Pakorn

The utilization of ultrasonication in the preparation of polymeric nano/microparticles has attracted vast attention due to its simplicity with low cost and high energy efficiency. The occurrence of acoustic cavitation generated by the ultrasonication process can enhance droplet disintegration, allowing the improved stability of emulsions. In this study, eumelanin-loaded nano/microparticles were prepared by employing polylactide stereocomplex (sc-PLA) formed by blending of poly(D-lactide-caprolactone-D-lactide), P(DLA-b-CL-b-DLA), triblock copolymer with linear poly(L-lactide), PLLA. The particles were prepared through an oil in water emulsion (O/W) technique followed by a solvent evaporation. The effect of ultrasonication on the encapsulation efficiency and physicochemical properties of the nano/microparticles was evaluated. The results showed that ultrasonication treatment could control nano/microparticles' shape, size, and size distribution, which enhanced the particle stability and the entrapment efficiency of eumelanin. Particle size distribution of ultrasound-assisted nano/microparticles was in a range of 0.38–135 μm, while those without ultrasonication had a broader distribution from 0.38–153 μm. Higher zeta potential was also observed in the ultrasound-assisted nano/microparticles, reflecting their higher suspension stability. The ultrasonication treatment led to higher eumelanin encapsulation efficiency (46.6%) with higher thermal stability and crystallinity, promoted by the formation of higher contents of the stereocomplex crystallites. The results confirm that ultrasonicate-assisted emulsification can be effectively used to fabricate the functional eumelanin-loaded nano/microparticles.
