Chemical and bioactive potential of the nests of Polistes nimpha, Polistes dominula, and Vespa crabro (Hymenoptera: Vespidae)

Ertürk, Ömer; Can, Zehra; Yaman, Mustafa

This work was accomplished to establish the chemical components and bioactivity potential of the nest materials of Polistes nimpha (Christ), Polistes dominula (Christ), and Vespa crabro (L.). The biological and chemical compounds of materials and their molecular functionalities were detected using FRAP (ferric reducing antioxidant power) method, DPPH (2,2-diphenyl-1-picryl-hydrazyl-hydrate) free radical method, and Energy Dispersive X-Ray Analysis. Finally, the bioactivity potentials of nest extracts were assayed and phenolic components were determined. C, N, O, Na, Ca, K, Mg, Al, Si are the elemental components of the nest materials. All nest extracts of three species had high biological activity against nine bacteria and one fungus causing common infections. The maximum antibacterial and antifungal activity was seen when gram-negative Pseudomonas aeruginosa, gram-positive Bacillus cereus, and gram-positive Candida albicans were exposed to ethanol extracts of nest samples in 25 μL. High antioxidant activity can be found in the nest extracts. These extracts might help researchers find novel antifungal and antibacterial substances.

Antioxidant, antimicrobial activities and fatty acid compositions of wild berberis spp. by different techniques combined with chemometrics (PCA and HCA)

    Gıdık, Betül

Interest in medicinal plants and fruits has increased in recent years due to people beginning to consume natural foods. This study aims to investigate the total phenolic flavonoid content, antiox-idant activity, condensed tannin content, oil content, and fatty acid compositions of five local breeds of Berberis spp. from Bayburt, Turkey, and their antioxidant and antimicrobial activities. The fatty acid composition of samples was performed with gas chromatography-mass spectrometry (GC-MS), and the total fatty acid content of samples was between 6.12% and 8.60%. The main fatty acids in Berberis spp. samples were α-linolenic acid (32.85–37.88%) and linoleic acid (30.98–34.28%) followed by oleic acid (12.85–19.56%). Two antioxidant assays produced similar results, demonstrating that extracts of wild B. vulgaris L. had the highest ferric reducing antioxidant power (FRAP) (621.02 µmol FeSO4.7H2O/g) and 1,1-diphenyl-2-picrylhydrazyl radical (DPPH) (0.10 SC50 mg/mL) values. According to principal component analysis (PCA), four components were determined. In addition, two main groups were determined according to hierarchical cluster analysis (HCA), and wild and culture of B. vulgaris L. were in different subgroups. This is the first original report about the fatty acid composition and oil content of Berberis spp. grown in Bayburt, Turkey. The obtained results indicate that B. integerrima Bunge and B. vulgaris, which have especially remarkable fatty acid content, antioxidant, and antimicrobial activity, could be potential sources for these properties in different areas of use.

Isolation and Biophysical Characterisation of Bioactive Polysaccharides from Cucurbita Moschata (Butternut Squash)

Jiwani, Shahwar Imran; Gillis, Richard B.; Besong, David; Almutairi, Fahad; Erten, Tayyibe; Kök, M. Samil; Harding, Stephen E.; Paulsen, Berit S.; Adams, Gary G.

Cucurbits are plants that have been used frequently as functional foods. This study includes the extraction, isolation, and characterisation of the mesocarp polysaccharide ofCucurbita moschata. The polysaccharide component was purified by gel filtration into three fractions (NJBTF1, NJBTF2, and NJBTF3) of different molecular weights. Characterisation includes the hydrodynamic properties, identification of monosaccharide composition, and bioactivity. Sedimentation velocity also indicated the presence of small amounts of additional discrete higher molecular weight components even after fractionation. Sedimentation equilibrium revealed respective weight average molecular weights of 90, 31, and 19 kDa, with the higher fractions (NJBTF1 and NJBTF2) indicating a tendency to self-associate. Based on the limited amount of data (combinations of 3 sets of viscosity and sedimentation data corresponding to the 3 fractions), HYDFIT indicates an extended, semi-flexible coil conformation. Of all the fractions obtained, NJBTF1 showed the highest bioactivity. All fractions contained galacturonic acid and variable amounts of neutral sugars. To probe further, the extent of glycosidic linkages in NJBTF1 was estimated using gas chromatography-mass spectrometry (GCMS), yielding a high galacturonic acid content (for pectin polysaccharide) and the presence of fructans-the first evidence of fructans (levan) in the mesocarp. Our understanding of the size and structural flexibility together with the high bioactivity suggests that the polysaccharide obtained fromC. moschatahas the potential to be developed into a therapeutic agent.
