Uzunçakmak, Sevgi Karabulut; Özcan, Halil; Dirican, Ebubekir

Objective The aim herein was to investigate mitochondrial cytochrome B (MT-CYB) mutations in individuals with bipolar disorder. Stanniocalcin-1 (STC1) and uncoupling protein 2 (UCP2) mRNA expressions and their relationship with clinical data and each other were also investigated. Method The blood samples of 100 individuals were included in this study. Real-time PCR was used to evaluate mRNA expressions of STC1 and UCP2. Genetic alterations were investigated via Sanger DNA sequencing. An in silico analysis was performed to reveal the phenotypic effects of MT-CYB mutations. Results In the MT-CYB gene of the bipolar disorder patients, the most seen mutations were the T194A A>G mutation at position 1532, G deletion at position 15498, and C>A L236I mutation at position 15452. Most of the mutations appeared to be neutral or benign. The UCP2 and STC1 mRNA expression levels were significantly higher in the patients than in the healthy controls (P = 0.0124 and P < 0.0001, respectively). The area under the curve values of the receiver operating characteristic curve analysis for UCP2 and STC1 were 0.6631 (P = 0.0123) and 0.8059 (P < 0.0001), respectively. No significant relationship was observed between the gene expressions and the routine laboratory findings. There was a positive correlation between the UCP2 and STC1 mRNA expressions in the bipolar disorder patients (r = 0.03559, P = 0.0306). Conclusion Expression of UCP2 and STC1 may be important parameters in bipolar disorder. MT-CYB mutations may be related to gene expressions. Comprehensive studies on bipolar disorder will help better understand UCP2 and STC1 gene functions.

Evaluating STC-1 and STC-2 mRNA expressions in Schizophrenia patients with increased oxidative stress; [Évaluation des expressions d'ARNm de STC-1 et STC-2 chez des patients schizophrènes présentant un stress oxydatif accru]

Karabulut Uzunçakmak, Sevgi; Özcan, Halil; Dirican, Ebubekir; Tavacı Özçelik, Ayşegül

Introduction: Schizophrenia is a chronic psychiatric disorder characterized by cognitive impairment. Oxidative stress is associated with disease progression in patients with schizophrenia. Stanniocalcin (STC)-1 and STC-2 are two proteins commonly expressed in mammals belonging to the stanniocalcin family and they are involved in oxidative stress. The aim of this study is to investigate STC-1 and STC-2 mRNA expressions in schizophrenia patients with altered oxidative stress parameters. Methods: For this purpose, 70 patients with schizophrenia and 40 healthy individuals were recruited for the study. Peripheral blood samples were obtained from all participants. Glutathione (GSH) activities, superoxide dismutase (SOD) activities, and malondialdehyde (MDA) levels were measured. STC-1 and STC-2 expressions were assessed by real-time polymerase chain reactions. Results: SOD activity levels were lower in patients than in healthy individuals (p = 0.0309), while the patients’ MDA levels were higher (p = 0.039). STC-1 and STC-2 expressions were lower in patients than in healthy individuals (p = 0.1049 and p < 0.0001, respectively). Furthermore, these two genes had a positive correlation among the patients (r = 0.435, p = 0.0025). According to area under the curve (AUC) values, STC-2 (AUC = 0.8332, p < 0.0001) had better diagnostic power than STC-1 (AUC = 0.6167, p = 0.1037). Conclusions: The expression of stanniocalcins in schizophrenia was investigated here for the first time. Decreased STC-2 expression in patients with schizophrenia with increased oxidative stress parameters may guide the understanding of the pathophysiological mechanisms caused by oxidative stress, which may be increased in cognitive diseases such as schizophrenia, and it also has the potential to be a prognostic factor that can be used in the diagnosis of schizophrenia.

Unveiling the significance of Gremlin-1 as a novel biomarker for enhanced severity prediction in COVID-19 patients

Celik, Muhammet; Tekin, Erdal; Koçak, Mehmet Nuri; Uzunçakmak, Sevgi Karabulut; Bayraktar, Mustafa

Gremlin-1 is associated with lung disease and plays a role in the initiation and progression of pulmonary fibrosis. This suggests that Gremlin-1 may be associated with lung involvement in COVID-19 and poor clinical outcomes and warrants further investigation. This prospective, cross-sectional, single-blind study was the first to investigate Gremlin-1 levels in COVID-19 patients and whether Gremlin-1 levels could be used to predict and guide clinical follow-up in outpatients and inpatients. In this context, serum Gremlin-1 levels were measured and compared in 2 groups of patients who were diagnosed as positive for COVID-19 by RT-PCR and were followed up as outpatients (n = 28) or required hospitalization (n = 30). The median Gremlin-1 values were statistically significantly different between outpatients and intensive care unit patients (0.367 ng/mL [IQR = 0.377], 1.858 ng/mL [IQR = 2.245], respectively) (P < .0001). The area under the ROC curve value to determine the discriminative power of Gremlin-1 was found to be 0.772 (95% CI: 0.672-0.871; P < .0001), and the cutoff value of Gremlin-1 to discriminate between outpatients and hospitalized patients was found to be 1.242 ng/mL with 66.7% sensitivity and 67.2% specificity. Serum Gremlin-1 level is an important biomarker that can be used as a clinical decision-making tool for COVID-19 positive patients.

