Anatomical and Phytochemical Characterization of Ranunculus pinardii and Ranunculus constantinopolitanus: LC-MS/MS-Based Bioactive Compound Profiling

Feizollahi, Sepideh; Gercek, Yusuf Can; Canlı, Deniz; Basturk, Fatma Nur; Yesil, Yeter; Kösesakal, Taylan; Ecem Bayram, Nesrin

The aim of this study was to characterize Ranunculus pinardii and Ranunculus constantinopolitanus species anatomically and phytochemically. In order to contribute to the taxonomic characterization of R. pinardii and R. constantinopolitanus, anatomical structures of the stem, leaf, and petiole of species were examined with light microscopy. Total phenolic-flavonoid content, antioxidant activity, total anthocyanin, total proanthocyanidin, and individual phenolic compounds of extracts prepared with different solvents were determined. The results showed that the total phenolic contents of the species had a similar range (0.09-0.45 mg GAE/g for R. pinardii and 0.01-0.44 mg GAE/g for R. constantinopolitanus), but R. constantinopolitanus exhibited higher antioxidant activity (29.28 mg TE/g). Higher individual phenolic compound recovery was achieved from R. pinardii compared to R. constantinopolitanus with all solvents, except methanol. Both species were rich in luteolin, a natural flavonoid (163.98-34417.11 µg/kg for R. pinardii and 44.93-129009.44 µg/kg for R. constantinopolitanus). These results indicate that these plants contain bioactive ingredients that may have applications in different fields, such as medicine, cosmetics, and food. In particular, the detection of luteolin compound at high levels in these species compared to other components indicates that optimization studies aimed at the recovery of luteolin are important for future research.

Exploring the Potential of Isoindole-1,3-Dione Derivatives as Novel Inhibitors of Aldose Reductase: An In Silico and In Vitro Insight into Therapeutic Strategies for Diabetic Complications

Şengül, Bülent; Gündoğdu, Özlem; Kilinç, Namık; Kishali, Nurhan

This study explores the potential of isoindole-1,3-dione derivatives as novel inhibitors of aldose reductase (AR), focusing on their in silico and in vitro effects for therapeutic strategies against diabetic complications. Aldose reductase, a critical enzyme in the polyol pathway, plays a significant role in glucose metabolism and has been linked to diabetic complications. In this comprehensive study, isoindole-1,3-dione derivatives were synthesized and evaluated for their inhibitory effects on the recombinant human AR enzyme. The compounds’ inhibitory activities were measured both in vitro and through in silico techniques, employing molecular docking and free binding energy calculations and ADME studies. The newly synthesized compounds demonstrated varied inhibitory effects, with ethyl and phenyl substituents at specific positions enhancing inhibitory activity. Notably, compounds with carboxylic acid derivatives exhibited potent inhibitory effects, especially compound 6 with an IC50 value of 1.649 μM. In conclusion, this study provides valuable insights into the inhibitory potential of isoindole-1,3-dione derivatives against AR, suggesting their potential therapeutic application in mitigating diabetic complications. The combination of experimental and computational approaches offers a comprehensive understanding of the compounds’ interaction mechanisms and pharmacokinetic profiles, supporting their further exploration as antidiabetic agents.

Synthesis of Arylsulfonyl Hydrazone Derivatives: Antioxidant Activity, Acetylcholinesterase Inhibition Properties, and Molecular Docking Study

Demirci, Yasin; Kalay, Erbay; Kara, Yakup; Güler, Halil İbrahim; Can, Zehra; Şahin, Engin

In this current paper, fifteen novel sulfonyl hydrazone derivatives have been successfully synthesized and evaluated for antioxidant activity as well as their effects on inhibitory activity toward acetylcholinesterase (AChE). By using 1H NMR, 13C NMR, FT-IR, and high-resolution mass spectrometry methods, the full characterization data of the novel compounds were obtained. The synthesized compounds capacity to inhibition glucosidase and exhibit antioxidant activity were tested in vitro. It was determined that compounds (E)-4-((2-((4-chlorophenyl)sulfonyl)hydrazone)methyl)-2-methoxyphenylfuran-2-carboxylate (21), (E)-2-methoxy-4-((2-(phenylsulfonyl)hydrazone)methyl)phenylfuran-2-carboxylate (17) and (E)-4-((2-((4-bromophenyl)sulfonyl)hydrazone)methyl)-2-methoxyphenylfuran-2-carboxylate (25) showed good antioxidant activity. Upon examining the acetylcholinesterase inhibitory activity, it was determined that compounds (E)-2-methoxy-4-((2-((4-methoxyphenyl)sulfonyl) hydrazone)methyl)phenylacetate (27) (10.39 μM), (E)-4-((2-((4-chloro phenyl)sulfonyl) hydrazone)methyl)-2-methoxyphenylfuran-2-carboxylate (21) (10.81 μM), (E)-4-((2-((4-chloro phenyl)sulfonyl)hydrazone)methyl)-2-methoxyphenyl thiophene-2-carboxylate (22) (12.92 μM) and (E)-2-methoxy-4-((2-(phenylsulfonyl)hydrazone)methyl)phenylfuran-2-carboxylate (17) (12.93 μM) showed potent inhibitory effects. Molecular docking simulations were used to investigate the interactions of novel sulfonyl hydrazone derivatives with human acetylcholinesterase protein. The ligands exhibited strong binding to the receptor protein with potent inhibition.
