BACKGROUND: One of the major contributors to postharvest losses in tomatoes is the development of infectious mold diseases. In this study, four distinct biodegradable film formulations were developed incorporating postbiotics derived from Lactiplantibacillus plantarum Y48 and the polysaccharide alternan. The formulations included alternan, sodium alginate and glycerol (AAG), alternan, postbiotic, sodium alginate and glycerol (APAG), postbiotic, sodium alginate and glycerol (PAG), and sodium alginate and glycerol (AG) as a control. The films were characterized in terms of their physical, mechanical, thermal, and morphological properties. To assess antifungal efficacy, tomatoes artificially inoculated with Alternaria alternata were coated with each film and stored at ambient temperature for 8 days. RESULTS: The incorporation of alternan and postbiotics enhanced both the mechanical strength and thermal stability of the films. Fourier-transform infrared (FTIR) analysis confirmed the successful integration of the bioactive components into the polymer matrix. Scanning electron microscopy (SEM) imaging revealed that postbiotic addition contributed to a rougher and more rigid surface morphology, while alternan improved the structural cohesion of the films. All film formulations exhibited significant inhibitory effects against A. alternata, with the PAG formulation demonstrating the most pronounced antifungal activity. CONCLUSION: These results suggest that the PAG film, enriched with postbiotics, represents a promising biocontrol strategy for mitigating fungal spoilage in tomatoes. In conclusion, this study underscores the potential of combining postbiotics and alternan for the development of active, biodegradable edible films with antifungal functionality for postharvest preservation.

Bayaman, Rabia Yusra; Alkay, Zuhal; Ispirli, Humeyra; Arioglu-Tuncil, Seda; Dere, Sevda; Can, Hasan; Gonzales, Miguel Angel Alvarez; Sagdic, Osman; Lindemann, Stephen R.; Tuncil, Yunus Emre

The production of novel oligosaccharides with potential prebiotic effects is of interest to expand the current market and explore the effectiveness of new functional carbohydrate forms. The utilization of glucansucrases is a cost-effective and environmentally friendly biotechnological strategy for producing novel gluco-oligosaccharides through acceptor reactions. In this study, an active glucansucrase (GS53) was used to produce gluco-oligosaccharides via its acceptor reactions with glucose, maltose, and maltotriose, and these oligosaccharides were tested in terms of structure and their gut microbiome modulatory effects. The formations of oligosaccharides were monitored by TLC analysis, and GS53 was active for the three acceptors but not for the other sugars tested. The structural characterization of the gluco-oligosaccharides by 1H NMR analysis revealed the glycosylation of each acceptor with alpha-(1 -> 3) and alpha-(1 -> 6) linkages, whereas LC-MS analysis demonstrated the formations of DP 8, DP 7, and DP 6 oligosaccharides with acceptors maltose, maltotriose, and glucose, respectively. In vitro fecal fermentation analysis, in which microbial short-chain fatty acids (SCFAs) and microbial compositional changes were assessed using gas chromatography and 16S rRNA sequencing, respectively, demonstrated that the gluco-oligosaccharides formed SCFAs-particularly propionate and butyrate-at levels comparable to those observed with inulin, a well-established prebiotic. Additionally, the gluco-oligosaccharides were found to promote the growth of Bifidobacterium adolescentis and Blautia OTUs, which are known to have important physiological functions beneficial to human health. Overall, these results demonstrate that gluco-oligosaccharides synthesized using GS53 through acceptor reactions exhibit prebiotic potentials and could be utilized in the future as dietary supplements as well as in the development of functional foods targeting colonic health.

ElObeid, Tahra; Yilmaz, Mustafa Tahsin; Ispirli, Humeyra; Sagdic, Osman; Taylan, Osman; Basahel, Abdulrahman; Karaboga, Dervis; Durak, Muhammed Zeki; Dertli, Enes