The potential health-promoting effects of probiotics against intoxication by pesticides is a topic of increasing commercial interest with limited scientific evidence. In this study, we aimed to investigate the positive effects of probiotic Saccharomyces boulardii on the male reproductive system under low dose neonicotinoid pesticide exposure conditions. We observed that acetamiprid and imidacloprid caused a degeneration and necrosis of the spermatocytes in the tubular wall, a severe edema of the intertubular region and a hyperemia. This was concomittant to increased levels of 8-hydroxy-2 '-deoxyguanosine reflecting oxidative stress, and an increase in caspase 3 expression, reflecting apoptosis. According to our results, Saccharomyces boulardii supplementation mitigates these toxic effects. Further in vivo and clinical studies are needed to clarify the molecular mechanisms of protection. Altogether, our study reinforces the burden of evidence from emerging studies linking the composition of the gut microbiome to the function of the reproductive system.

LRIG1 levels in chronic rhinosinusitis with nasal polyps

Uzunçakmak, Sevgi Karabulut; Sahin, Abdulkadir; Özçelik, Aysegul Tavaci; Halici, Zekai

Purpose: Nasal polyps (NPs), usually occurring together with chronic rhinosinusitis (CRS), are benign masses of mucosal origin arising from inflammation. The transmembrane protein known as leucine-rich repeats and immunoglobulin-like domains 1 (Lrig1) is a member of the Lrig family. Lrig1 is frequently expressed in the respiratory tract and epithelial tissues and can inhibit several signaling pathways involved in cell proliferation. The aim of this study was to determine Lrig1 levels in NP tissues of patients with CRS. Material and Methods: This study included 36 patients with CRS and NPs and 15 patients who underwent rhinoplasty as the control group. The Lrig1 levels of all participants were measured by the ELISA method. Results: This study revealed that Lrig1 levels were significantly lower in NP tissues than in tissues of the control group. The mean level of Lrig1 of the NP tissues was 22.2 ng/ml, while the mean level of the control group was 28.5 ng/ml. According to the results of ROC analysis, Lrig1 levels have the power to distinguish polyp tissues from control tissues (AUC=0.794). Lrig1 levels were higher in tissues with scores of 4-8 than in tissues with scores of 16-20 based on the results of computed tomography scoring. According to endoscopic evaluations, Lrig1 levels of tissues with scores of 5-8 or 9-11 were relatively lower than those of tissues with scores of 2-4.Conclusion: Lrig1 levels were found to be decreased in NP tissues. Thus, Lrig1 may be used to confirm the presence of NPs. Lrig1 may also be helpful in NP grading. Increasing the Lrig1 levels in cases of NPs has the potential to become a targetable treatment modality.

A comparison of the associations between bone turnover markers and different sports fields: combat versus team sports

    Yıldız, K.; Uzunçakmak, S. Karabulut; Halıcı, Z.; Özçelik, C.; Yazıcı, A.G.

Sedentary life brings risks that include osteoporosis, while physical activity has many benefits for health in general and the skeletal system. Irisin is a hormone-like myokine regulated by mechanical force and it contributes to cortical and trabecular bone mineral density. This study aimed to investigate the effects of different types of sports on bone health by measuring elite athletes’ fibronectin type III domain 5 (FNDC5), N-terminal collagen type I extension propeptide (PINP), and C-terminal cross-linking telopeptide of type I collagen (CTXI) levels. Combat sports athletes, team sports athletes, and sedentary control subjects were enrolled. Serum levels of proteins were measured by ELISA. FNDC5 levels of elite athletes were higher compared to the sedentary controls (P=0.0014). The highest FNDC5 level was measured in wrestlers among all considered groups. The PINP levels of the athletes were also higher than those of the sedentary control group (P=0.1431). The highest PINP level was determined in boxers. The CTXI levels of the athletes were lower than those of the sedentary control (P<0.0001). The lowest CTXI level was seen in boxers. The FNDC5 and PINP levels of combat sports athletes were higher than those of team sports athletes (P=0.0134 and P=0.0262, respectively), while the CTXI levels of combat sports athletes were lower than those of team sports athletes (P<0.0001). Our results indicated that FNDC5, PINP, and CTXI levels are associated with physical activity. The effect of the exercises performed by athletes of specific sports on bone health has not been studied in much detail before. Combining different exercises for athletes of a particular sport may be more beneficial for bone health.

