Comparative evaluation of antioxidant activity and phenolic content of Lupinus albus L. (white bitter lupin, termiye) grown in different regions of Türkiye by RP-HPLC-PDA

    Kara, Yakup; Can, Zehra

Lupinus albus L. (white bitter lupin) is an important legume species widely cultivated in the Mediterranean region. This study aimed to comparatively evaluate the antioxidant capacity and phenolic composition of L. albus samples collected from different regions of Türkiye. Samples were obtained from Konya (Centre, Akşehir, Doğanhisar), Isparta (Central), and Burdur (Central). Total phenolic content, total flavonoid content and antioxidant activities of samples were determined using FRAP and DPPH assays. Additionally, the qualitative and quantitative profiles of phenolic compounds were analyzed by RP-HPLC-PDA. The total phenolic content was determined to be in the range of 1.96–2.47 mg GAE/g, whereas flavonoids were not detected in any of the samples. The results demonstrated that Lupinus albus L. extracts exhibited notable antioxidant activity, with FRAP values ranging from 1.14 to 1.95 μmol Fe²⁺/g and DPPH (SC₅₀) values ranging from 37.98 to 56.60. The phenolic composition analysis revealed the presence of major compounds including p-hydroxybenzoic acid, p-coumaric acid, ferulic acid, chrysin, pinocembrin, and galangin. Overall, the results indicate that L. albus exhibits moderate antioxidant potential despite its relatively low phenolic content, and its phenolic composition is influenced by geographical origin.

Usability of calcite mineral, which develops in the crack fillings of carbonate rocks, as ornamental stone

    Camuzcuoğlu, Murat

Turkey has a significant amount of mineral content from a geological perspective, offering essential resources, especially in mining (ornamental stone production). In this study, methods are being developed for the extraction, processing, and usability of calcite minerals, which develop as vein fillings (secondary formations) among fragmented limestone along the Bayburt-Erzurum highway for ornamental stone use. The chemical and crystallographic properties of the mineral were determined through XRD and FTIR analyses, and its suitability for ornamental stone use was evaluated. (durable, clean, and transparent) epoxy was used as a binding material for ornamental stone production, and the use of calcite minerals in jewelry production was examined, revealing its potential economic value in terms of durability, aesthetics, and rarity. Generally, while calcite minerals are widely used in various industrial fields, their use as ornamental stones remains limited. This study presents a new perspective on evaluating calcite minerals as ornamental stones, demonstrating the possibility of processing a low-hardness mineral as an ornamental stone using binding materials and its potential economic value. These findings significantly evaluate natural minerals in different applications and enhance their financial potential.

Determination of cytotoxic, anti-acetylcholinesterase and antioxidant activity of some medicinal Artemisia spp.

Gıdık, Betül; Kara, Yakup; Akar, Zeynep; Demir, Elif Ayazoglu; Can, Zehra

The study aims to investigate the antioxidant activities, phenolic compounds, acetylcholinesterase enzyme inhibition and cytotoxic effects of two different Artemisia spp. (Artemisia dracunculus L. and Artemisia dracunculoides Pursh) cultivated in Türkiye organically, for the first time. The total phenolic and total flavonoid contents of the plants were determined spectrophotometrically while antioxidant activity DPPH•, CUPRAC, and FRAP was determined using the colorimetric method. And also, acetylcholinesterase in vitro enzyme inhibition activity and anti-cancer activity against human melanoma (VMM917, CRL-3232), lung carcinoma (A549, CCL-185), and normal human fibroblast (hGF, PCS-201-018) cells were studied. Total phenolic (225-324 μg GAE/mL extract) and total flavonoid contents (0.066-0.085 μg QE/mL extract), antioxidant activity Color Scavenging Concentration 50% (DPPH• (CSC50: 1.371-1.655 mg/mL), CUPRAC (0.246-0.344 μM CTEAC) and FRAP (462.133-726.661 μM CTEAC)). A. dracunculus and A. dracunculoides extracts inhibited 40.09 ± 0.65%, and 39.48 ± 3.68% of acetylcholinesterase activity. It was determined that demonstrated the selective effect of A. dracunculus and A. dracunculoides on the cytotoxicity of A549 and hGF cells. According to our results, both tarragon plant species, it may be said that it is not only consumed as food but also used for therapeutic purposes will be beneficial for health.

Molecular docking study on acetylcholinesterase and urease enzyme inhibition effects of Amygdalin

Güler, Halil Ibrahim; Kolaylı, Sevgi; Kara, Yakup; Birinci, Ceren; Can, Zehra

This study investigated that the acetylcholinesterase, urease enzyme inhibition and these enzymes molecular docking as well as antioxidant activity of commercially available amygdalin. While Amygdalin displayed effect anti-urease activity compared to acetohydroxamic acid but, anti-acetylcholine esterase activity a little ineffective compared to donepezil. The molecular docking was performed in order to check binding interactions between the amygdalin and the enzymes. DPPH and FRAP assays were preferred to determine the antioxidant activity. The antioxidant activity (3.39±0.33 µmol Fe2SO47H2O /g, SC50 18.74±0.72 mg/mL using the FRAP and DPPH assays, respectively. Amygdalin's in vitro and in vivo studies are needed for demonstrated that is a therapeutic agent for the treatment of various diseases.