In this study, the alternan/selenium nanoparticles (Alt/SeNPs) were characterized with respect to their formation, morphology, size, selenium distribution, molecular, crystallographic, and thermal properties using UV–Vis spectroscopy, SEM, TEM, EDAX, FTIR, XRD, DSC and TGA measurements. UV–VIS measurements confirmed the synthesis of nanoparticles by observing a maximum surface plasmon resonance peak at 212 nm. In this study, alternan contributed to stabilizing the dispersion of SeNPs, resulting in a cluster of spherical and well-dispersed nanoparticles ranging in size from 50 to 90 nanometers. Nanoparticles were found to be highly thermally stable and in a nanocrystalline structure. The ABTS and CUPRAC radical scavenging activities of Alt/SeNPs were remarkable (95% and 78% at 4 and 6 mg/mL levels of Alt/SeNPs, respectively). Alt/SeNPs had also good inhibitory activities (3.5–4.0 and 4–15 mm of inhibition zone levels at 5 mg/mL level of Alt/against foodborne pathogenic bacteria and fungi, respectively).

Dere, Sevda; İspirli, Hümeyra; Korkmaz, Kader; Tuncil, Yunus Emre; Sagdic, Osman; Narbad, Arjan; Ayyash, Mutamed; Dertli, Enes

Glucansucrases (GS) are functional enzymes for the production of distinct glucan and gluco-oligosaccharide structures with potential bioactive characteristics. In this study, an active glucansucrase (GS40) was identified within the genome of Leuconostoc mesenteroides YTU 40 that was further cloned, heterologously expressed and biochemically characterised. GS40 was capable of production of a branched dextran with ~ 20% (1→3)-α-D-Glc residues detected by1H and13C NMR analysis. Acceptor reactions of GS40 with maltose and cellobiose were performed and distinct maltose/cellobiose derivative acetylated gluco-oligosaccharides were produced detected by TLC, NMR and LC-MS analysis. The bioactive characteristics of branched dextran and gluco-oligosaccharides were tested by determination of their immune-modulatory roles in terms of triggering IL-12, IL-10, IL-4 and TNF-α cytokine levels. These findings can be important to unveil the role of glucansucrases for the production of gluco-oligosaccharides that can be used as functional ingredients for food and other industries.

Özpınar, Fatma Beyza; İspirli, Hümeyra; Kayacan, Selma; Korkmaz, Kader; Dere, Sevda; Sagdic, Osman; Alkay, Zuhal; Tunçil, Yunus Emre; Ayyash, Mutamed; Dertli, Enes

Exopolysaccharide (EPS) producing Lactic Acid Bacteria (LAB) species can be presented in distinct environments. In this study, Turkish fermented sausage (sucuk) was tested for the presence of EPS producer LAB strains and slimy-mucoid colonies were selected for further tests. Among the isolates, Weissella confusa strain S6 was identified and tested for the physicochemical characterisation of its EPS. This strain was found to produce 0.74 g L−1 of EPS in modified BHI medium conditions. Structural characterisation of EPS S6 by 1H and 13C NMR demonstrated that EPS S6 was a highly branched dextran type glucan formed by mainly (1 → 2)-linked α-D-glucose units together with low levels of (1 → 3)-linked α-D-glucose units as branching points. This structure was further confirmed by methylation analysis detected by GC–MS. An average molecular weight of 8 × 106 Da was detected for dextran S6. The FTIR analysis supported the dextran structure and revealed the presence of distinct functional groups within dextran S6 structure. A strong thermal profile was observed for dextran S6 detected by DSC and TGA analysis and dextran S6 revealed a degradation temperature of 289 °C. In terms of physical status, dextran S6 showed amorphous nature detected by XRD analysis. SEM analysis of dextran S6 demonstrated its rough, compact and porous morphology whereas AFM analysis of dextran S6 detected in its water solution showed the irregularity with no clear cross-link within the dextran chains. These technological features of dextran S6 suggests its potential to be used for in situ or ex situ application during meat fermentations.