Uzuncakmak, Sevgi Karabulut; Dirican, Ebubekir; Ozcan, Halil; Takim, Ugur

Objective: Schizophrenia is a serious mental disorder. Mutations in mitochondrial genes can change energy metabolism. Telomere is a tandem sequence at the end of chromosomes. Shorter telomere length has been shown in schizophrenia. The aim of this study was to determine the relationship between ATPase6 gene mutations and telomere length in schizophrenia patients. Methods: Blood samples of 34 patients and 34 healthy controls were used. In this study conventional PCR, Sanger sequencing technic and real-time PCR were utilized. Results: Five different mutations (A8860G, A8836, G8697A, C8676T, and A8701G) in the ATPase6 gene were identified in schizophrenia patients. The most seen mutation was A8860G (94%). Telomere length analysis indicated the relation of ATPase6 gene mutations and telomere length variations (p = 0.001). Patients carrying the A8860G mutation had shorter telomere lengths than patients carrying other mutations. Comparing telomere length between schizophrenia patients and healthy controls revealed that the mean telomere length of schizophrenia patients was shorter than healthy controls (p = 0.006). The demographic analysis demonstrated a significant relationship between marital status and telomere length (p = 0.011). Besides that, the duration of the illness is another factor that impacts telomere length (p = 0.044). There is no significant relation between telomere length and other clinical and demographic characteristics including education status, age, gender, etc. Conclusion: In conclusion, telomere length and ATPase6 gene mutations have a significant relation. Studies with larger patient populations and investigation of other mitochondrial gene mutations will make the clearer link between telomere length and mitochondrial mutations.

Evaluation of IGFBP5 expression and plasma osteopontin level in COVID-19 patients

Karabulut Uzunçakmak, Sevgi; Aksakal, Alperen; Kerget, Ferhan; Aydın, Pelin; Halıcı, Zekai

Purpose: The aim of this study is to investigate insulin-like growth factor binding protein 5 (IGFBP5) expression in coronavirus disease 2019 (COVID-19) patients and its relationships with COVID-19 laboratory findings and plasma osteopontin (OPN) levels. Materials and methods: We enrolled 60 patients with COVID-19 and 30 healthy individuals in this study. mRNA expression of IGFBP5 was measured by RT-PCR. Plasma OPN levels were measured via the ELISA method. Results: Plasma OPN levels were higher and IGFBP5 expression levels were lower in COVID-19 patients than in the healthy individuals (p = 0.0057 and p = 0.0142, respectively). Critically ill patients had higher OPN and lower IGFBP5 than non-critically ill patients. Patients with affected lungs demonstrated increased OPN and decreased IGFBP5 (p = 0.00032 and p = 0.044, respectively). Receiver operating characteristic (ROC) analysis indicated that IGFBP5 expression and OPN levels can be used discriminate non-critically from critically ill patients (p = 0.049; p = 0.0016, respectively). Conclusion: This study demonstrated that patients with a poor prognosis had increased OPN and decreased IGFBP5. High values of OPN and low values of IGFBP5 may be considered as signs of disease severity. Tissue-specific IGFBP5 expression may contribute to understanding the role of IGFBP5 in the lungs in COVID-19 cases.

The melatonin agonist ramelteon attenuates bleomycin-induced lung fibrosis by suppressing the NLRP3/TGF-Β1/HMGB1 signaling pathway

Aydin, Pelin; Aksakalli-Magden, Zeynep B.; Civelek, Maide S.; Karabulut-Uzuncakmak, Sevgi; Mokhtare, Behzad; Ozkaraca, Mustafa; Alper, Fatih; Halici, Zekai

Purpose: The possible effects of ramelteon, a melatonin receptor agonist on bleomycin-induced lung fibrosis were analyzed via transforming growth factor β1 (TGF-β1), the high mobility group box 1 (HMGB1) and Nod-like receptor pyrin domain-containing 3 (NLRP3) which are related to the fibrosis process. Materials and methods: Bleomycin (0.1 mL of 5 mg/kg) was administered by intratracheal instillation to induce pulmonary fibrosis (PF). Starting 24 h after bleomycin administration, a single dose of ramelteon was administered by oral gavage to the healthy groups, i.e. PF + RM2 (pulmonary fibrosis model with bleomycin + ramelteon at 2 mg/kg) and PF + RM4 (pulmonary fibrosis model with bleomycin + ramelteon at 4 mg/kg) at 2 and 4 mg/kg doses, respectively. Real-time polymerase chain reaction (real-time PCR) analyses, histopathological, and immunohistochemical staining were performed on lung tissues. Lung tomography images of the rats were also examined. Results: The levels of TGF-β1, HMGB1, NLRP3, and interleukin 1 beta (IL-1β) mRNA expressions increased as a result of PF and subsequently decreased with both ramelteon doses (p < 0.0001). Both doses of ramelteon partially ameliorated the reduction in the peribronchovascular thickening, ground-glass appearances, and reticulations, and the loss of lung volume. Conclusions: The severity of fibrosis decreased with ramelteon application. These effects of ramelteon may be associated with NLRP3 inflammation cascade.
