Biosynthesis of Functionalized Silver Nanomaterials Utilizing Seeds of Black Rosehip (Rosa pimpinellifolia L.) and its Remarkable Antioxidative Capacity

    Kizil, Hamit Emre

Background: Silver nanoparticles (AgNPs) have gained significant attention due to their unique antimicrobial, catalytic and optical properties. This study focuses on the green synthesis of silver nanoparticles using black rosehip (Rosa pimpinellifolia L.) seeds (Rp-AgNPs) and evaluates their biological activities. Methods: Rp-AgNPs were synthesized using R. pimpinellifolia seed extract and characterized using multiple analytical techniques including UV-Vis spectroscopy, XRD, FTIR, TEM, SEM, EDAX, zeta potential analysis, DSC, SERS and XPS. The biological activities were assessed through DPPH free radical scavenging assay, cytotoxicity testing against H460 lung cancer cells and antimicrobial activity evaluation against various pathogens. Result: The synthesized Rp-AgNPs exhibited spherical morphology with an average size of 51.27 nm and demonstrated stability with a zeta potential of-18.9. The nanoparticles showed enhanced antioxidant activity (IC50 = 30.89 μg/mL) compared to the seed extract (IC50 = 136.4 μg/mL). Cytotoxicity studies revealed significant effects against H460 cells with an IC50 of 65.8 μg/mL. Antimicrobial testing demonstrated effectiveness against both Gram-positive and Gram-negative bacteria (MIC 50-100 μg/mL) and Candida strains (MIC 100-200 μg/mL).

The present study focuses on synthesis of silver nanoparticles (AgNPs) by employment of a green and facile synthesis method. Aqueous extract of Helichrysum arenarium was used as a reducing and capping agent. The bioreduction of silver particles was monitored using UV–Vis spectroscopy based on increase in color intensity led by surface plasmon resonance. SEM, TEM, FTIR, DSC, and XRD analyses were followed for characterization, divulging that the nanoparticles were thermally resistant in the size of approximately 20 nm with a quasi-spherical shape without any agglomeration. AgNPs possessed a nanocrystal form with presence of crystalline Ag. EDAX analysis was used to verify that the nanoparticles were silver as revealed by a typical optical absorption peak of metallic silver nanocrystallites. The AgNPs were shown to have strong antibacterial and antifungal effects on some foodborne pathogens. The method developed in this study could offer a green, facile biomimetic, and cost-effective method.

Synthesis of silver nanoparticles prepared with a dextran-type exopolysaccharide from Weissella cibaria MED17 with antimicrobial functions

İspirli, Hümeyra; Sagdic, Osman; Dertli, Enes

Microbial polysaccharides can find distinct applications as stabilizer agents including synthesis of nanoparticles. In this study, a dextran-type exopolysaccharide (EPS) was used as the stabilizer agent for the green synthesis of silver nanoparticles (AgNPs-Dex) with antimicrobial characteristics. UV–vis spectroscopy analysis was used to test the formation of AgNPs-Dex. The uniform morphology at around 10 nm size was observed for AgNPs-Dex by TEM analysis and importantly EDX analysis demonstrated the embedment of Ag+ ions within dextran as the stabilizer agent. XRD analysis confirmed the crystalline nature of AgNPs-Dex and FTIR analysis demonstrated the interactions of dextran functional groups with silver. DSC and TGA analysis showed the alteration in the thermal stability of AgNPs-Dex compared to the stabilizer dextran. The antibacterial and antifungal activities of AgNPs-Dex were determined against food originated pathogenic bacteria and fungi and important inhibition levels were observed at 1 mg ml−1 concentration of AgNPs-Dex and this activity was observed to be concentration dependent.

Synthesis and characterisation of alternan-stabilised silver nanoparticles and determination of their antibacterial and antifungal activities against foodborne pathogens and fungi

Yilmaz, Mustafa Tahsin; İspirli, Hümeyra; Taylan, Osman; Dertli, Enes

In this study, alternan isolated from Lactobacillus reuteri E81 was used as the stabilising agent to produce alternan-stabilised silver nanoparticles (AgNPs-Alt) and AgNPs-Alt were characterized and their antibacterial and antifungal activities were tested. The formation and the stability of AgNPs-Alt were determined by UV–vis spectroscopy analysis. SEM and TEM analysis revealed the uniform morphology of AgNPs-Alt with an average size of ~50 nm. The crystalline nature of the AgNPs-Alt was confirmed by XRD analysis. The interactions of alternan functional groups with silver was observed by FTIR analysis and thermal characteristics of AgNPs-Alt were determined by DSC analysis. Finally, the antibacterial and the antifungal activities of AgNPs-Alt were tested and AgNPs-Alt revealed inhibitory effects to pathogenic strains at 0.25 mg ml−1 concentration and this activity was observed to be concentration dependent. For the antifungal activity, a higher level of AgNPs-Alt was required compared to the antibacterial activity.