The development of new starter cultures is a crucial task for the food industry to meet technological requirements and traditional products are important reservoirs for new starter cultures. In this respect, this study aimed to isolate, identify, and determine the technological characteristics of Lactobacillus delbrueckii subsp. bulgaricus and Streptococcus thermophilus strains originated from traditional yogurt samples. Genotypic discrimination of 200 isolates revealed the presence of distinct 19 S. thermophilus and 11 Lb. delbrueckii subsp. bulgaricus strains as potential starter cultures. Strain-specific properties determined the acidification capacity of the yogurt starter cultures and a higher acidification capacity was observed for S. thermophilus strains compared to Lb. delbrueckii subsp. bulgaricus strains. Proteolytic activity was found between 0.012–0.172 and 0.078–0.406 for S. thermophilus and Lb. delbrueckii subsp. bulgaricus strains, respectively. 4 of S. thermophilus and 3 of Lb. delbrueckii subsp. bulgaricus strains were found resistant to all tested bacteriophages. The antibiotic susceptibility tests of the isolates revealed that a very low antibiotic resistance was observed for the yogurt starter cultures. Finally, the growth kinetics of selected strains were determined and the maximum specific growth rate of selected S. thermophilus and Lb. delbrueckii subsp. bulgaricus was calculated as 0.527 h−1 and 0.589 h−1, respectively.

Utilisation of an active branching sucrase from Lactobacillus kunkeei AP-37 to produce techno-functional poly-oligosaccharides

İspirli, Hümeyra; Korkmaz, Kader; Arioglu-Tuncil, Seda; Bozkurt, Fatih; Sağdıç, Osman; Tunçil, Yunus Emre; Narbad, Arjan; Dertli, Enes

Glucansucrase AP-37 was extracted from the culture supernatant of Lactobacillus kunkeei AP-37 and characteristics of the glucan produced by the active glucansucrase in terms of structural and functional roles were determined in this study. A molecular weight around 300 kDa was observed for glucansucrase AP-37 and its acceptor reactions with maltose, melibiose and mannose were also conducted to unveil the prebiotic potential of the poly-oligosaccharides formed via these reactions. The core structure of glucan AP-37 was determined by 1H and 13C NMR and GC/MS analysis which revealed that glucan AP-37 was a highly branched dextran composing of high levels of (1 → 3)-linked α-D-glucose units with low levels of (1 → 2)-linked α-D-glucose units. The structural features of the glucan formed, demonstrated that glucansucrase AP-37 was an α-(1 → 3) branching sucrase. Dextran AP-37 was further characterised by FTIR analysis and XRD analysis demonstrated its amorphous nature. A fibrous compact morphology was observed for dextran AP-37 with SEM analysis whereas TGA and DSC analysis revealed its high stability as no degradation was observed up to 312 °C. Finally, the prebiotic potential of the dextran AP-37 and the gluco-oligosaccharides produced with the acceptor reaction of α-(1 → 3) branching sucrase AP-37 were determined and promising results were found for the gluco-oligosaccharides to act as prebiotics.

Ecem Bayram, Nesrin; Çebi, Nur; Çelik, Saffet; Gerçek, Yusuf Can; Bayram, Sinan; Tanugur Samanci, Asli Elif; Sagdiç, Osman; Özkök, Asli

Royal jelly is one of the most remarkable commercial bee products used in cosmetics, medicine, and as a dietary supplement. This study aimed to determine some quality parameters (amino acid, multi-elemental, total phenolic-flavonoid, 10-HDA, sugar, and moisture content) as well as antioxidant and antimicrobial activities of different royal jelly samples (domestic and non-domestic samples). In this study, royal jelly samples exhibited high antioxidant and antimicrobial activity. 10-HDA values were found to be higher in domestic royal jelly samples (2.17-2.71%) compared to non-domestic samples (0.31-1.71%). Also, it was found that the acidity value varied between 2.57-3.37 mL 0.1 N NaOH/g and 3.87-4.97 mL 0.1 N NaOH/g in non-domestic and domestic royal jelly samples, respectively. L-Proline was the amino acid with the highest rate of occurrence in both domestic (21972.39-35053.00 nmol/g) and non-domestic (19688.31-21711.50 nmol/g) samples. It was determined that the new method developed for amino acid analysis has many advantages over other methods. Chemometrics of principal component analysis (PCA) showed that amino acid profile, elemental profile, and chemical parameters were capable of presenting characteristic and descriptive properties of royal jelly samples. In addition to these findings, Raman spectroscopy combined with chemometrics of hierarchical cluster analysis (HCA) was successfully used to distinguish domestic and non-domestic royal jelly samples. The obtained results will provide an important basis for subsequent research on the evaluation of the amino acid content, physicochemical properties, phenolic content, and the antioxidant and elemental profile of royal jelly.
