Kırbaş, Zila Özlem; Bayraktar, Bülent; Odabaşi Aktaş, Elif
Background: Hormones are molecules that contribute significantly to mental and physical health due to their role in emotion regulation (ER) processes. However, the role and effect of hormones on the ER has not yet been fully elucidated. In this study, it was aimed to examine the salivary Brain-derived neurotrophic factor (BDNF) and cortisol hormone response to support ERS in primary school students depending on some parameters. Method: This cross-sectional and correlational study was conducted in the Eastern Black Sea Region of Türkiye between April 2023 and June 2023. In the sample selection, after the number of students was determined in proportional stratification, the sample selection method was made by simple random selection from each class list, first according to the school and then the grade level of each school. A total of 177 healthy students between the ages of 8 and 10 who attended these schools, were allowed to participate in the research by their families, and were volunteers, were included in the study. Data were collected using the Personal Information Form and the Emotion Regulation Scale for Children and saliva collection cups. Human-specific Brain-derived neurotrophic factor ELISA Kit (BT LAB, Cat.No E1302Hu, CHINA) for measuring BDNF levels in saliva; In measuring the level of cortisol hormone in saliva, Human Cortisol ELISA Kit (BT LAB, Cat.No E 1 003Hu, CHINA) was used in accordance with the procedure specified in the manufacturer’s catalogue. Data were evaluated using the SPSS 26.0 for Windows (SPSS, Chicago, Il, USA) package program. Independent Samples Test and One-Way ANOVA test, correlation and Multiple Linear Regression analysis were used to evaluate the data. P <.01 and p <.05 significance levels were used as statistical significance values. Results: A statistically significant correlation was found between the mean total score of the Emotion Regulation Skills Scale for Children and the mean level of salivary BDNF, and a negative correlation between the mean level of salivary cortisol (p˂0.01). Conclusions: Within the scope of the data obtained in our current study, BDNF levels were found to be high and cortisol hormone levels to be low in children with high ERS.

Erkiliç, T. Orkun; Bayraktar, B.
Background: The aim of this study was to investigate the effects of Cannabidiol on myokine (Irisin), cerebral (BDNF) and cardiac (cTnI) responses in sepsis-induced rats by Cecal Ligation Perforation (CLP) method. Methods: In this study, 40 adult male Wistar albino rats, aged between 8-10 weeks, were used. 5 groups were created, with 8 rats in each group: Control (C), Sham (S), CLP, CLP+CBD 2.5 mg/kg, CLP+CBD 5 mg/kg group. Rats in groups C and Sham were sacrificed at the end of the 10th hour and intracardiac blood was taken immediately afterwards. In groups CLP, CLP+2.5 mg/kg, CLP+5 mg/kg, intra-abdominal sepsis was induced and at the end of the 10th hour after sepsis occurred with CLP application, rats were sacrificed and intracardiac blood was taken immediately afterwards. İrisin, BDNF and cTnI levels in the serum samples were measured by ELISA method. Result: In the present study, mean serum irisin, BDNF and levels were determined to be at the lowest level in the CLP group, while cTnI levels were at the highest level (p<0.01). In CLP groups, serum irisin and BDNF levels increased due to CBD addition and the most significant decrease in cTnI levels occurred in 5 mg/kg CBD groups. As a result, it was concluded that CLP may be safe and beneficial when administered at a dose of 5 mg/kg in sepsis-induced rats.

Objective: It is known that neuropathic pain is accompanied by alterations in the levels of neurotrophic factors and synaptic proteins in the microenvironment of the spinal dorsal horn. Such changes contribute to hyperalgesia and allodynia processes; thus, analgesic drugs can exert their pharmacological effects by affecting the expressions, levels, or functions of these endogenous substances. In this study, based on the knowledge that reboxetine (a selective noradrenaline reuptake inhibitor) has the potential for antihyperalgesic efficacy in diabetic neuropathy, we aimed to examine the probable effects of this drug on diabetes-induced changes in brain-derived neurotrophic factor (BDNF), synaptophysin (the pre-synaptic marker of synaptic integration), and postsynaptic density-95 (PSD-95) (the postsynaptic marker of synaptic integration) levels in the superficial laminae of the dorsal horn. Methods: Experimental diabetes was induced by a single-dose injection of streptozotocin (STZ) (50 mg/kg) in rats. After four week-long induction period of painful diabetic neuropathy, rats were treated orally with 8 mg/kg reboxetine for two weeks. Hyperalgesia responses were evaluated by using the Randall-Selitto and Hargreave's tests. Following the pain tests, immunohistochemical studies were performed. Results: Two weeks of reboxetine administration increased the reduced paw withdrawal thresholds and shortened the paw withdrawal latencies of diabetic rats in neuropathic pain tests, indicating the antihyperalgesic efficacy of this drug. Moreover, augmented BDNF and synaptophysin levels in diabetic rats reversed by reboxetine treatment. However, there was no alteration in the densities of PSD-95, in both STZ-diabetic and reboxetine-treated STZ-diabetic rats. Conclusion: The obtained results suggested that inhibition of central sensitization and modulation of spinal plasticity seem to be pharmacological mechanisms underlying reboxetine's antihyperalgesic effects on diabetic rats. However, further studies are still needed to clarify the exact mechanism of action.

İncele - MakaleAçık Erişim2023Beneficial Effect of Atomoxetine Treatment on Scopolamine-Induced Cognitive Impairments and Molecular Changes in Rats
Yucel, Nazli Turan; Ucel, Umut Irfan; Kandemir, Ummuhan; Can, Ozgur Devrim
Objective: Atomoxetine is a noradrenaline re-uptake inhibitory drug that is currently used in the pharmacological treatment of attention deficit/hyperactivity disorder in children, adolescents, and adults. Based on the findings that noradrenergic system is closely associated with learning and memory processes, dysfunction of this system might contribute to the pathophysiology of cognitive disorders. Therefore, atomoxetine, which alters the noradrenergic neurotransmission in the synaptic cleft, has a potential to modulate learning and memory functions in various areas of the central nervous system. In the present study, it was aimed to investigate the effects of atomoxetine treatment on the cognitive impairments in amnesic animals using behavioral and immunohistochemical methods. Methods: Male Sprague-Dawley rats (adult, weighing 250-300 g) were used in the experiments. Atomoxetine at doses of 3 or 6 mg/kg (i.p.) was administered to the rats for 14 days. Experimental amnesia was induced in the animals by applying scopolamine (0.5 mg/kg, i.p.) injection. Following the treatment protocol, Morris water maze and passive avoidance tests were conducted to assess spatial and emotional learning and memory parameters of rats, respectively. Piracetam (300 mg/kg daily for 14 days) was used as reference drug in these experiments. In addition, motor performances of the animals were evaluated by activity cage and Rota-rod tests. Subsequent to behavioral tests, rats were euthanized and immunohistochemical analyses were performed to determine the alterations in the brain-derived neurotrophic factor (BDNF) and nerve growth factor (NGF) levels in the hippocampi of the animals. Results: In the Morris water maze test, amnesic rats found the hidden platform slower and spent less time in the target quadrant compared to the healthy controls. On the other hand, atomoxetine treatments significantly decreased the escape latencies, and also increased the target quadrant time values of the amnesic animals at both doses. Data obtained from the passive avoidance test showed that second transition latency values of the amnesic rats were significantly shortened with respect to the control animals; while atomoxetine treatment prolonged these shortened values significantly. It was observed that atomoxetine did not alter motor coordination or spontaneous locomotor activities of animals. Moreover, this drug increased the BDNF and NGF levels in the hippocampal sub-regions which were diminished by scopolamine administration. Conclusion: The obtained findings showed that atomoxetine treatment significantly improved the impaired learning and memory performances of the rats induced by scopolamine administration. Furthermore, anti-amnesic effects of this drug were concomitant with increased neurotrophic factors levels in the hippocampal brain regions. These results suggest a notable therapeutic potential of this drug in the treatment of dementia and several cognitive disorders; however, it should be underlined they are needed to be confirmed by welldesigned clinical trials.

This study was conducted to investigate adipokine [Apelin and brain-derived neurotrophic factor (BDNF)] and cardiac troponin (cTnI) response that emerged after adding an essential oil mixture (EOM) (Eucalytus glabutus labii, Thymus vulgaris, Cymbopogon nardus, and Syzygium aromaticum) at different rates to the drinking water of broilers in which thyroid dysfunction (hypo-and hyperthyroidism) had been experimentally induced. In the present study, 150 1-day-old Ross-308 male broiler chicks were used. They were divided into five groups, each with 30 animals. The groups were designed to include five subgroups: control (C), hypothyroid, hypothyroid + 250 ppm EOM, hyperthyroid, and hyperthyroid + 250 ppm EOM, with six animals in each group. At the end of the experiment, Apelin, p-BDNF, cTnI, T3, T4, and thyroid-stimulating hormone (TSH) levels were investigated in blood serum samples obtained by cervical dislocation from four randomly selected animals from each subgroup, making 100 animals in total. It was found that EOM administration resulted in a dose-dependent increase in p-BDNF and apelin levels, and a decrease in T4 levels in the experimentally induced hypo-and hyper-thyroidism groups, but did not affect T3 and cTnI levels. Conversely, an increase in TSH level was observed in the hypothyroidism groups, whereas a decrease was observed in groups with hyperthyroidism. This study is the first to examine adipokine (Apelin and BDNF) and cTnI response to EOM administration in thyroid dysfunction.
